甲醇酵母表达系统甲醇酵母基因表达系统是一种最近发展迅速的外源蛋白质生产系统。甲醇酵母是可利用甲醇作为唯一碳源的酵母，主要有h.polymorpha、candida bodinii、pichia pastoris三种，其中pichia pastoris作为基因表达系统使用得最多、最广泛［1］。与以往的基因表达系统相比，它具有无可匹敌的高表达特性，已被认为是最具有发展前景的生产蛋白质的工具之一。1. 外源蛋白质的基因在甲醇酵母中的高效表达目前已有多种蛋白质的基因在该表达系统中克隆成功，包括蛋白酶、酶抑制剂、受体、单链抗体等(表1)。尽管各种外源蛋白质产生的水平不一，但各种蛋白质在甲醇酵母中的产生水平均为在细菌、昆虫或哺乳动物等表达系统中产量的10～100倍［2］。如表皮生长因子(egf)在酿酒酵母中的产量为7.4mg/l，而在甲醇酵母中为450mg/l，提高了60倍［3］。椐报道，外源蛋白质在甲醇酵母中的产量最高可达12g/l［9］。2. 实现外源蛋白质高效产生的关键因素2.1 表达载体 首先，甲醇酵母中没有稳定的质粒，所以其表达载体采用整合型质粒。利用醇氧化酶(甲醇代谢的关键酶)基因-1(aox1)的启动子(paox1)和转录终止子(3′aox1)构建成整合型表达载体。paox1是一个极强的启动子，醇氧化酶的产量最高可占甲醇酵母中可溶性蛋白质的30%［2］，所以能使外源蛋白质在它的控制下高效产生。其次，在载体中加入酿酒酵母的分泌信号和前导肽序列(α因子)构建分泌型载体，一方面可以减轻宿主细胞的代谢负荷，另一方面可以减少宿主细胞蛋白水解酶对外源蛋白质的降解，ppic9k、pmetαa、b、c均为此类载体。此外，使多拷贝目的基因整合入甲醇酵母染色体，形成多个表达单元，产生高产的菌株，此类载体有pao815、ppic3.5、ppic9等。2.2 受体菌 在以葡萄糖或甘油为碳源的培养基上生长时，甲醇酵母中aox1基因的表达受到抑制，而在以甲醇为唯一碳源时，诱导基因表达，使外源蛋白质的产量更高。甲醇酵母适合高密度连续培养的条件，细胞干重达100g/l以上，蛋白质产量极高［14］。受体菌采用蛋白水解酶缺陷型，从而大大降低产物的降解。常用的甲醇酵母受体菌有gs115、km71、pmad11、pmad16等。3. 应用前景经过近几年的发展完善，甲醇酵母表达系统已日趋成熟，应用也日趋广泛。国内已有多篇在甲醇酵母中生产外源蛋白质成功的报道(表2)。美国invitrogen公司也已开发出多种新型的甲醇酵母表达系统试剂盒，如multi-copy pichia expression kit、easyselect pichia expression kit、pichia methanolia expression system等，利用甲醇酵母表达系统克隆一个外源基因已十分方便。相信随着对甲醇酵母研究的进一步深入，它在生产外源蛋白质方面将日益展示出诱人的前景。
