皮肤睾丸(卵睾)组织和中期羊水细胞培养皮肤睾丸(卵睾)组织和中期羊水细胞培养 诊法。染色体检测的细胞培养技术之一。取小块皮肤或性腺,用无菌之含双抗的Ha~nks液清洗数遍后,在无菌条件下将标本剪成05~1mm3小块种入25ml小培养瓶内,倒置,加15~2ml培养液(RPMI1640或TC199等配制的培基,37℃,CO2孵箱内静置培养,24小时后翻瓶,使培养物全部浸泡于培养液内,一周不必翻动。此后每3~5天或视培养基pH值下降即液体转黄时换培养液一次,使培养液保持一定pH值,清澈透亮。一般7~10天培养物即开始生长。待细胞长满后可传代,部分细胞可液氮内保存备用;部分细胞以胰酶消化后加秋水仙素,制作染色体标本及显带分析。羊水细胞培养后染色体标本制备与上类同。
