癌基因癌基因oncogene人类或其他动物细胞(以及致癌病毒)固有的一类基因。又称转化基因,它们一旦活化便能促使人或动物的正常细胞发生癌变。对于癌基因的研究目前尚存在争议,抱乐观态度的人认为这是肿瘤研究中的重要突破。癌基因的分离成功不仅有助于阐明癌变的机理,而且有重要的实用价值。在理论上,它说明化学致癌物和致癌病毒引起癌的根本原因可能在于激活了细胞中内在的原癌基因。在实用意义上,由于癌基因的激活使细胞合成相应的、特异的转化蛋白,后者有可能被用于诊断。而且如果能抑制癌基因的激活或使转化蛋白失活,那么将有可能提供癌治疗的新途径。持保留态度的人则认为癌变是一个多阶段的过程,把它看作只需要一个癌基因的激活的结果似乎是过于简单化了。尤其是在正常细胞中癌基因也并不是完全不表达的。这一现象的发现使问题更为复杂化。迄今为止,已分离的癌基因多与致癌的RNA病毒有关,而且都是依据它们对小鼠成纤维细胞转化受体系统的致癌活性来判定的。因此还可以怀疑这种系统的局限性,可能某些癌基因用现有的手段尚无法检出。
