亚甲蓝【通用名称】亚甲蓝【其他名称】亚甲蓝 亚甲蓝 拼音名:Yajialan 英文名:Methylthioninium Chloride 书页号:2000年版二部-225 C16H18ClN3S.3H2O 373.90 本品为氯化3,7-双(二甲氨基)吩噻嗪-5-{翁}三水合物。按干燥品计算,含C16H18 ClN3S不得少于98.5%。 【性状】 本品为深绿色、有铜光的柱状结晶或结晶性粉末;无臭。 本品在水或乙醇中易溶,在氯仿中溶解。 【鉴别】 (1) 取本品约10mg,加水50ml溶解后,显深蓝色;分取溶液10ml,加稀 硫酸1ml 与锌粉0.1g,蓝色即消失,滤过,滤液置空气中或加过氧化氢试液1 滴,复显 蓝色;另取溶液10ml,加碘化钾试液数滴,即生成深蓝色的绒毛状沉淀,沉淀后,上层 溶液显淡蓝色;再取溶液10ml,加0.1mol/L碘溶液数滴,即显深棕色;加0.1mol/L硫代 硫酸钠溶液复显蓝色。 (2) 本品的红外光吸收图谱应与对照的图谱(光谱集143 图)一致。 【检查】 干燥失重 取本品,在105℃干燥至恒重,减失重量不得过18.0%(附 录Ⅷ L)。 炽灼残渣 不得过1.2%(附录Ⅷ N)。 锌盐 取本品0.10g ,加硫酸数滴湿润后,炽灼,残渣中加稀盐酸5ml 与水5ml , 煮沸,加氨试液5ml ,滤过,滤液中加硫化铵试液2 滴,不得发生沉淀或浑浊。 砷盐 取本品 0.20g,加氢氧化钙0.5g,混合,加水少量,搅拌均匀,干燥后,先 用小火烧灼使炭化,再在600 ~700 ℃炽灼使完全灰化,放冷,加盐酸5ml 与水23ml使 溶解,依法检查(附录Ⅷ J第一法),应符合规定(0.001%)。 【含量测定】 取本品约0.2g,精密称定,置烧杯中,加水40ml溶解后,置水浴上 加热至75℃,精密加入重铬酸钾滴定液(0.01667mol/L)25ml,摇匀,在75℃保温20分钟, 放冷,用垂熔玻璃漏斗滤过,烧杯与漏斗用水洗涤4 次,每次2.5ml ,滤过,合并滤液 与洗液,移置具塞锥形瓶中,加水250ml 、硫酸溶液(1→5)25ml与碘化钾试液10ml,摇 匀,用硫代硫酸钠滴定液(0.1mol/L)滴定,至近终点时,加淀粉指示液2ml,继续滴定至 蓝色消失,并将滴定的结果用空白试验校正。每1ml重铬酸钾滴定液(0.01667mol/L) 相当于10.66mg 的C16H18ClN3S 。 【类别】 解毒药。 【贮藏】 遮光,密封保存。 【制剂】 亚甲蓝注射液
