肝微粒体药物代谢酶分子式:CAS号:性质:肝脏微粒体中含有多种酶系统,是许多药物、杀虫剂、除草剂、污染物及食物添加剂等外源性高脂溶性物质在体内的主要生物转化场所,也称为肝微粒体药物代谢酶,简称为肝药酶。该酶系统底物面广,活性个体差异大,影响活性因素多。其中最重要的是混合功能氧化酶系统,可催化许多不同型的氧化反应,如羟基化,烷基化,脱氨基化,脱卤素,硫原子氧化等。该酶系统中一个主要成分是细胞色素P450,是由与一氧化碳结合后,在光谱450nm处有吸收峰而得名。细胞色素P450含量高低反映了混合功能氧化酶系统活性的高低,这个系统将分子氧中一个氧原子氧化药物,另一个氧原子生成水,没有产生相应的还原物,故也称为细胞色素单加氧酶。肝药酶主要生物学意义是使大多数药物转化后降低或消失活性,极性增加,易于排出体外。少数药物经生物转化后可产生药理活性,如环磷酰胺,在细胞色素P450催化下氧化成4-羟基环磷酰胺,开环后变成磷酰胺氮芥才有烷化作用。个别药物如非那西丁,经转化生成对氨基苯乙醚,毒性加大。也有少数化合物如多环芳香烃经转化可成为有致癌及致突变的中间体。
