脂多糖  革兰氏阴性细菌的细胞壁外膜的组成成分,结构较复杂,分子量超过10000。脂多糖是热稳定的,易从细胞壁抽提、纯化。它是确定菌体抗原特性的决定因子,因此又称为O-抗原。    脂多糖分子由O-侧链、核心寡糖和脂质 A组成(图1):后两部分合在一起又叫R-核心。O-侧链是由2~4个糖基组成寡糖的重复结构的长链。核心寡糖是9~10个糖基组成的分枝寡糖链,其中包括一种不常见的七碳糖──L-甘油-D-甘露型庚糖(Hep),和一种不常见的八碳糖酸── 3-脱氧-D-甘露型-辛酮糖酸(DO)。Hep和DO的羟基部分地被磷酸、焦磷酸、乙醇胺和磷酸乙醇胺取代。脂质A部分是脂多糖的骨架,基本组成单位是以β-1,6糖苷键相联的D-葡萄糖胺双糖,其中一个糖基的呌位上联着核心寡糖,两个糖基的C1和C4位上的羟基被磷酸取代,其他位置上的羟基和胺基都被脂肪酸取代。脂质A上的脂肪酸常见的有12C,14C,16C的饱和脂肪酸和β-羟十四烷酸。      在革兰氏阴性菌包被物中,脂多糖、磷脂和蛋白质构成肽聚糖层外面的外膜,三者的相对含量因菌种而异。例如,沙门氏菌的外膜中,磷脂:脂多糖:蛋白质=3:3:10,而大肠杆菌则为2∶7∶10。脂多糖的脂质 A部分埋在厚约3NM的磷脂组成的脂双层中,O-侧链伸向细胞外介质中去(图2)。蛋白质则分布在磷脂层中,并和脂多糖的脂质A部分的糖链形成某种程度的网络结构。      脂多糖的O-侧链是抗原决定因子,其糖基组成和结构,不但在不同菌种之间,而且在同一菌种内部不同类群之间,都有所差别,成为细菌免疫学分类的基础。例如,鸭沙门氏菌和鼠伤寒沙门氏菌的O-侧链,其重复寡糖单位虽然都含有相同顺序的半乳糖、甘露糖和鼠李糖,但是因为联结的糖苷键不同,结果成为不同的抗原决定因子。沙门氏菌这一个种,根据O-侧链的抗原性差别,可分成40个以上的类群。不同类群细菌的核心寡糖和脂质A部分,在组成和结构上,也稍有不同。脂质A部分具有内毒素性质,使寄主产生发热等病理症状。    对菌体本身说来,脂多糖有保护作用,它对裂解酶类和寄主细胞的吞噬作用有一定的抗性,并对有害物质的透过有一定程度的阻拦作用。
