磷酸化酶b激酶分子式:CAS号:性质:又称磷酸化酶b激酶。系糖原的合成或分解过程中,又起催化又起调控作用的一个重要而复杂酶蛋白,EC.2.7.1.38,分子量1.2×106。其分子中有α,β,γ,δ四种亚基、十六个多肽组成的大分子。这个酶有双重控制,酶蛋白被蛋白质激酶磷酸化后,可由无活性型转化成活性型,催化磷酸化反应;另外,酶分子中的δ亚基是钙调蛋白,可以与Ca2+结合改变构象使酶部分活化,并发生自身磷酸化。在生物体中,它往往在激素(如胰岛素、肾上腺素)或cAMP作用下与蛋白激酶,磷酸化酶等构成级联放大反应系统(Cascade system)起催化和调控作用。在肌肉组织中,三者分子比为:蛋白激酶:磷酸化酶激酶:磷酸化酶=1:20:120。磷酸化酶激酶在体内是催化来自腺苷三磷酸(ATP)的磷酸基转移,使糖原磷酸酶b磷酸化成为糖原磷酸酶a活化型,也就是说,它把ATP上一个磷酸基转移至糖原磷酸化酶b上14位上丝氨酸而变成有活性的a型。这一共价修饰调控是可逆的,丝氨酸上这一磷酸基仍可被磷酸化酶磷酸酶(phosphorylase phosphatase,EC 3.1.3.17.)水解下来而成为无活性的磷酸化酶b。
