放射性示踪剂量估算放射性示踪剂量估算treatment of date from radi-active counting  放射性示踪剂t估算(estima‘ion of radio-tracer dose)示踪试验前估算所需的示踪剂(比)活度和引入的总量。估算的目的是为使试验样品有足够的计数率,保证试验的准确结果,又不致由于引入剂量过大而带来对试验生物体的辐射效应一般悄况下,要求最终样品的计数率不低于本底的一倍,但又不要求超过本底很高的活度。虽然样品的放射性活度越高.灵敏度越高,测量结果越准确,测量时间也可缩短,然而若引入剂量过大,对试验生物产生辐射效应,影响生物体正常的生理活动,同样影响试验结果的准确性.同时高活度样品,增加辐射损伤,并造成示踪剂的浪费. 估算引入剂量,要考虑以下因素:①示踪剂在试验体系内的稀释程度.示踪剂进入生物体后经运转、分配和随着植株生长,遭到物理稀释,使样品比活度变小,最后所需总活度应由要求样品达到的比活度和试验体的总量计算。另一方面,试验生物体内本来存在的或其他来源的同一种非放射性物质或元素使示踪剂遭到物理化学稀释,使测定成分或标记代谢产物的比活度降低,这要估计最后样品中待测成分的总量和示踪养分的吸收率及结合到待测组分的程度来估算引入示踪剂的稀释倍数以确定引入示踪剂的比活度和引入t。②示踪剂在体内分布的不均匀性。由于植株各部位对示踪剂的选择吸收或植株的生理特性,造成示踪剂在各器官、组织中分布不均匀。试验时,要使分配最少部位的样品有足够的计数率.不均匀性还指示踪核家结合到各组成物中量的差异,在代谢或物质转化研究中,要使各待测组分有足够的(比)活度。③时间因素。一般试验从开始到结束,要经历一段时间,短半衰期示踪剂因衰变而减少活度。因此,试验时必须使衰变后的活度仍符合测量要求.将最后要求达到的总计数率除以衰变常数K,即为所需引入剂量.④测量效率.所有的放射性探测仪不可能将样品中的放射性衰变数全部检侧出来,因此还需将要求达到的总计数率(C尸M,每分钟脉冲数)除以仪器的计数效率,求出需要引入的实际活度(D尸M,每分钟衰变数)。此外必须考虑制备放射性测量祥品时,从样品中放射性的回收率,将要求的总活度除以回收率。经上述各项估算样本要求的总活度除以示踪剂的比活度,即为引入示踪剂的t。
