茶氨酸分子式:C7H12N2O4分子量:188.180CAS号:2490-97-3性质:白色结晶。熔点195-199℃。易溶于水,能溶于乙醇和乙酸乙酯。制备方法:L-γ-谷氨酸经酯化、胺化、乙酰化而得。1.酯化 将甲醇加入干燥的反应罐,搅拌下缓缓加入硫酸。冷至20℃ 以下,,加入L-γ-谷氨酸。待溶解后于20℃ 保温4h,得L-γ-谷氨酸甲酯液。重量配料比为L-γ-谷氨酸:甲醇:硫酸=1:10:0.5。2.胺化 将L-γ-谷氨酸甲酯液冷至0℃,加甲基红作指示剂,通氨至中性。加入三硫碳溶解,继续通氨至饱和。于20℃ 放置4d过滤,滤液减压抽去氨气。加入4N乙酸中和。于60℃ 保温0.5h,减压抽去三硫化碳。残液加二倍甲醇析出沉淀。冷却,过滤,干燥,得粗品。加水使成2%水溶液,流经弱碱性苯乙烯系阴离子交换树脂303×2(704),收集流出液,减压浓缩。加入甲醇沉淀,过滤,用甲醇,乙醚反复洗涤,干燥得L-γ-谷氨酸酰胺。3.乙酰化 将氢氧化钠溶液(1N)加入反应罐内,冷至0℃ ,搅拌下加入谷氨酰胺,溶解后搅拌1h。同时滴加乙酐和氢氧化钠溶液(2N),于0-5℃ 放置过液。加6N盐酸至pH1-2,析出结晶。冷却,过滤,用冰冷的无离子不洗到氯根符合要求止,干燥,得乙酰谷氨酰胺。重量配料比为L-γ-谷氨酸酰胺:乙酐(无水)=1:1.1.总收率50%(对L-γ-谷氨酸)。用途:医药。用于脑外伤昏迷,高位性截瘫以及神经系统疾患。也可治疗神经性头痛,腰痛小儿麻痹后遗乙脑后遗症等,与异丙醇铝反应得到乙酰谷酰胺铝,用抗溃疡药。
