超滤 选择性地将分子量小于某一范围的极小颗粒或大分子,通过一定孔径的滤膜,从溶液中分离出来的过程。分离的决定因素主要是分子的大小,也与分子的形状及荷电有关。超滤膜的孔径均匀,按其截留分子量的大小不同,通常可分成分子量自5百至1百万的不同规格。因滤膜孔径极小,滤过率慢,可在滤膜上方通以高压氮气以加压;也可在滤膜下方施以真空负压减压抽滤。影响超滤效率的因素主要是浓度极化,即在膜上积聚一层大分子胶层,阻碍了滤过性。为提高超滤效率,可加悬吊式电磁搅拌,或将滤液加泵以循环之,循环液体通过螺旋式细沟从膜流过,使膜上不出现浓度极化,液面得以不断更新。中空纤维超滤系统可大大增加膜的面积,使滤过速度达到每分钟数百至数千毫升。超滤法可在冷室进行,实验过程温和,使生物大分子在处理过程中不致失活。其用途很广,尤适用于生物大分子的快速浓缩,也是分离和纯化一定分子量物质的有效方法,还可用作透析和脱盐。尤其当样品体积很大时,超滤法常属首选步骤。人工肾透析,就是利用超滤法以排除体内毒性小分子。
