抗原抗原antigen具有免疫原性和反应原性的物质。又称免疫原。免疫原性是指刺激机体的免疫活性细胞产生免疫应答的能力。反应原性是指抗原与抗体或致敏淋巴细胞发生特异性结合而出现可见反应的能力。抗原在化学结构上与机体自身不同，具有异物性：①异种物质。从生物进化过程来看，异种动物间的血缘关系越远，则免疫原性越强。如马的血清和各种微生物与人的血缘关系远，所以免疫原性强。而马的血清与驴、骡的血缘关系近，所以免疫原性相对就弱。②同种异体物质。如人的红细胞抗原物质和人的白细胞抗原等。③自身物质。自身物质一般不具免疫原性。有些物质如隐蔽的自身成分（眼晶体蛋白、精子等），在正常情况下与免疫系统是隔绝的。但是一旦屏障遭到破坏，这些物质进入血流，即可与免疫活性细胞接触而成为自身抗原异物。另外，自身物质在外伤、感染、药物和射线的影响下，其理化性质发生质的改变时，也可成为具有免疫原性的抗原物质。抗原具有特异性，反映在两个方面：①能刺激机体产生具有高度针对性的免疫应答（免疫原性的特异性）。②只能与其相对应的抗体或致敏淋巴细胞结合而不能与无关的抗体或致敏淋巴细胞结合（反应原性的特异性）。与人类有关的抗原有：①病原微生物。在医疗中将病原微生物制成疫苗进行预防接种，可以提高人的免疫力。也可以根据微生物抗原的特异性进行各种免疫学试验，帮助诊断疾病。②同种异体抗原。有两大类，一类是红细胞血型抗原，包括A、B、O血型抗原，Rh血型抗原等。不同血型间相互输血，可引起严重的输血反应；另一类是存在于人类白细胞细胞膜上的人类白细胞抗原（HLA），又称主要组织相容性抗原。它们与血型抗原一样，也是由遗传决定的，受染色体上的基因控制。不同的个体（同卵双生者除外）其组织细胞的组织相容性抗原绝大多数不完全相同，因此，在同种异体进行皮肤或脏器移植时，常因供者移植物中存在受者所没有的抗原成分，刺激受者产生对移植物的免疫反应，导致移植物受到排斥而坏死脱落。③动物免疫血清。临床上常用的各种抗毒素血清，一般都是用免疫马来制备的。一方面，抗毒素能中和与其相应的外毒素，起到防治疾病的作用；另一方面，它能刺激人体产生抗马血清蛋白的抗体，当再次接受马的免疫血清时，有可能发生超敏反应。④嗜异性抗原。一类与种属特异性无关的、存在于人以及某些动物、植物、微生物的性质相同的抗原。⑤肿瘤抗原。由物理的、化学的因素或某些病毒诱发的实验动物肿瘤，其细胞中或细胞表面均出现特异性抗原，称为肿瘤特异性抗原。已证实在某些人类肿瘤中心存在着与病毒密切相关的抗原。
