羧化酶分子式:CAS号:性质:通常指属于EC6.4、催化CO2(HCO3—)参入有机物分子形成C-C键的一群合成酶,都是以生物素为辅基的生物素蛋白,反应需ATP。HCO3—在ATP、Mg2+在下先形成活性形式,即N-羧化生物素·酶中间复合物,然后转移羧基至底物碳原子上,如丙酮酸羧化酶(EC6.4.1.1.)、乙酰CoA羧化酶(EC6.4.1.2.)、丙酰CoA羧化酶(EC6.4.1.3.)等。此外,尿素羧化酶(EC6.3.4.6.)也是生物素蛋白,利用HCO进行C-N键合成,产生脲基甲酸,继之水解为NH3和CO2属于EC4·1的羧基裂解酶(carboxylyase),催化底物脱羧和羧化,其中的某些酶也称羧化酶,如核酮糖-1,5-二磷酸羧化酶(EC4.1.1.39.),是一种铜酶,在光合作用中起二氧化碳固定作用,是生物圈中最丰富的一类酶。再如磷酸核糖氨基异吡唑羧化酶(EC4.1.1.21.),其参与嘌呤核苷酸合成,在反应中CO2不需先活化。
