反向打点杂交技术分子式:CAS号:性质:系指分子遗传学中一项具体操作技术。它由Conner等人于1983年首创的等位基因特异性寡核苷酸(ASO)探针杂交技术上发展起来,由聚合酶链式反应(PCR)技术创立者之一Saiki等人改进并建立了这一反向打点杂交技术。传统的ASO探针杂交技术是将靶DNA(基因组DNA或PCR扩增片段)点在膜上,使其与液相中的探针退火,经洗膜后检出信号。而改进后的技术是将ASO探针固定在尼龙膜上,用扩增的DNA样品与膜上探针杂交。由于ASO探针长度有限(一般为15~20个碱基),为使探针序列不受固定过程的影响,当ASO合成后,用末端转移酶在其3′一端加一上段多聚核苷酸(dT)尾(一般为400个左右),这种带尾的ASO点在膜上后,经紫外光照射,其多聚dT即可与尼龙膜表面的氨基发生作用而结合在膜条上。这一个“反向”体系的突出特点之一是能将多种不同序列的ASO均固定在同一条膜上。关键是ASO的设计需根据其碱基组成选择探针长度,以使各种固定的ASO在杂交时具有尽可能接近的解链温度(Tm)。这样一张膜经一次杂交即可鉴定出样品中多种不同的突变。其缺点是对于未知DNA序列及其突变类型的检测对象无能为力。本方法主要是作为基因诊断和遗传病、DNA多态性及癌基因分析等领域中。
