氯霉素【通用名称】氯霉素【其他名称】氯霉素 氯霉素 拼音名:Lumeisu 英文名:Chloramphenicol 书页号:2000年版二部-929 C11H12Cl2N2O5 323.13 本品为D-苏式 -(—)-N-〔a-(羟基甲基)- β -羟基- 对硝基苯乙基〕-2,2-二氯 乙酰胺。按干燥品计算,含C11H12Cl2N2O5应为98.0%~102.0%。 【性状】 本品为白色或微带黄绿色的针状、长片状结晶或结晶性粉末;味苦。 本品在甲醇、乙醇、丙酮或丙二醇中易溶,在水中微溶。 熔点 本品的熔点(附录Ⅵ C)为149 ~153 ℃。 比旋度 取本品,精密称定,加无水乙醇溶解并稀释成每1ml中含50mg的溶液,依法 测定(附录Ⅵ E),比旋度为+18.5°至+21.5°。 【鉴别】 (1) 取本品10mg,加稀乙醇1ml 溶解后,加1 %氯化钙溶液3ml 与锌粉 50mg,置水浴上加热10分钟,倾取上清液,加苯甲酰氯约0.1ml,立即强力振摇1 分钟, 加三氯化铁试液0.5ml 与氯仿2ml ,振摇,水层显紫红色。如按同一方法,但不加锌粉 试验,应不显色。 (2) 取本品的无水乙醇溶液,测其旋光度,应为右旋;另取本品的醋酸乙酯溶液, 测其旋光度,应为左旋。 (3) 本品的红外光吸收图谱应与对照的图谱(光谱集507 图)一致。 (4) 取本品50mg,加乙醇制氢氧化钾试液2ml 使溶解,防止乙醇挥散,在水浴中加 热15分钟,溶液显氯化物的鉴别反应(附录Ⅲ)。 【检查】 酸碱度 取本品,加水制成每1ml 中含25mg的混悬液,依法测定(附录 Ⅵ H),pH值应为4.5 ~7.5 。 干燥失重 取本品,在105 ℃干燥至恒重,减失重量不得过0.5 %(附录Ⅷ L)。 炽灼残渣 不得过 0.1%(附录Ⅷ N)。 【含量测定】 取本品适量,精密称定,按氯霉素每10mg加乙醇1ml 使溶解,再用 水稀释成每1ml中含20μg的溶液,照分光光度法(附录Ⅳ A),在278nm的波长处测定吸 收度,按C11H12Cl2N2O5的吸收系数(E1% 1cm)为298计算。 【类别】 抗生素类药。 【贮藏】 密封保存。 【制剂】 (1) 氯霉素片 (2) 氯霉素胶囊 (3) 氯霉素眼膏 (4) 氯霉素滴耳液 (5) 氯霉素滴眼液
