细胞内钙稳态失调在细胞功能的调节中,ca2+可作为第二信使起着信号传导的关键作用,同时ca2+ 也是多种参与蛋白质、磷脂和核酸分解的酶的激活分子之一。正常情况下,细胞内钙稳态是由质膜ca2+转位酶和细胞内钙池系统共同操纵控制的。细胞损害时,这一操纵过程紊乱可导致ca2+内流增加,ca2+从细胞内储存部位释放与/或通过质膜逐出抑制,从而导致细胞内ca2+浓度不可控制的持续增加,细胞内ca2+ 浓度持续高于生理水平以上必然导致维持细胞结构和功能的重要大分子难以控制的破坏。而且这种持续增加将会完全破坏正常生命活动所必需的由激素和生长因子刺激而产生的短暂的ca2+浓度瞬变,危及线粒体功能和细胞骨架结构,最终激活不可逆的细胞内成分的分解代谢过程。毒物可在不同水平上干扰细胞信号的传递,导致细胞内ca2+ 对激素及生长因子的正常反应的丧失。另外,钙信号系统的异常活化也是毒物引起细胞死亡的一个重要机制。当前,细胞内钙稳态失调是细胞损害与机制研究方面最为热门的话题,大量证据表明,细胞钙的持续增高可能活化各种不同组织和细胞的毒性机理,因而曾被称为“细胞死亡的最终共同途径”。
