二酰基甘油分子式:CAS号:性质:激素-信息传递的磷酸肌醇系统中具有第二信使作用的化学信息分子。是一个甘油分子的三个羟基中有两个羟基和两个脂肪酸缩合失去两分子水形成的酯。CDP-二脂酰同与肌醇可形成肌醇磷脂(磷脂酰肌醇)。肌醇磷脂经激酶作用生成磷脂酰肌醇-4,5-二磷酸(三磷酸肌醇,PIP2)。当激素、神经递质与膜受体结合后,激活G蛋白介导的磷酯酶C(磷酸肌醇酯酶,PIC),催化PIP2水解产生肌醇三磷酸(IP3)和DG的反应。IP3能够促使内质网Ca2+库释放Ca2+,引起胞内游离Ca2+浓度瞬间增加,启动胞内Ca2+信号系统,继而激发一系统生理反应。DG则通过Ca2+增加激活蛋白激酶C(PKC),以磷酸化形式对许多蛋白质和酶进行修饰,进而调节和控制另外一系列的生理过程。同此可见PIP2水解可以分别产生两个胞内信使,分别激动IP3-Ca2+ tDG-PKC两个信号传递途径,这两个信使途径相辅相成,又互相制约,从而建立了肌醇磷酯第二信使系统。
