维A酸【通用名称】维A酸【其他名称】维A酸 维A酸 拼音名:Wei A Suan 英文名:Tretinoin 书页号:2000年版二部-781 C20H28O2 300.44 本品为全反式维A酸。按干燥品计算,含C20H28O2应为97.0%~103.0 %。 【性状】 本品为黄色或淡橙色的结晶性粉末。 本品在乙醇或氯仿中微溶,在水中几乎不溶。 【鉴别】 (1) 取本品,加酸性异丙醇溶液(取0.1mol/L盐酸溶液1ml ,加异丙醇 至1000ml),制成每1ml 中含4μg的溶液,照分光光度法(附录Ⅳ A)测定,在352nm 的波长处有最大吸收。 (2) 本品的红外光吸收图谱应与对照的图谱(光谱集445 图)一致。 【检查】 干燥失重 取本品,在105 ℃干燥3 小时,减失重量不得过0.5 %(附 录Ⅷ L)。 炽灼残渣 取本品1.0g,依法检查(附录Ⅷ N),遗留残渣不得过0.1 %。 重金属 取炽灼残渣项下遗留的残渣,依法检查(附录Ⅷ H第二法),含重金属不 得过百分之二十。 异维A酸 避光操作。取异维A酸对照品适量,精密称定,加二氯甲烷5ml 使溶解, 用异辛烷制成每1ml 中含12.5μg 的溶液,作为对照品溶液。另取本品适量,精密称定, 加二氯甲烷5ml 使溶解,用异辛烷制成每1ml 中含250μg的溶液作为供试品溶液。取上 述溶液,照高效液相色谱法(附录Ⅴ D)测定,用5μm硅胶为填充剂;异辛烷-异丙醇 -冰醋酸(99.65:0.25:0.1)为流动相;检测波长为352mn 。理论板数按异维A酸计算应 不低于1000,异维A酸和维A酸峰的分离度应大于1.0 ,供试品中异维A酸的含量不得 过5.0 %。 【含量测定】 取本品约0.24g ,精密称定,加二甲基甲酰胺50ml溶解后,加麝香 草酚蓝的二甲基甲酰胺溶液(1→100)3 滴,用甲醇钠滴定液(0.1mol/L)滴定至溶液显绿 色,并将滴定的结果用空白试验校正。每1ml甲醇钠滴定液(0.1mol/L)相当于30.04mg的 C20H28O2。 【类别】 抗皮肤角化异常药、细胞诱导分化药。 【贮藏】 遮光,密封保存。 【制剂】 维A酸片
