枸橼酸他莫昔芬【通用名称】枸橼酸他莫昔芬【其他名称】枸橼酸他莫昔芬 枸橼酸他莫昔芬 拼音名:Juyuansuan Tamoxifen 英文名:Tamoxifen Citrate 书页号:2000年版二部-452 C26H29NO.C6H8O7 563.65 本品为(Z)-N, N-二甲基-2-〔4-(1,2-二苯基-1-丁烯基)苯氧基]—乙胺枸橼酸盐。按干燥品计 算,含 C26H29NO.C6H8O7不得少于99.0%。 【性状】本品为白色或类白色结晶性粉末;无臭。 本品在冰醋酸中易溶,在甲醇中溶解,在乙醇或丙酮中微溶,在氯仿中极微溶解,在水中几乎不 溶。 熔点 本品的熔点(附录Ⅵ C)为142~148℃,熔融时同时分解。 【鉴别】(1)取本品适量,加醋酐-吡啶(1:5)5ml,摇匀,置水浴上加热,溶液颜色由黄色变为红 色。 (2)取本品,加无水乙醇制成每1ml中含10μg的溶液,照分光光度法(附录Ⅳ A)测定,在238nm与 278nm的波长处有最大吸收。 (3)本品的红外光吸收图谱应与对照的图谱(光谱集265图)一致。 【检查】干燥失重 取本品,置105℃干燥4小时,减失重量不得过0.5%(附录Ⅷ L)。 E-异构体 照高效液相色谱法(附录Ⅴ D)测定。 色谱条件与系统适用性试验 用十八烷基硅烷键合硅胶为填充剂;乙腈-磷酸盐缓冲液(取磷酸二 氢钠 0.9g、N,N-二甲基辛胺4.8g,加水至1000ml,用2mol/L磷酸溶液调pH值至3.0)(40:60)为流动相; 检测 波长为240nm。理论板数按枸橼酸他莫昔芬峰计算应不低于2000,E-和Z-异构体峰的分离度应符合规 定。 测定法 取本品适量,加流动相制成每1ml中含0.2mg的溶液,作为供试品溶液;另取E-异构体对 照 品适量,加流动相制成每1ml含1.0μg的溶液,作为对照品溶液。取上述两种溶液各20μl,分别注入液 相色 谱仪,记录色谱图,供试品溶液的色谱图中如出现与对照品溶液相应的色谱峰,其峰面积不得大于对 照品溶 液主成分的峰面积(0.5%)。 【含量测定】取本品约0.35g,精密称定,加冰醋酸50ml,微温使溶解后,加结晶紫指示液1滴, 用高氯 酸滴定液(0.1mol/L)滴定,至溶液显蓝绿色,并将滴定的结果用空白试验校正。每1ml高氯酸滴定液 (0.1mol/L)相当于56.36mg的C26H29NO.C6H8O7。 【类别】抗肿瘤药。 【贮藏】遮光,密封,在干燥处保存。 【制剂】枸橼酸他莫昔芬片
