1.Objective To evaluate the feasibility of two-dimensional gel electrophoresis liver in ratswith idiopathic hyperoxaluria and labeling before DIGE, and to lay experimentalfoundation for DIGE.方法:选择3只24小时尿草酸排泄量较高的特发性高草酸尿症模型大鼠作为实验组,选择3只24小时尿草酸排泄量在正常范围的SD大鼠作为对照组,切取其肝脏组织后分别提取总蛋白,然后利用双向电泳技术分离蛋白,扫描后获得二维电泳的图谱,并对提取的蛋白样品进行荧光标记实验。

高草酸尿高草酸尿hyperoxaluria 正常人每日尿的草酸排泄量为15~60mg,超过此范围为高草酸尿。常见的原因有摄入含草酸丰富的食物,维生素B6缺乏,镁摄入少,回肠疾病致草酸吸收增多和先天性α-酮戊二酸-乙醛酸裂解酶(Ⅰ型)及d-甘油脱氢酶(Ⅱ型)的缺陷造成内源性草酸增多。例如,Ⅰ型缺乏乙醛酸代谢所需要的酶,造成乙醛酸堆积,氧化为草酸。Ⅱ型缺乏d-甘油脱氢酶引起1-甘油酸增加,于是草酸增多。
