一种融合蛋白及其应用的制作方法

本发明涉生物医药技术领域,尤其涉及一种融合蛋白及其应用。



背景技术:

磷脂酸肌醇-3-激酶(phosphoinositide3-kinases,pi3k)是一种胞内磷脂酰肌醇激酶接受来自酪氨酸激酶和g蛋白偶联受体的信号后,在质膜处将底物小分子ptdins(4,5)p2催化形成ptdins(3,4,5)p3(以下简称pi3p)。pi3p激活akt等下游蛋白。该信号通路广泛存在与各种细胞,是细胞内重要的促生存信号通路,调节细胞增殖、分化、生长、存活、代谢和迁移等多种活性。研究表明,pi3k/akt通路异常与人类肿瘤、神经退行性疾病、胰岛素抵抗相关疾病如ii型糖尿病、心脑血管疾病等的发生、发展密切相关。通过对该信号通路的快速、有效以及特异性的开关控制,有助于了解信号的级联传递规律,对于研究与该信号通路密切相关的疾病具有十分重要的意义。

传统方法采用化学物质(如igf-1)激活pi3k/akt通路用于研究正常细胞、肿瘤细胞的迁移和细胞形态变化等,该方法时空分辨率较低,且诱导方法比较复杂。

光遗传学作为一门新兴的学科在以神经科学为主的很多领域被广泛应用,光遗传学技术结合蛋白的遗传学表达与激光的光控和成像,可实现细胞内特定信号通路分子的快速激活与调控。目前已经实现了对pi3k/akt通路的光学调控,如idevall-hagreno等人采用光控工具来控制pi3k在质膜处的产生,见图1。该方法将pi3k的催化亚基p110α利用光控蛋白作用对cibn/cry2将其拉到质膜附近,从而催化pi3p的形成。该方法比传统化学小分子诱导方法简便,时空分辨率较高,但是需要在细胞中需要表达2个融合蛋白,即cibn-caax和mch-cry2-ish2达到最佳效果所需的每种蛋白的表达量需要耗费大量时间精力去摸索,而且ish2是来源于pi3k亚基,过多表达可能对诱导结果产生影响,不利于pi3k/akt通路的控制。



技术实现要素:

有鉴于此,本发明的目的在于提供一种融合蛋白及其应用。本发明提供的融合蛋白可快速有效、高时空分辨率地局部激活pi3k/akt信号通路。

为了实现本发明的目的,本发明采用如下技术方案:

本发明提供了一种融合蛋白,包括质膜定位序列、dab1蛋白、光敏蛋白和荧光蛋白;所述质膜定位序列为lyn蛋白定位序列或kras蛋白定位序列。

在一些实施方案中,所述lyn蛋白定位序列为:

(1)seqidno:1所示的氨基酸序列;

或(2)seqidno:1所示的氨基酸序列经缺失、取代或添加一个或多个氨基酸残基获得具有同等功能的氨基酸序列。

在一些实施方案中,所述kras蛋白定位序列为:

(1)seqidno:2所示的氨基酸序列;

或(2)seqidno:2所示的氨基酸序列经缺失、取代或添加一个或多个氨基酸残基获得具有同等功能的氨基酸序列。

在一些具体实施例中,所述光敏蛋白为隐花色素cry2olig蛋白。

在一些具体实施例中,所述荧光蛋白为红色荧光蛋白mcherry或黄色荧光蛋白mcit。

在一些实施方案中,所述融合蛋白具有如seqidno:3~5所示的氨基酸序列或在seqidno:3~5所示的氨基酸序列中取代、缺失和/或添加一个或几个氨基酸获得的具有同等功能的氨基酸序列。

在一个具体实施例中,所述融合蛋白具有如seqidno:3所示的氨基酸序列。该融合蛋白由lyn蛋白定位序列、dab1蛋白、隐花色素cry2olig蛋白和红色荧光蛋白mcherry组成,即lyn-dab1-cry2olig-mch。

在一个具体实施例中,所述融合蛋白具有如seqidno:4所示的氨基酸序列。该融合蛋白由lyn蛋白定位序列、dab1蛋白、隐花色素cry2olig蛋白和黄色荧光蛋白mcit组成,即lyn-dab1-cry2olig-mcit。

在一个具体实施例中,所述融合蛋白具有如seqidno:5所示的氨基酸序列。该融合蛋白由dab1蛋白、隐花色素cry2olig蛋白、黄色荧光蛋白mcit和kras蛋白定位序列组成,即dab1-cry2olig-mcit-kras。

本发明还提供了编码本发明所述融合蛋白的dna分子。

在一些实施方案中,本发明提供了编码具有seqidno:3所示的氨基酸序列的dna分子,其核苷酸序列如seqidno.6。

在一些实施方案中,本发明提供了编码具有seqidno:4所示的氨基酸序列的dna分子,其核苷酸序列如seqidno.7。

在一些实施方案中,本发明提供了编码具有seqidno:5所示的氨基酸序列的dna分子,其核苷酸序列如seqidno.8。

由于密码子的简并性,除了seqidno.6~8所示的核苷酸序列,还可以存在很多种能够编码本发明所述的融合蛋白的dna分子。

本发明还提供了一种重组dna载体,包含本发明所述dna分子。

重组dna载体是一种表达载体,本领域技术人员将所述融合蛋白的dna分子克隆到表达载体中,转化宿主细胞,通过诱导表达获得融合蛋白。所述表达载体包括pcag系列载体。在一些实施方案中,所述表达载体为pcag系列中的pcag-dsred2。

本发明还提供了一种宿主细胞,包含本发明所述重组dna载体。

进一步地,本发明还提供了所述融合蛋白在制备pi3k信号通路激活剂中的应用。所述与pi3k/akt信号通路相关的疾病包括神经退行性疾病、心脑血管疾病和糖尿病

研究表明,本发明提供的融合蛋白可以分别定位于质膜lipidraft(脂筏)/non-lipidraft(非脂筏)区域的lyn-opto-dab1/opto-dab1-kras,可以更加精细的研究性质不同的质膜区域中pi3k激酶活化的效果。经研究发现本发明融合有特定质膜定位序列的lyn-opto-dab1/opto-dab1-kras蛋白,与opto-dab1融合蛋白相比,仅有质膜上的扩散过程,不含胞浆快速扩散的过程,空间分辨率更高。

本发明提供的融合蛋白将lyn的质膜定位序列、dab1蛋白与光敏蛋白及荧光蛋白融合,通过光照可局部地、高时空分辨率地实现pi3k/akt信号通路的快速激活与调控。对该信号通路快速、有效以及特异性的开关控制,有助于了解信号的级联传递规律,对于研究与该信号通路密切相关的疾病具有十分重要的意义。

具体实施方式

本发明公开了一种融合蛋白及其应用,本领域技术人员可以借鉴本文内容,适当改进工艺参数实现。特别需要指出的是,所有类似的替换和改动对本领域技术人员来说是显而易见的,它们都被视为包括在本发明。本发明的方法及应用已经通过较佳实施例进行了描述,相关人员明显能在不脱离本发明内容、精神和范围内对本文所述的方法和应用进行改动或适当变更与组合,来实现和应用本发明技术。

对所公开的实施例的说明,使本领域专业技术人员能够实现或使用本发明。对这些实施例的多种修改对本领域的专业技术人员来说将是显而易见的,本文中所定义的一般原理可以在不脱离本发明的精神或范围的情况下,在其它实施例中实现。因此,本发明将不会被限制于本文所示的这些实施例,而是要符合与本文所公开的原理和新颖特点相一致的最宽的范围。

本发明采用的试材皆为普通市售品,皆可于市场购得。

下面结合实施例,进一步阐述本发明:

实施例1lyn-dab1-cry2olig-mch融合蛋白的制备

利用分子克隆基本技术,将编码lyn蛋白质膜定位序列、dab1蛋白与光敏感蛋白cry2olig及红色荧光蛋白mch(mcherry)的dna序列连接起来,导入表达载体pcag-dsred2中,获得重组表达载体,转化入宿主菌大肠杆菌,诱导表达获得融合蛋白(结构示意图见图2),具有如seqidno:3所示的氨基酸序列,核苷酸序列如seqidno.6所示。

实施例2lyn-dab1-cry2olig-mcit融合蛋白的制备

利用分子克隆基本技术,将编码lyn蛋白质膜定位序列、dab1蛋白与光敏感蛋白cry2olig及黄色荧光蛋白mcit(mcit)的dna序列连接起来,导入表达载体pcag-dsred2中,获得重组表达载体,转化入宿主菌大肠杆菌,诱导表达获得融合蛋白,具有如seqidno:4所示的氨基酸序列,核苷酸序列如seqidno.7所示。

实施例3dab1-cry2olig-mcit-kras融合蛋白的制备

利用分子克隆基本技术,将编码dab1蛋白、光敏感蛋白cry2olig、黄色荧光蛋白mcit(mcit)和kras蛋白质膜定位序列的dna序列连接起来,导入表达载体pcag-dsred2中,获得重组表达载体,转化入宿主菌大肠杆菌,诱导表达获得融合蛋白,具有如seqidno:5所示的氨基酸序列,核苷酸序列如seqidno.8所示。

实施例4

首先,克隆akt-ph-mch基因片段,将其转入表达载体pcag-dsred2中,获得含akt-ph-mch的真核表达载体。

分别将实施例2含lyn-opto-dab1-mcit的重组表达载体和含akt-ph-mch的表达载体通过lipofectamine2000试剂盒转染培养在玻璃底的培养皿中的cos7细胞(购自atcc公司),过夜培养后,用不含血清的、不含酚红的培养基(购自gibco公司)饥饿细胞16小时。利用商业化的共聚焦显微镜(比如zeisslsm780共聚焦显微镜),利用红色荧光(可以561nm激发,580-620nm接收荧光),找到表达的akt-ph-mch探针的细胞。实验中,用488nm/561nm扫描一块区域,荧光共聚焦成像结果见图3。

采用相同的方法对融合蛋白dab1-cry2olig-mcit-kras进行上述实验,结果见图4。

由图3~4结果可知,随着时间从0s到200s,akt-ph-mch发生向质膜转移的现象,结果表明,lyn-dab1-cry2olig-mcit和dab1-cry2olig-mcit-kras均能成功地激活pi3k通路,并在质膜处产生了大量的pi3p小分子。

实施例5

将实施例1含lyn-opto-dab1-mch的重组表达载体通过lipofectamine2000试剂盒转染培养在玻璃底的培养皿中的cos7细胞(购买自atcc公司),过夜培养后,用不含血清的、不含酚红的培养基(购自gibco公司)饥饿细胞16小时。利用商业化的共聚焦显微镜(比如zeisslsm780共聚焦显微镜),利用红色荧光(可以561nm激发,580-620nm接收荧光),找到表达opto-dab1-mch的细胞。选取细胞边缘一小块区域(比如一个直径为6.6μm的圆形区域),用0.2%功率(根据需要确定大小)的488nm激发线以每个像素0.79μs停留时间的方式做10次扫描,进行激活,然后利用红色荧光通道及明场通道分别观察细胞荧光和明场下的形态。每5秒成一次像。可见在200s处,圆形区域及附近的细胞膜运动很明显,出现皱褶及向细胞外空间延伸,见图5。

由图5结果可知,用488nm激发光快速扫描白色示意的圆圈区域(即激活区域),即可观察到激活区域红色信号的增强,随后在明场图中可观察到细胞质膜的变形,结果显示,本发明提供的融合蛋白可局部地、高时空分辨率地实现pi3k/akt信号通路的快速激活,可实现高时空分辨率地观察细胞形态变化。

以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
