本发明属于基因工程技术领域。



背景技术:

松芪是一种白藜芦醇衍生物,发现于葡萄、蓝莓等植物中,具有抗毒素及抗氧化剂的作用。葡萄等多种植物有编码白藜芦醇o-甲基转移酶(romt)的基因,表达的romt可以使白藜芦醇的羟基甲基化,产生松芪或紫檀芪。相比白藜芦醇,松芪和紫檀芪稳定性更强,其中松芪在预防及治疗癌症方面比白藜芦醇更有效,松芪的市场价格比白藜芦醇和紫檀芪更高,松芪的高效生物合成方法有重要价值。

目前发现的多数romt,其产物以紫檀芪为主(wangy,bhuiyamw,zhour,etal.annalsofmicrobiology,2015,65(2):817-826.),少数以松芪为主要产物的romt活性很低(jeongyj,anch,woosg,etal.enzymeandmicrobialtechnology,2014,54:8-14.)。表达葡萄来源romt(vvromt)的大肠杆菌工程菌,可催化白藜芦醇转化为松芪和紫檀芪,产物以紫檀芪为主,松芪仅有微量的积累。表达高粱来源romt(sbromt3)的大肠杆菌工程菌,产物虽然以松芪为主,但由于酶活性低,工程菌48h松芪产量仅有34mg/l。



技术实现要素:

本发明的目的是通过易错pcr等诱变技术改造酶的基因,然后根据特定的改造目的,筛选有价值的可以定向进化构建高效转化白藜芦醇生产松芪的romt突变体。

本发明通过定向进化构建的高效转化白藜芦醇生产松芪的romt突变体,由葡萄来源的vvromt基因,运用易错pcr方法,通过多轮重组和随机突变,经定向进化而获得的romt突变体。

本发明vvromt基因核苷酸突变方法:利用易错pcr技术在体外向vvromt基因引入核苷酸突变:

易错pcr的反应条件如下

上游引物f:5’-caggatccggatctggcaaatggtgtgatcagtg-3’;

下游引物r:5’-atgcggccgctcacggatacacttcaatcagactgcgcag-3’

pcr扩增条件:94℃3min;94℃1min,59℃1min,72℃2min,30个循环;72℃10min;

易错pcr扩增产物经dna纯化试剂盒纯化后,限制性内切酶bamhi和noti分别对易错pcr扩增产物和质粒prsf进行消化,连接,转化至大肠杆菌bl21(de3)感受态细胞;涂布于lb(含50μg/ml卡那霉素)平板,37℃培养12h后,构成romt突变文库;

利用上述同样的方法,以突变体基因为模板进行多轮易错pcr,构建突变文库。

本发明romt突变体的获得:

用灭菌牙签将lb平板上生长出的卡那霉素抗性转化子复制到lb平板,其中含50μg/ml卡那霉素,同时将对照菌大肠杆菌bl21(de3)/prsf-vvromt接种至lb平板,其中含50μg/ml卡那霉素,37℃培养12h;

96孔板筛选:向1.8ml/孔的96孔板加入100μllb(含50μg/ml卡那霉素)液体培养基,向其中接入保藏于lb(含50μg/ml卡那霉素)平板上的菌株(同时接入大肠杆菌bl21(de3)/prsf-vvromt作为对照),37℃,200rpm摇床中过夜培养,补加400μllb(含50μg/ml卡那霉素和0.125mmiptg)液体培养基到96孔板中,在18℃,200rpm摇床中培养24h,将诱导表达1天的96孔板发酵液4℃4000g离心5min,去除上清培养基,用300μl的改良m9(含50μg/ml卡那霉素、0.1mmiptg、228mg/l白藜芦醇)培养基重悬菌体,在18℃,200rpm摇床中培养48h,即得到带有表达romt基因的大肠杆菌bl21(de3)转化白藜芦醇的发酵液,将300μl带有表达romt基因的大肠杆菌bl21(de3)转化白藜芦醇的发酵液添加3μl6mhcl混匀,37℃培养箱温育30min,后用300μl乙酸乙酯萃取发酵液,取200μl乙酸乙酯层真空旋转离心干燥后用5μl无水乙醇重悬,取1μl做tlc分析,用imagej软件对薄层色谱检测图中松芪化合物点进行灰度分析,分析松芪产量,相同条件下,松芪产量明显比对照工程菌bl21(de3)/prsf-vvromt产量高的菌株为筛选目的菌株,相同条件下,对照工程菌bl21(de3)/prsf-vvromt仅产生微量松芪;

以易错pcr方法构建的突变文库为基础进行筛选,获得8株白藜芦醇产松芪效率明显提高的菌株,提取其中romt表达质粒,测定romt核苷酸序列,利用三联体密码子推测romt的氨基酸序列,氨基酸的改变方式及产松芪相对活性如表1所示,以对照工程菌bl21(de3)/prsf-vvromt(菌株编号vvromt)的产松芪相对活性为100%,根据imagej软件对tlc检测图的分析结果确定romt突变文库中工程菌产松芪的相对活性;

本发明高产松芪工程菌表达romt突变体氨基酸序列:seqidno.2、seqidno.3、seqidno.4、seqidno.5、seqidno.6、seqidno.7、seqidno.8。

本发明的定向进化属于非理性设计,是指针对某一蛋白质酶的基因,通过易错pcr等诱变技术改造酶的基因,然后根据特定的改造目的,筛选有价值的天然酶。目前,定向进化技术已成功用于提高酶的催化反应活性,改进底物特异性,提高热稳定性,对映立体选择性等方面。本发明酶活力的提高可以用表达突变型romt工程菌转化白藜芦醇生产松芪的效率表示,表达突变型romt工程菌产松芪效率的提高,即代表了突变型romt转化白藜芦醇产松芪活性的提高。

具体实施方式

本发明通过定向进化构建的高效转化白藜芦醇生产松芪的romt突变体,由葡萄来源的vvromt基因(genbank登录号fm178870)seqidno.1,运用易错pcr方法,通过多轮重组和随机突变,经定向进化而获得的romt突变体,在突变体的氨基酸序列中,包含氨基酸突变为phe311trp、phe311trp/ser67thr/gly300ala、phe311trp/val19leu、phe311tyr/glu330gln、phe311tyr/ser9thr、phe311tyr、phe311arg/asn46gln、phe311arg/cys69asn/val118lle。以表达对应romt突变体的工程菌转化白藜芦醇产松芪的产量表示,相比表达野生型vvromt的对照工程菌,这些突变体转化白藜芦醇产松芪的效率得到了提高。

本发明涉及到的培养基配方如下:

lb液体培养基(胰蛋白胨1%;酵母提取物0.5%;nacl1%;ph7.0)

lb固体培养基(胰蛋白胨1%;酵母提取物0.5%;nacl1%;琼脂2%;ph7.0)

改良m9培养基(na2hpo4·12h201.71%;kh2po40.3%;nacl0.05%;nh4cl0.1%;mgso40.036%;cacl20.0011%;甘油0.5%;酵母提取物0.125%)

培养基中单位为%(w/v)

薄层层析色谱(tlc)条件:使用烟台江友硅胶开发有限公司的薄层色谱硅胶板,型号为hsgf254,展开剂为二氯甲烷:丙酮体积比为40:2含0.1%甲酸。

液相色谱质谱联用(lc-ms)条件:利用安捷伦1260-6120液质联用系统对所提取的化合物中白藜芦醇和松芪进行定性和定量分析,产物通过反向色谱zorbaxsb-c18(粒径为1.8μm;2.1×50mm)进行分离,洗脱液为100%的乙腈含0.1%(v/v)的甲酸(溶液a)和0.1%(v/v)的甲酸(溶液b)。样品用50%的无水乙醇进行稀释,梯度洗脱程序如下:20%溶液a洗脱8min,20%-50%溶液a线性洗脱7min,50%溶液a洗脱7min,20%溶液a洗脱13min,流速为0.2ml/min,进样体积1μl,检测模式正离子模式,检测电压1.56kv,雾化气(n2)流速1.5l/min,干燥气(n2)压力100kpa,离子收集时间30ms,碰撞能量50%,ms扫描范围100-600m/z,通过外标法对样品进行定量分析。

96孔板筛选:向1.8ml/孔的96孔板加入100μllb(含50μg/ml卡那霉素)液体培养基,向其中接入保藏于lb(含50μg/ml卡那霉素)平板上的菌株(同时接入大肠杆菌bl21(de3)/prsf-vvromt作为对照),37℃,200rpm摇床中过夜培养,补加400μllb(含50μg/ml卡那霉素和0.125mmiptg)液体培养基到96孔板中,在18℃,200rpm摇床中培养24h,将诱导表达1天的96孔板发酵液4℃4000g离心5min,去除上清培养基,用300μl的改良m9(含50μg/ml卡那霉素、0.1mmiptg、228mg/l白藜芦醇)培养基重悬菌体,在18℃,200rpm摇床中培养48h,即得到带有表达romt基因的大肠杆菌bl21(de3)转化白藜芦醇的发酵液,将300μl带有表达romt基因的大肠杆菌bl21(de3)转化白藜芦醇的发酵液添加3μl6mhcl混匀,37℃培养箱温育30min,后用300μl乙酸乙酯萃取发酵液,取200μl乙酸乙酯层真空旋转离心干燥后用5μl无水乙醇重悬,取1μl做tlc分析,用imagej软件对薄层色谱检测图中松芪化合物点进行灰度分析,分析松芪产量,相同条件下,松芪产量明显比对照工程菌bl21(de3)/prsf-vvromt产量高的菌株为筛选目的菌株,相同条件下,对照工程菌bl21(de3)/prsf-vvromt仅产生微量松芪;

以易错pcr方法构建的突变文库为基础进行筛选,获得8株白藜芦醇产松芪效率明显提高的菌株,提取其中romt表达质粒,测定romt核苷酸序列,利用三联体密码子推测romt的氨基酸序列,氨基酸的改变方式及产松芪相对活性如表1所示,以对照工程菌bl21(de3)/prsf-vvromt(菌株编号vvromt)的产松芪相对活性为100%,根据imagej软件对tlc检测图的分析结果确定romt突变文库中工程菌产松芪的相对活性;

本发明表达高效产松芪romt突变体的工程菌转化白藜芦醇产松芪的产量测定

测定表达高效产松芪romt突变体的工程菌在50ml培养体系中转化白藜芦醇产松芪的产量。

摇瓶发酵:将对照菌大肠杆菌bl21(de3)/prsf-vvromt以及8株表达高效产松芪romt突变体的工程菌,分别接种至50ml离心管中的5mllb(含50μg/ml卡那霉素)液体培养基,37℃,200rpm摇床中培养12小时,作为种子液,取0.5ml种子液接入到250ml三角瓶中的50mllb(含50μg/ml卡那霉素)培养基,于37℃,200rpm的摇床中发酵培养。然后用分光光度计于600nm波长下检测发酵液的吸光度,待发酵液在600nm波长下吸光度达到0.6时,添加iptg到上述发酵液中至终浓度为100μm。将发酵液置于18℃,200rpm的摇床之中培养8个小时,得到对照菌株bl21(de3)/prsf-vvromt以及8株表达高效产松芪romt突变体工程菌的发酵液,4℃4000g离心15min,去除上清培养基,用50ml改良m9(含50μg/ml卡那霉素、0.1mmiptg、228mg/l白藜芦醇)培养基重悬菌体,在18℃,200rpm摇床中培养12h,即得到对照菌株bl21(de3)/prsf-vvromt以及表达高效产松芪romt工程菌的发酵液。分别取500μl发酵液加500μl无水乙醇,涡旋振荡30s,于离心机中12000rpm离心10min,上清用0.45μm有机滤膜过滤后用lc-ms测定松芪产量。表达高效产松芪romt突变体工程菌(菌株编号romt01-08)在12h内松芪产量及其相比对照菌株bl21(de3)/prsf-vvromt(菌株编号vvromt)的产量提高倍数如表2所示。
