蓖麻毒蛋白分子式:CAS号:性质:又称蓖麻毒素。自大戟科(Eu-phorbiaceca)植物蓖麻(Ricinus communis Linne)成熟种子分离而得的天然糖蛋白,为毒性硬蛋白。分子量65 000,略溶于水、乙醇、氯仿及乙醚,遇酸不生成盐。粗制剂溶于水和稀盐溶液。干燥的产品在室温下稳定,甚至可干热到100℃;潮湿产品稳定性下降,煮沸即完全丧失活性,即100℃ 20min即失效,等电点pI7.1。一种毒性很高的细胞毒类植物蛋白。半致死剂量为7~10μg/kg。其分子由A链(分子量32 000)和B链(分子量34 000)组成;二者之间有二硫键(-S-S-)连接。A链能催化灭活核糖体60S亚基,从而抑制蛋白质生物合成;B链起载体作用,能使毒素与细胞表面结合。它对RNA合成有中度抑制作用,对DNA合成无作用。对移植性动物肿瘤具有明显抑制作用,抗瘤谱较广,对鳞癌和腺癌均有效。将毒素或其A链与抗体分子连接制成的免疫毒素,可用于特异性杀伤各种癌瘤细胞。蓖麻毒的有效成分是蓖麻碱(ricinine;ricidine)1,2-二氢-4-甲氧基-1-甲基-2-氧代-3-吡啶腈。纯蓖麻碱比整个糖蛋白毒性更大,为VX的1000倍。蓖麻毒素是19世纪以来在生产蓖麻油和蓖麻子制品时发现的。第二次世界大战期间,英、美共试生产了蓖麻毒素1700kg,代号WA。制成武器,最小致死剂量为0.02mg/kg。
