放射免疫分析放射免疫分析radioimmunoassay,RIA 放射免疫分析的基础是放射性标记抗原和非标记抗原,同时与限量的特异性抗体进行竞争性免疫结合反应,这种竞争关系可以用以下反应式来表示:〓〓式中*Ag代表标记性抗原,Ag代表非标记抗原,Ab代表特异抗体,*Ag-Ab代表标记抗原抗体复合物,Ag-Ab代表非标记抗原抗体复合物。由于*Ag与Ag免疫活性相同,对Ab有同样的亲和力,当*Ag与Ab为恒量,Ag和*Ag的总量大于Ab上有效结合点时,三者混合后*Ag-Ab的形成量与Ag量成反比。剩下的未结合或游离的*Ag则与Ag量成正比。因此测定*Ag-Ab或*Ag即可推算出被测的Ag量。
