二氢叶酸还原酶分子式:CAS号:性质:是催化叶酸还原成四氢叶酸(H4FA)的酶。H4FA形成过程分两步进行,由同一个酶催化,H4FA作为一碳单位载体,为嘌呤核苷酸从头合成和胸嘧啶核苷酸合成等提供一碳单位。氨基蝶呤和氨甲蝶呤的结构类似叶酸,与二氢叶酸还原酶结合的平衡常数为10-10mol/L,比叶酸与该酶的结合能力强1000倍,故是一种极强的竞争性抑制剂。肿瘤细胞经用氨甲蝶呤处理,增殖停止,最终细胞死亡,正是因为核苷酸合成受阻,进而阻断核酸合成的结果。
