茶树育种抗性鉴定茶树育种抗性鉴定appraisal of resistance in tea breeding 茶树育种抗性鉴定(appraisal of resi-stance in tea breeding)茶树育种过程中对抵抗不良环境因子所表现的能力进行鉴定的方法。扰性鉴定主要包括对抗寒性、抗早性、抗病性与抗虫性的鉴定。 扰寒性鉴定茶树不同品种抗寒性差别很大,南方类型的大叶种只能忍受一3一一5’C的低温。北方类型的中、小叶种能忍受一15一一18’C的低温。低温冻容对茶叶产t、品质形响很大,因此茶树育种必须进行抗寒性鉴定。主要方法有: 5级评分法在自然或人工低温条件下,根据受冻害程度用5级评分标准鉴定抗寒性强弱。 1级茶芽和叶片在5%以内受害; 2级茶芽和叶片有5一25%受害,1年生枝条在 5%以内受冻. 3级茶芽和叶片有25一50%受害,1年生枝条有 5一25%受害,骨干枝受害在5%以内; 4级茶芽和叶片有50一75%受害,l年生枝条有 25一50%受害,骨干枝有5一25%受害,地 上部受害在5%以内; 5级茶芽和叶片受害达75%以上,1年生枝条有 50%以上受伤,骨干枝有25%以上受害,地 上部受容在5%以上。 早期导电度鉴定法它的主要原理是:当茶树受低沮影响时破坏了细胞腆,伤害原生质结构,引起透性增加,结果细胞内含物有不同程度外渗,使外液导电度增大,用电导仪可以侧定出来,导电度愈大,表示受伤愈重,抗寒性愈弱,反之,则抗寒性愈强。为编短育种年限,可将新品系成熟枝条!于0℃下30分钟(双冷),取出放在需要测定的低温下(如一10℃)处理2小时,然后移至室沮下水培2一3天,检查受冻害程度。 抗寒性等级可根据在不同低温下(人工调整低沮冰箱沮度)受冻容程度(导电度)决定。 叶片组织结构鉴定茶树叶片上表皮厚度、栅状组织厚度,姗状组织厚度与叶片总厚度的比值、栅状组织和海绵组织厚度比值等与抗寒性有密切关系.它们的相关系数依次为0.78、0.81、0.84、0.85,均呈较强的正相关。根据它们的厚度和比值可以鉴定不同品种的抗寒性。叶片表皮角质膜厚度也是茶树抗寒性标诸,用角质膜与叶全厚比值、角质膜与海绵组织比值大小,可以鉴定抗寒性强弱。此外,生化成分含量多少也可以鉴定不同品种抗寒性强弱:氮基酸含量高,尤其是其中茶氮酸含量高,叶绿素总量含量高,茶多酚含量较低,酚氮比较少,儿茶素组成中的L一EGC含量较高,抗寒性强,反之,抗寒性弱。
