替加氟【通用名称】替加氟【其他名称】替加氟 替加氟 拼音名:Tijiafu 英文名:Tegafur 书页号:2000年版二部-805 C8H9FN2O3 200.17 本品为1-( 四氢-2- 呋喃基)-5-氟-2,4(1H,3H)-嘧啶二酮。按干燥品计算,含C8H9 FN2O3应为98.0%~102.0%。 【性状】 本品为白色结晶性粉末;无臭。 本品在甲醇、丙醇或氯仿中溶解,在水或乙醇中略溶,在苯或乙醚中几乎不溶。 熔点 本品的熔点(附录Ⅵ C)为164 ~169 ℃。 【鉴别】 (1) 取本品约0.1g,加碳酸钠和碳酸钾各0.1g,混匀,置坩锅中,缓缓 灼烧至完全炭化,放冷,加水10ml使溶解,加稀醋酸酸化,滤过,取滤液1ml ,加氯化 钡试液,即生成白色沉淀。 (2) 取鉴别(1) 项下的滤液5ml ,加0.1 %茜素磺酸钠溶液与0.1 %硝酸锆溶液的 等容混合液0.2ml ,紫红色即消失。 (3) 取本品约50mg,加水5ml ,加热使溶解,放冷,加溴试液1ml ,振摇,红色即 消失。 (4) 取含量测定项下的溶液,照分光光度法(附录Ⅳ A)测定,在270nm 的波长处 有最大吸收,在235nm 的波长处有最小吸收。 【检查】 溶液的澄清度 取本品0.10g ,加乙醇10ml溶解后,溶液应澄清。 含氯化合物 取本品0.10g ,加水10ml与0.1mol/L氢氧化钠溶液2 滴,置水浴中加 热15分钟,放冷,分取2.0ml ,加水至10ml,加稀硝酸5 滴与硝酸银试液 5滴,依法检 查(附录Ⅷ A),与标准氯化钠溶液4.0ml 制成的对照液比较,不得更浓(0.2%)。 有关物质 取本品,加乙醇制成每1ml 中含10mg的溶液,作为供试品溶液;精密量 取适量,加乙醇稀释成每1ml 中含0.10mg的溶液,作为对照溶液。照薄层色谱法(附录 Ⅴ B)试验,吸取上述供试品溶液20μl 与对照溶液12μl,分别点于同一硅胶HF薄层板(用羧甲基纤维素钠溶液制备)上,以苯-丙酮(5:4) 为展开剂,展开后, 晾干,置紫外光灯(254nm) 下检视,供试品溶液如显杂质斑点,其荧光强度与对照溶液 的主斑点比较,不得更强。 干燥失重 取本品,在105 ℃干燥至恒重,减失重量不得过0.5 %(附录Ⅷ L)。 重金属 取本品0.50g ,依法检查(附录Ⅷ H第三法),含重金属不得过百万分之 二十。 【含量测定】 精密称取本品适量,加无水乙醇制成每1ml 中约含10μg 的溶液, 照分光光度法(附录Ⅳ A),在270nm 的波长处测定吸收度,按C8H9FN2O3 的吸收系数 (E1% 1cm)为439 计算,即得。 【类别】 抗肿瘤药。 【贮藏】 遮光,密封保存。 【制剂】 (1)替加氟片 (2)替加氟注射液
