药物释放用载体材料药物释放用载体材料earrier material fordelivery system  药物释放用载体材料earrier material forddelivery system制作药物释放体系中药物载体合物医学材料。药物载体用于药物贮藏和控制释放,处是由生物医学材料制成的胶囊、管、微球或骨架等,艰生常是体系。不同的药物释放体系,只有选用适宜的材料作载体,才能达到药物控制释放的目的。 种类用于药物释放的载体材料不仅具有良好的生物相容性和可加工性,还必须和药物相容,不损害药物的性能。可分为降解性和非降解性两大类。前者如聚酞胺、多肤类、聚醋类、聚原酸酷类、聚缩醛类、多糖类、聚氰基丙烯酸甲醋类等;后者有无机陶瓷材料和有机聚合物材料,如聚乙烯醇及其共聚物、交联的聚硅氧烷、乙烯一醋酸乙烯共聚物、甲基丙烯酸酷类共聚物、聚氨醋等。常用的载体材料有以下4种。 ①用于扩散控制释放的聚合物材料:眼疗法、生育控制及经皮吸收体系中使用的微胶囊、空心纤维、脂质体和膜材料。常用的为乙基羚丙基纤维素及其衍生物,其次有交联水凝胶。可用于含氟无机盐、前列腺素、麻醉剂、抗菌素等药物载体,如聚乙烯醇与N一乙烯基毗咯烷酮共聚物,以其适宜的亲水性,用于亲水性和疏水性药物释放体系;聚硅氧烷,用于疏水性药物体系;乙烯一醋酸乙烯共聚物(EVA),醋酸乙烯醋(VAC)含量一般为30一50%,用于控制生育药物、蛋白质或经皮吸收药物体系。 ②生物降解性聚合物:分散于聚合物中的药物,因聚合物降解而被释放。如聚原酸醋受环境中pH值变化,可改变释药速率。常用的还有聚乳酸、聚乙醇酸及其共聚物。另一类是将药物分子接枝到聚合物链上,经水解、酶解而使药物得以释放,如将炔诺酮结合到聚(经烷基一左旋)谷氨酞胺上。 ③具有溶剂活性聚合物:由于溶剂对聚合物的渗透和溶胀,溶剂进入药剂核心,并以等体积输出药物溶液。如以纤维素为基础的多层膜可制成微渗透泵,甲基丙烯酸经乙醋与甲基丙烯酸甲醋共聚物、EVA共聚物、交联聚乙烯醇,均可用作载体材料。 ④生物陶瓷:是新发展的一类药物载体材料。可以通过加入短射程的放射性同位素制成生物陶瓷微球,利用血液传递并在肿瘤部位浓集达到治疗效果;也可以在生物活性玻璃陶瓷中加入磁性物质,如Fe,03,将其植入骨癌所在部位的骨中,利用微波加热杀死癌细胞;也可利用多孔A12O3陶瓷,通过孔隙大小和孔隙率的控制,使药物通过孔隙缓慢释放。药物控制释放体系的重要组成部分。 药物控制释放体系是新一代给药制剂。其施药方式不同于传统的按剂量配制的药剂,需要通过按时口服或注射等来维持药物在血浆中的浓度和疗效,而是根据治疗的需要,按设定的施药速率或施药程序持续施药,或集中于患病的器官或靶位按确定的时间和程序定点施药。
