磷酸单酯酶分子式:CAS号:性质:多种酶蛋白的总称,属于水解酶类中的磷酸酶(phosphatase)类。它们仅能催化酯化一次的磷酸基,即把磷酸单酯化合物中磷酸单酯键切断而使磷酸基游离。根据它们对所作用的底物的专一程度,可把它们分为特异性和非特异性两种。前者常冠以底物名以便与后者区别,如己糖-6-磷酸酯酶等。后者又根据其催化反应时最适pH值条件将其分为四个类型,即Ⅰ至Ⅳ型。Ⅰ型,又称碱哇磷酸酯酶(alkaline phosphatase;EC 3.1.3.1。;分子量8.0×104~1.9×105),最适pH值8.6~9.4,pH值7.5~8.5稳定,可被Mg2+或其他二价阳离子活化。对水解磷酸甘油(为该类酶的标准底物)中的α-异构体比β-异构体快,在机体中主要分布在骨、肠黏膜、肾、乳腺等。在细胞中它属于膜结合蛋白之一。Ⅱ型,又称酸性磷酸酯酶(acid phosphatase;EC3.1.3.2.;分子量2.3×104~9.6×104),最适pH值5.0~5.5,pH值5.0~6.0时最稳定,不能被Mg2+所活化,氟化物对它们有明显抑制作用,对磷酸甘油作用同Ⅰ型。Ⅲ型,最适pH值3.0~4.2,pH值4.5~5.5最稳定,pH值7.0时很不稳定,Mg2+对它们有抑制作用,对标准底物作用与Ⅰ型同。Ⅳ型,最适pH值5.0~6.0,pH值6.5~7.5下最稳定,Mg2+、Mn2+对它们有活化作用,对标准底物作用同I型,在红细胞、微生物中均含有之。
