本发明属于微生物代谢工程领域,具体涉及改造微生物代谢途径,从单糖直接合成植物生长素的方法。
背景技术:
植物生长素是植物中最为重要的信息素之一,是植物体内普遍存在的内源生长素,对于植物体的生长发育至关重要,具有促进细胞分裂、维管束分化、茎伸长、顶端优势、种子发芽、侧根和不定根形成、根瘤形成、子房壁生长、种子和果实生长、坐果等,在农业生产上具有广阔的应用前景与经济地位。这些植物生长素包括吲哚乙酸、苯乙酸、对羟基苯乙酸等。
目前已商品化的植物生长素均为通过化学法合成。化学法合成生长素需涉及到多个反应步骤导致合成成本较高。通过生物技术手段,实现微生物发酵单糖合成各种生长素,可有效的降低成本。
技术实现要素:
本发明的目的在于提供一种微生物体内合成植物生长素的方法,在微生物内构建一条由单糖转化到植物生长素的代谢途径。
为实现上述目的,本发明提供了在大肠杆菌和毕赤酵母合成植物生长素苯乙酸、对羟基苯乙醛和吲哚乙酸的方法,主要步骤为:
(1)敲除基因构建菌株gdy
以大肠杆菌mg1655菌株出发,依次敲除该菌株中的trpr基因和七个醛还原酶基因(dkgb、yeae、yqhc、yqhd、dkga、yahk和yjgb),敲除上述8个基因的菌株命名为gdy。
(2)构建产苯乙酸工程菌gdy1
将来自酿酒酵母菌的kdc和aro8基因及来自大肠杆菌的aldh基因,重组到载体pet28a(+)上,得到载体pdy001。将来自大肠杆菌的arogfbr基因和pheafbr基因,重组到载体pbbr1mcs1上,得到载体pdy002。将载体pdy001和pdy002转入到大肠杆菌gdy中,实现上述5个基因在大肠杆菌中的高量表达,得到大肠杆菌工程菌株gdy1。表达解除反馈抑制的arogfbr基因和pheafbr基因用于加强大肠杆菌胞内苯丙氨酸的合成。表达aro8用于催化苯丙氨酸转化苯丙酮酸,表达kdc用于催化苯丙酮酸脱羧为苯乙醛,表达aldh用于随后催化苯乙醛氧化形成苯乙酸。上述构建的gdy1工程菌可有效的将单糖转化为苯乙酸。
(3)构建产对羟基苯乙酸工程菌gdy2
将来自大肠杆菌的arogfbr基因和tyrafbr基因,重组到载体pbbr1mcs1上,得到载体pdy003。将pdy003和前面构建好的载体pdy001同时转入到大肠杆菌gdy中,实现arogfbr、tyrafbr、kdc、aro8和aldh5个基因在大肠杆菌中的高量表达,得到大肠杆菌工程菌株gdy2。表达解除反馈抑制的arogfbr基因和tyrafbr基因用于加强大肠杆菌胞内酪氨酸的合成。表达aro8用于催化酪氨酸转化对羟基苯丙酮酸,表达kdc用于催化对羟基苯丙酮酸脱羧为对羟基苯乙醛,表达aldh用于随后催化对羟基苯乙醛氧化形成对羟基苯乙酸。上述构建的gdy2工程菌可有效的将单糖转化为对羟基苯乙酸。
(4)构建产吲哚乙酸工程菌gdy3
将来自大肠杆菌的arogfbr、trpefbr和serafbr基因,重组到载体pbbr1mcs1上,得到载体pdy004。trpabcd基因(包含trpa、trpb、trpc和trpd四个基因)重组到载体pdy001上,得到载体pdy005。将载体pdy004和pdy005同时转入到大肠杆菌gdy中,实现arogfbr、trpefbr、serafbr、trpabcd、kdc、aro8和aldh7个基因在大肠杆菌中的高量表达,得到大肠杆菌工程菌株gdy3。表达解除反馈抑制的arogfbr、trpefbr和serafbr基因及trpabcd基因用于加强大肠杆菌胞内色氨酸的合成。表达aro8用于催化色氨酸转化为吲哚丙酮酸,表达kdc用于催化吲哚丙酮酸脱羧为吲哚乙醛,表达aldh用于随后催化吲哚乙醛氧化形成吲哚乙酸。上述构建的gdy3工程菌可有效的将单糖转化为吲哚乙酸。
(5)构建产植物生长素工程菌gdy4
将来自大肠杆菌的aldh基因,重组到载体pgapza,得到载体pdy006。将来自酿酒酵母菌的kdc和aro8基因,分别重组到载体pgapza,得到载体pdy14和pdy15。将pdy14和pdy15分别以bamhi和bglii双酶切,得到kdc基因表达框(包含gap启动子,kdc基因和aox1tt)和aro8基因表达框(包含gap启动子,aro8基因和aox1tt)。将kdc基因表达框和aro8基因表达框分别重组到pao815载体,得到载体pdy007和pdy008。将载体pdy006、pdy007和pdy008分别线性化后,依次导入毕赤酵母gs115中,实现aro8、aldh和kdc基因的组成型表达,该工程菌命名为gdy4。表达aro8用于催化苯丙氨酸、酪氨酸和色氨酸转化为苯丙酮酸、对羟基苯丙酮和吲哚丙酮酸。表达kdc用于催化苯丙酮酸、对羟基苯丙酮酸和吲哚丙酮酸分别转化为苯乙醛、对羟基苯乙醛和吲哚乙醛。表达aldh用于催化苯乙醛、对羟基苯乙醛和吲哚乙醛分别转化为苯乙酸、对羟基苯乙酸和吲哚乙酸。
本发明的优点是在微生物中引入外源基因构建一条合成植物生长素的代谢途径,最终实现以单糖为原料合成植物生长素的目的,不需要通过过多的化学合成反应,减少化学合成过程中有毒物质的释放,也减少了合成成本。同时,由于微生物生长速度快,遗传操作技术成熟,且发酵过程不会对环境造成污染破坏。本发明的毕赤酵母菌株可作为农业微生物,直接利用环境土壤中的营养物质合成植物生长素,从而促进植物的生长。

具体实施方式
以下实施例用于进一步说明本发明,但不应理解为对本发明的限制。
实施例1构建产苯乙酸大肠杆菌工程菌
1.基因敲除菌株gdy的构建
以pkd4质粒为模板,pcr扩增打靶片段,该片段包含(kanr序列和与待敲除目的基因两端互补的同源臂);将扩增的打靶片段电转到带有pkd46质粒的大肠杆菌中。利用pkd46质粒上带有的red同源重组系统,使待敲除目的基因与打靶片段进行重组,目的基因被抗性基因取代。随后将温敏质粒pcp20导入菌株,利用该质粒上的flp翻转酶去除目的菌株染色体上的抗性基因。利用上述方法依次敲除大肠杆菌mg1655菌株中的trpr、dkgb、yeae、yqhc、yqhd、dkga、yahk和yjgb基因,获得大肠杆菌dgy。所述trpr基因具有seqidno:25所示的核苷酸序列编码;所述trpr基因具有seqidno:26所示的氨基酸序列;所述dkgb基因具有seqidno:27所示的核苷酸序列编码;所述aldh基因具有seqidno:28所示的氨基酸序列;所述yeae基因具有seqidno:29所示的核苷酸序列编码;所述yeae具有seqidno:30所示的氨基酸序列;所述yqhc基因具有seqidno:31所示的核苷酸序列编码;所述yqhc基因具有seqidno:32所示的氨基酸序列;所述yqhd基因具有seqidno:33所示的核苷酸序列编码;所述yqhd基因具有seqidno:34所示的氨基酸序列;所述dkga基因具有seqidno:35所示的核苷酸序列编码;所述dkga基因具有seqidno:36所示的氨基酸序列;所述yahk基因具有seqidno:37所示的核苷酸序列编码;所述yahk基因具有seqidno:38所示的氨基酸序列;所述yjgb基因具有seqidno:39所示的核苷酸序列编码;所述yjgb基因具有seqidno:40所示的氨基酸序列;敲除基因用的引物为:trpr-f:atggcccaacaatcaccctattcagcagcgatggcagaacaggtgtaggctggagctgcttc

2.载体pdy001的构建
利用引物kdc-xbai(atctctagatttaagaaggagatataatggcacctgttacaattgaaaagttc)和kdc-nhei-bamhi(tctggatccgctagcctattttttatttcttttaagtgccgctg)pcr扩增来自酿酒酵母菌yph499的kdc基因,随后将扩增的片段以xbai和bamhi插入到pet28a(+)中得到载体pdy01。利用引物aldh-xbai(atctctagatttaagaaggagatataatgaattttcatcatctggcttactg)和aldh-nhei-bamhi(tcaggatccgctagctcaggcctccaggcttatcc)pcr扩增来自大肠杆菌的aldh基因,利用xbai和bamhi插入到pet28a(+)中得到载体pdy02。利用引物aro8-xbai(aactctagatttaagaaggagatataatgatgactttacctgaatcaaaagacttttc)和aro8-nhei-bamhi(acaggatccgctagcctatttggaaataccaaattcttcgtataa)pcr扩增来自酿酒酵母菌yph499的kdc基因,随后将扩增的片段以xbai和bamhi插入到pet28a(+)中得到载体pdy03。利用xbai和xhoi双酶切pdy02得到aldh表达框,将其插入以nhei和xhoi双酶切的载体pdy01上,得到载体pdy04。利用xbai和xhoi双酶切pdy03得到aro8表达框,将其插入以nhei和xhoi双酶切的载体pdy04上,得到载体pdy001。所述kdc基因具有seqidno:19所示的核苷酸序列编码;所述kdc具有seqidno:20所示的氨基酸序列;所述aldh基因具有seqidno:21所示的核苷酸序列编码;所述aldh基因具有seqidno:22所示的氨基酸序列;所述aro8基因具有seqidno:23所示的核苷酸序列编码;所述aro8基因具有seqidno:24所示的氨基酸序列;
3.载体pdy002的构建
解除反馈抑制的arogfbr和pheafbr基因委托金唯智生物科技有限公司合成,以合成的基因为模板利用引物arogfbr-xbai(gtatctagaaagaggagatataatgaattatcagaacgacgatttacgc)和arogfbr-spei-bamhi(tatggatccactagtttacccgcgacgcgctttta)pcr扩增arogfbr基因,随后将扩增的片段以xbai和bamhi插入到pet28a(+)中得到载体pdy05。以合成的基因为模板,利用引物pheafbrr-xbai(gtatctagaaagaggagatataatgacatcggaaaacccgttactg)和pheafbr-spei-sali(atatgtcgacactagttcacaacgtggttttcgccgga)pcr扩增来pheafbr基因,随后将扩增的片段以xbai和sali插入到pet28a(+)中得到载体pdy06。利用xbai和xhoi双酶切pdy06得到pheafbr表达框,将其插入以spei和xhoi双酶切的载体pdy05上,得到载体pdy07。利用xbai和xhoi双酶切pdy07得到arogfbr和pheafb表达框,将其插入以xbai和xhoi双酶切的载体pbbr1mcs1上,得到载体pdy002。所述arogfbr基因具有seqidno:1所示的核苷酸序列;所述arogfbr基因具有与seqidno:2所示的氨基酸序列;所述pheafbr基因具有seqidno:3所示的核苷酸序列编码;所述pheafbr基因具有seqidno:4所示的氨基酸序列;4.将重组载体pdy001和pdy002同时导入大肠杆菌gdy中,得到gdy1工程菌。该工程菌可实现arogfbr、pheafbr、kdc、aldh和aro8基因的高表达,可有效的将单糖转化为苯乙酸。
实施例2构建产对羟基苯乙酸大肠杆菌工程菌
1.载体pdy003的构建
解除反馈抑制的tyrafbr基因委托金唯智生物科技有限公司合成,以该合成的基因为模板利用引物tyrafbrr-xbai(gtatctagaaagaggagatataatggttgctgaattgaccgcat)和tyrafbr-spei-sali(atatgtcgacactagtttactggcgattgtcattcgcc)pcr扩增来tyrafbr基因,随后将扩增的片段以xbai和sali插入到pet28a(+)中得到载体pdy08。利用xbai和xhoi双酶切pdy08得到tyrafbr表达框,将其插入以spei和xhoi双酶切的载体pdy05上,得到载体pdy09。利用xbai和xhoi双酶切pdy09得到arogfbr和tyrafbr表达框,将其插入以xbai和xhoi双酶切的载体pbbr1mcs1上,得到载体pdy003。所述tyrafbr基因具有seqidno:5所示的核苷酸序列编码;所述tyrafbr基因具有seqidno:6所示的氨基酸序列;
2.将重组载体pdy001和pdy003同时导入大肠杆菌gdy中,得到gdy2工程菌。该工程菌可实现arogfbr、tyrafbr、kdc、aldh和aro8基因的高表达,可有效的将单糖转化为对羟基苯乙酸。
实施例3构建产吲哚乙酸大肠杆菌工程菌
1.载体pdy004的构建
解除反馈抑制的trpefbr和serafbr基因委托金唯智生物科技有限公司合成。以合成的基因为模板,利用引物trpefbr-xbai(gtatctagaaagaggagatataatgcaaacacaaaaaccgactctc)和trpefbr-spei-bamhi(tatggatccactagttcagaaagtctcctgtgcatgatg)pcr扩增trpefbr基因,随后将扩增的片段以xbai和bamhi插入到pet28a(+)中得到载体pdy10。以合成的基因为模板,利用引物serafbr-xbai(gtatctagaaagaggagatataatgacatcggaaaacccgttactg)和serafbr-spei-sali(atatgtcgacactagttcacaacgtggttttcgccgga)pcr扩增来serafbr基因,随后将扩增的片段以xbai和sali插入到pet28a(+)中得到载体pdy11。利用xbai和xhoi双酶切pdy10得到trpefbr表达框,将其插入以spei和xhoi双酶切的载体pdy01上,得到载体pdy12。利用xbai和xhoi双酶切pdy10得到trpefbr表达框,将其插入以spei和xhoi双酶切的载体pdy12上,得到载体pdy13。利用xbai和xhoi双酶切pdy13得到serafbr和trpefbr表达框,将其插入以xbai和xhoi双酶切的载体pbbr1mcs1上,得到载体pdy004。所述trpefbr基因具有seqidno:7所示的核苷酸序列编码;所述trpefbr基因具有seqidno:8所示的氨基酸序列;serafbr所述serafbr基因具有seqidno:9所示的核苷酸序列编码;所述serafbr基因具有seqidno:10所示的氨基酸序列;

以大肠杆菌mg1655基因组为模板,利用引物trpabcd-xbai(ccatctagatttaagaggagatataatggctgacattctgctgctcgata)和trpabcd-xhoi(gattctcgagttaactgcgcgtcgccgct)pcr扩增trpabcd基因(包含trpa、trpb、trpc和trpd四个基因),随后将扩增的片段以xbai和xhoi插入到pdy001中,得到载体pdy005。所述trpa具有seqidno:11所示的核苷酸序列编码;trpa基因具有seqidno:12所示的氨基酸序列;所述trpb具有seqidno:13所示的核苷酸序列编码;所述trpb基因具有seqidno:14所示的氨基酸序列;所述trpc具有seqidno:15所示的核苷酸序列编码;所述trpc基因具有seqidno:16所示的氨基酸序列;所述trpd具有seqidno:17所示的核苷酸序列编码所述;所述trpd基因具有seqidno:18所示的氨基酸序列;
3.将重组载体pdy004和pdy005同时导入大肠杆菌gdy中,得到gdy3工程菌。该工程菌可实现arogfbr、trpefbr、serafbr、trpabcd、kdc、aldh和aro8基因的高表达,可有效的将单糖转化为吲哚乙酸。
实施例4构建产植物生长素毕赤酵母工程菌gdy4
以大肠杆菌mg1655基因组为模板,利用引物aldh-bstbi(agcattcgaaatgaattttcatcatctggcttactggc)和aldh-xhoi(gaacctcgagtcaggcctccaggcttatccagat)pcr扩增aldhr基因,随后将扩增的片段以bstbi和xhoii插入到pgapza中得到载体pdy006。以酿酒酵母yph499基因组为模板,利用引物kdc-bstbi(agcattcgaaatggcacctgttacaattgaaaagttc)/kdc-xhoi(cttcctcgagctattttttatttcttttaagtgccgctg)、引物aro8-bstbi(agcattcgaaatgactttacctgaatcaaaagacttttc)/aro8-xhoi(cttcctcgagctatttggaaataccaaattcttcgtataa)pcr扩增kdc和aro8基因,随后将扩增的片段以bstbi和xhoi分别插入到pgapza中,得到载体pdy14和pdy15。将pdy14和pdy15分别以bamhi和bglii双酶切,得到kdc基因表达框(包含gap启动子,kdc基因和aox1tt)和aro8基因表达框(包含gap启动子,aro8基因和aox1tt)。将kdc基因表达框重组到经bamhi酶切的pao815载体,得到载体pdy007。将aro8基因表达框重组到经bamhi酶切的ppic3.5k载体,得到载体pdy008。将载体pdy006、pdy007和pdy008分别线性化后,依次导入毕赤酵母gs115中,实现aro8、aldh和kdc基因的组成型表达,该工程菌命名为gdy4。表达aro8用于催化苯丙氨酸、酪氨酸和色氨酸转化为苯丙酮酸、对羟基苯丙酮和吲哚丙酮酸。表达kdc用于催化苯丙酮酸、对羟基苯丙酮酸和吲哚丙酮酸分别转化为苯乙醛、对羟基苯乙醛和吲哚乙醛。表达aldh用于催化苯乙醛、对羟基苯乙醛和吲哚乙醛分别转化为苯乙酸、对羟基苯乙酸和吲哚乙酸。所述kdc基因具有seqidno:19所示的核苷酸序列编码;所述kdc具有seqidno:20所示的氨基酸序列;所述aldh基因具有seqidno:21所示的核苷酸序列编码;所述aldh基因具有seqidno:22所示的氨基酸序列;所述aro8基因具有seqidno:23所示的核苷酸序列编码;所述aro8基因具有seqidno:24所示的氨基酸序列;
实施例5大肠杆菌工程菌生产植物生长素的发酵实验
1、将37度lb中过夜培养的大肠杆菌工程菌gdy1、gdy2和gdy3,分别接种到50mlm9发酵培养基中(包含2%葡萄糖),进行30度摇瓶发酵,待菌体od600为0.8左右,加入0.1mmiptg进行目的基因的诱导表达。
2、发酵培养24小时后收集菌体,超声波破细胞,利用乙酸乙酯萃取产物。
3、利用旋转蒸发仪将溶剂乙酸乙酯旋干,再用一定量的乙酸乙酯溶解产物。
4、gc-ms(气相色谱-质谱联用仪)检测样品。
5、实验结果:大肠杆菌工程菌利用葡萄糖合成植物生长素的gc-ms检测结果如图1、图2和图3所示。图1为大肠杆菌工程菌gdy1发酵产物gc-ms检测结果,表明菌株gdy1可有效利用葡萄糖合成植物生长素苯乙酸,摇床产量为1.67g/l。图2为大肠杆菌工程菌gdy2发酵产物gc-ms检测结果,表明菌株gdy2可有效利用葡萄糖合成植物生长素对羟基苯乙酸,摇床产量为1.07g/l。图3为大肠杆菌工程菌gdy3发酵产物gc-ms检测结果,表明菌株gdy3可有效利用葡萄糖合成植物生长素对吲哚乙酸,摇床产量为1.16g/l。
实施例6毕赤酵母工程菌生产植物生长素的发酵实验
6、将30度ypd中过夜培养的毕赤酵母工程菌gdy4,接种到50mlypd发酵培养基中(包含2%葡萄糖),进行30度摇瓶发酵。
7、发酵培养28小时后收集菌体,超声波破细胞,利用乙酸乙酯萃取产物。
8、利用旋转蒸发仪将溶剂乙酸乙酯旋干,再用一定量的乙酸乙酯溶解产物。
9、gc-ms(气相色谱-质谱联用仪)检测样品。
10、实验结果:毕赤酵母工程菌利用葡萄糖合成植物生长素的gc-ms检测结果如图4所示。结果表明菌株gdy4可有效利用葡萄糖合成植物生长素苯乙酸,对羟基苯乙酸和吲哚乙酸,产量分别为108mg/l,36mg/l和33mg/l。
以上所述仅是本发明的优选实施方案,应当指出,对于本技术领域的技术人员,在不脱离本分明原理的前提下,还可以做出适当的改进,这些改进也应视为本发明的保护范围。
