复方甘草片【通用名称】复方甘草片【其他名称】复方甘草片 复方甘草片 拼音名:Fufang Gancao Pian 英文名:Compound Liquorice Tablets 书页号:2000年版二部-503 本品每片中含无水吗啡(C17H19NO3) 应为0.36~0.44mg。 【处方】 甘草浸膏粉(中粉) 112.5g 阿片粉 4g 樟脑 2g 八角茴香油 2g 苯甲酸钠(中粉) 2g ───────────────── 制 成 1000片 【制成】 取甘草浸膏烘干,研碎,加苯甲酸钠、阿片粉均匀混合制成颗粒后,加 入用少量乙醇溶解的樟脑与八角茴香油,混匀压制成片,即得。 【性状】 本品为棕色片;有特臭,味甜;易吸潮。 【鉴别】 (1) 取本品2 片,研细,加水约7ml 混匀,加10%无水碳酸钠水溶液至 pH值约为9 ,用氯仿-异丙醇(3:1) 提取2 次,每次20ml,合并提取液,用少量氨试液 洗涤,再用少量水洗,然后浓缩蒸干,加甲醇0.3ml 使溶解,作为供试品溶液;另取吗 啡对照品适量,加甲醇溶解,制成每1ml 中含2mg 的溶液,作为对照品溶液。照薄层色 谱法(附录Ⅴ B)试验,取上述两种溶液各10μl ,分别点于同一硅胶G薄层板上,以 醋酸乙酯-甲醇-浓氨溶液(35:10:5) 为展开剂,展开后,晾干,喷以碘化铋钾试液, 使显色,供试品应出现与吗啡对照品位置相同,色泽亦相同的斑点。 (2) 取本品的细粉适量,加水使溶解,滤过,取滤液少量加稀硫酸1 ~2 滴,立即 析出极易溶于浓氨溶液的浅棕色沉淀。 【检查】 除崩解时限不检查外,应符合片剂项下有关的各项规定(附录Ⅰ A)。 【含量测定】 照高效液相色谱法(附录Ⅴ D)测定。 色谱条件与系统适用性试验 用十八烷基硅烷键合硅胶为填充剂;0.1mol/L磷酸二 氢钠溶液-甲醇(5:1) 为流动相,流速为每分钟1.2ml;检测波长为254nm 。理论板数按吗 啡峰计算应不低于1000。 测定法 取本品20片,精密称定,研细;精密称取适量(约相当于1 片重),置 小蒸发皿中,加水数滴,研磨均匀;用水7ml 定量转移至100ml 分液漏斗中,加10%无 水碳酸钠水溶液2.5ml ,加氯化铵0.2g,使溶解后,用氯仿-异丙醇(3:1) 混合液提取 5 次,第一次15ml,其他各次均为10ml,提取时用力振摇,合并提取液;用含1 滴浓氨溶 液的水10ml洗涤,然后用水再洗两次,每次10ml,氯仿层静置2 小时;三次水溶液 分别再用氯仿-异丙醇(3:1) 混合液10ml回提一次,回提液与原提取液合并。总提取液 中加入无水碳酸钠适量,滤入磨口锥形瓶中,减压蒸干溶,趁热加入甲醇2ml,密塞,使 残留物溶于甲醇中,摇匀,取10μl 注入液相色谱仪,记录色谱图;另取无水吗啡对照品 适量,精密称定,加甲醇制成每1ml 中含0.2mg 的溶液,同法测定。按外标法以峰面积计算, 即得。 【类别】 祛痰镇咳药。 【贮藏】 密封,在干燥处保存。
