1.本发明涉及含有2911植物元的组合物和含有2911植物元的金银花提取物的制备方法。


背景技术:

2.中药金银花为忍冬科植物忍冬(lonicera japonica thunb)的干燥花蕾,金银花性甘、寒,归肺、心、胃经,其药用历史悠久,有“中药中的抗生素”之称,具有清热解毒、疏散风热的功效。
3.中国药典2020版一部公布了金银花露成方制剂和单味制剂标准规格。
4.标准规格1、取金银花62.5g,用水蒸气蒸馏,收集蒸馏液约1000ml,取蒸馏液,调节ph值至约4.5,加矫味剂适量,滤过,制成1000ml,灌封,灭菌,或灭菌,灌封,即得。
5.标准规格2、取金银花100g,用水蒸气蒸馏,收集蒸馏液1400ml,加入单糖浆适量至1600ml,滤过,灌封,灭菌;或取蔗糖140g及苯甲酸钠3.2g,加水使溶解,兑入蒸馏液中,加水至1600ml,混匀,加适量枸橼酸调节ph值至4.0~4.5,混匀,滤过,灭菌,灌封,即得。
6.标准规格3、取金银花100g,用水蒸气蒸馏,收集蒸馏液1600ml,加入蔗糖30g,混匀,滤过,灌封,灭菌,即得。
7.金银花露成方制剂和单味制剂的鉴别的对照品是芳樟醇。
8.按照中国药典2020年版一部所制备的产品包括葵花药业集团(襄阳)隆中有限公司出品的金银花露(国药准字z42020517)(其成份为金银花、蔗糖、苯甲酸钠、枸橼酸)和湖北午时药业股份有限公司出品的金银花露(国药准字z42021971)(其成份为金银花、蔗糖、苯甲酸钠、枸橼酸)。
9.近年来,对金银花化学成分和生物活性物质研究较多,其现代药理学研究表明,金银花具有广谱抗菌、抗病毒、解热、抗炎、保肝利胆、止血等药理作用。然而,现有研究成果对于金银花的有效成分主要集中于其含有的绿原酸、异绿原酸和咖啡酰奎宁酸等化学单体。如上所述的中国药典中用芳樟醇作为对照品来鉴别金银花露成方制剂和单味制剂的规定,体现了现有技术和产品对金银花有效成分利用的局限。
10.中国授权专利(授权公告号cn 104922187b)公开了一种金银花口服液及其制备方法;其发明是基于“金银花的有效成份包括有机酸类、黄酮类、三萜皂苷类、挥发油类及微量元素等,其中以绿原酸和挥发油为主”。cn 104922187b公开的制备方法是将水蒸汽蒸馏法提取分为两次,首次1小时完成挥发性成份的提取,蒸馏器内煎液过滤,药渣进行二次提取;该提取方法得到的提取液比当时的现有方法中的绿原酸的含量提高约5%。
11.综上所述,无论是中国药典的金银花露,还是专利公开的金银花口服液,都是为了尽量保留或提高金银花提取液中有机酸特别是绿原酸的含量。而为了达到这个目的,cn 104922187b要求金银花煎煮的时间须在一小时内完成;亦即是说,当金银花煎煮的时间超过一个小时,绿原酸会被破坏,降低金银花口服液的质量。
12.微小核酸(microrna)是一类由内源基因编码的长度约为20-25个核苷酸的非编码单链rna分子,它们在动植物中参与转录后基因表达调控。
13.研究发现将金银花10克在100毫升水中煎煮30分钟得到大约50毫升的煎煮液;在煎煮液中发现微小核酸2911(mir2911)分子的存在(mir2911的核苷酸序列为:5

),rt-qpcr和northern印迹检测显示金银花中mir2911的浓度为0.34-0.40pmol/g,金银花煎煮液中mir2911的浓度为0.06-0.08pmol/g(0.2g金银花/ml)(zhou 2015)。进一步的研究发现含有mir2911的煎煮液能够抑制流感甲型病毒(zhou2015)。
14.微小核酸分子一般是不稳定的。尽管mir2911分子是一个特例,具有一定的稳定性,但是,把金银花在水中煎煮的时间限制在30分钟而制取含有mir2911分子的煎煮液(zhou2015),这一做法说明了更长时间的煎煮很可能还是会对微小核酸如mir2911分子的稳定性造成不利影响;同时,是否可以通过调节煎煮的条件如时间来提高煎煮液中微小核酸如mir2911分子的含量,也是未知的;煎煮时间如何影响煎煮液中微小核酸如mir2911分子的含量更是无法预测的。

20.为使本发明的目的、技术方案和优点更加清楚明了,下面结合具体实施方式,对本发明进一步详细说明。应该理解,这些描述只是示例性的,而并非要限制本发明的范围。此外,在以下说明中,省略了对公知结构和技术的描述,以避免不必要地混淆本发明的概念。
21.在本技术中,“植物元”指一类来源于植物,通过煎煮或其他特殊工艺获得的,对人体健康有益的含有特定序列的微小核酸(microrna)的功能分子。例如,“2911植物元”指来源于中药金银花或其他植物的花、茎、叶等部分(如茶叶等),含有修饰基团(如甲基化修饰、糖基化修饰等等)的微小核酸2911(mir2911)及其衍生物。
22.本发明的发明人探索了2911植物元在金银花提取过程中的稳定性,特别是煎煮时间对金银花提取液和提取物固体粉末中2911植物元含量的影响。研究发现2911植物元在金银花提取过程中具有稳定性,在煎煮时间为80分钟时,2911植物元的含量达到最高值的75%,而煎煮30分钟,2911植物元的含量则均不到最高值的20%;煎煮60分钟,2911植物元
的含量大约为最高值的40%。
23.本发明的发明人同时也探索了煎煮时间对金银花提取液和提取物固体粉末所制备的组合物特别是饮料的口感的影响。研究发现煎煮时间超过180分钟(如200分钟)时,口感会有显著下降。
24.从这两者的综合效果,本发明提供了含有2911植物元的金银花提取液和提取物固体粉末的制备方法。

26.粉粹干燥金银花样品得到金银花粉末;一些实施例中,金银花粉末的大小为10-200目;
27.将金银花粉末置于装有水的蒸馏器中进行煎煮得到煎煮液,水量倍数范围为5-50倍,煎煮液温度范围为90-121℃,时间范围为15-200分钟;(注:此处温度范围主要考虑了高原地区和高压环境)

101.实施例1中所得到的金银花提取液在真空-0.1mpa/50℃条件下浓缩,180℃喷雾干燥,收粉。固体粉末得率(得到的固体粉末的重量除以金银花干花原料的重量)是12-29%。采用荧光rt-pcr检测方法测定2911植物元的含量。金银花干花中2911植物元的含量为589-880pmol/g。
