丙酸倍氯米松【通用名称】丙酸倍氯米松【其他名称】丙酸倍氯米松 丙酸倍氯米松 拼音名:Bingsuan Beilumisong 英文名:Beclometasone Dipropionate 书页号:2000年版二部-101 C28H37ClO7 521.05 本品为16β-甲基-11β,17α,21-三羟基-9α- 氯孕甾-1,4-二烯-3,20-二酮 -17,21-二丙酸酯。按干燥品计算,含C28H37ClO7应为97.0%~103.0%。 【性状】 本品为白色或类白色粉末;无臭。 本品在丙酮或氯仿中易溶,在甲醇中溶解,在乙醇中略溶,在水中几乎不溶。 比旋度 取本品,精密称定,加二氧六环溶解并定量稀释制成每1ml 中含10mg的溶 液,依法测定(附录Ⅵ E),比旋度为+88°至+94°。 【鉴别】 (1) 取本品约25mg,照氧瓶燃烧法(附录Ⅶ C)进行有机破坏,以水20 ml与0.4%氢氧化钠溶液1ml为吸收液,燃烧完全后,溶液显氯化物的鉴别反应(附录Ⅲ)。 (2) 取本品约20mg,置试管中,加磷酸3~4滴,混匀,试管上端覆以5%硝酸镧 溶液润湿的滤纸,小火缓缓加热至沸,取下滤纸条加0.02mol/L碘溶液1滴后,再加氨 试液1 滴,滤纸渐变为蓝紫色。 (3) 取本品,精密称定,加乙醇溶解并定量稀释制成每1ml中含20μg 的溶液,照 分光光度法(附录Ⅳ A)测定,在239nm的波长处有最大吸收,吸收度为0.57~0.60; 在239nm与263nm波长处的吸收度比值应为2.25~2.45。 (4) 本品的红外光吸收图谱应与对照的图谱(光谱集71图)一致。 【检查】 其他甾体 取本品,加氯仿-甲醇(9:1) 制成每1ml 中含3.0mg 的溶液 ,作为供试品溶液;精密量取适量,加氯仿-甲醇(9:1) 稀释成每1ml 中含60μg 的溶 液,作为对照溶液。照薄层色谱法(附录Ⅴ B)试验,吸取上述两种溶液各5μl,分别 点于同一硅胶G薄层板上,以二氯乙烷-甲醇-水(95:5:0.2)为展开剂,展开后,晾干 ,在105 ℃干燥10分钟,放冷,喷以碱性四氮唑蓝试液,立即检视。供试品溶液如显杂 质斑点,不得多于2 个,其颜色与对照溶液的主斑点比较,不得更深。 干燥失重 取本品,在105℃干燥至恒重,减失重量不得过0.5%(附录Ⅷ L)。 炽灼残渣 不得过0.1%(附录Ⅷ N)。 【含量测定】 照高效液相色谱法(附录Ⅴ D)测定。 色谱条件与系统适用性试验 用十八烷基硅烷键合硅胶为填充剂;甲醇-水(74:26 ) 为流动相;检测波长为240nm 。理论板数按丙酸倍氯米松峰计算应不低于2500,丙酸 倍氯米松峰和内标物质峰的分离度应大于4.0 。 内标溶液的制备 取甲睾酮,加流动相制成每1ml中含0.12mg的溶液,即得。 测定法 取丙酸倍氯米松对照品约12.5mg,精密称定,置100ml量瓶中,加甲醇74 ml使溶解,用水稀释至刻度,摇匀;精密量取该溶液10ml与内标溶液5ml ,置50ml量瓶 中,加流动相稀释至刻度,摇匀,取20μl 注入液相色谱仪,记录色谱图;另取本品适 量,同法测定,按内标法以峰面积计算,即得。 【类别】 肾上腺皮质激素类药。 【贮藏】 密封,避光保存。 【制剂】 (1) 丙酸倍氯米松气雾剂 (2) 丙酸倍氯米松软膏
