背景技术
:高尿酸血症是一种慢性代谢性疾病,临床表现为血尿酸水平高于正常范围(男性>420μmol/l,女性>360μmol/l)。高尿酸血症患者除因尿酸结晶引发痛风外,还可能发展为肾病、尿路结石,心血管紊乱,脑血管紊乱等。正常生理状态下,人体内尿酸总量约1200mg,尿酸的排泄主要有两种途径,2/3左右通过肾脏以尿液形式排出,1/3以肠道形式排泄的。越来越多的临床研究数据表明,尿酸的肠道途径代谢障碍是导致高尿酸血症的重要因素。成人的肠道中大约有1.5-2.5kg的内容物,由未消化完的食物,消化液,胆汁,人体排出的废物等成分组成,同时,人体肠道内还存在着大约1014个微生物(也称为肠道菌群),约为人体细胞数量的10倍,肠道菌群构成微生态系统,参与人体营养物质消化吸收、合成、免疫调节、能量代谢等生命活动,构成肠道黏膜的保护屏障,因此,肠道菌群构成与生活习惯,饮食和许多疾病密切相关。在成年人个体中,这些肠道微生物的总重量约1271克,相当于肝脏的重量。这些微生物的编码基因总量是人类编码基因数目的50~100倍,被统称为宏基因组。人体每日排出粪便重量的50%以上由细菌及其“尸体”构成。对肠道排泄尿酸的机制进行研究,人们发现肠道内细菌在嘌呤和尿酸的产生和代谢过程中扮演着重要的角色。某些肠道菌自身具有分泌尿酸的能力,同时死亡菌体富含嘌呤,嘌呤被机体吸收后最终代谢产物也是尿酸,然而,许多研究结果也表明,部分肠道菌具有降解尿酸的能力。痛风患者体内肠道菌群失调导致尿酸代谢不平衡,可能是高尿酸症状的重要原因。目前,王海涛等人研究结果显示,通过结肠透析可以显著降低高尿酸血症(王海涛,陈椰,等.中国中西医结合杂志,2007年6月第27卷第6期:492-494);曹永孝,等人通过口服具有吸附作用的蒙脱石散和活性炭,也可以达到降低高尿酸血症的效果(zhaoma,yongxiaocao,et.al.journalofpharmacyandpharmacology2009;61:1499–1504),这些研究结果提示,降低肠道中的游离尿酸含量,可达到治疗高尿酸血症和痛风的目的。然而,以上方式的诸多缺陷,限制了临床上的应用,比如结肠透析操作复杂,需要专业的设备和技术人员才能操作,因此费用非常高;长时间口服蒙脱石散和活性炭,会带来便秘,肠道电解质失衡等后遗症等,因此无法在临床上广泛应用。肠道菌作为人体的长期共生菌,作为降低肠道中游离尿酸含量的媒介具有天然的优势。肠道菌降低尿酸的机制,主要可分为两类,一类是降解尿酸的前体物质,如嘌呤,核苷酸类的物质,降低食源性核苷酸的摄入,从而降低高尿酸血症。如日本明治公司(专利号:cn102747004b)开发的含有降嘌呤或核苷酸的益生菌的酸奶;还有一类,通过降解肠道中的尿酸,增加血中尿酸向肠道中的排泄,从而降低高尿酸血症,如张彦新等人将尿酸氧化酶构建进入乳酸菌,构建具有降尿酸能力的益生菌(张彦新,蒋云生.南京大学学报,2009年4月第45卷:223-224)。然而,血液中尿酸的来源,80%以上来自自身新陈代谢产生,食源性尿酸摄入的量很少,通过降低食源性的尿酸摄入,来试图降血尿酸的效果有限。此外,本专利发明人也曾构建过表达尿酸氧化酶的工程益生菌,试图通过增加益生菌的分解尿酸能力达到降低肠道环境尿酸浓度的目的,但体外模拟效果显示,效果并不理想。一次试验意外发现,当菌体裂解后,较完整菌体降解尿酸的能力显著上升,该试验结果提示专利发明人,表达尿酸氧化酶的工程益生菌降尿酸效果不佳,可能为胞外的尿酸无法有效的透过细胞膜到达胞内与尿酸氧化酶接触。因此,仅表达尿酸氧化酶的工程益生菌,虽然可以降解尿酸,但是效果不佳,不具有显著的应用价值。此后多次研究结果发现,对外源尿酸的摄取能力,可能是决定肠道菌降尿酸能力的关键因子。尿酸透过酶(uricacidpermease)是一种膜蛋白,具有12-14个跨膜区,存在于许多微生物中,具有转运胞外的尿酸以及尿酸相关衍生物(如黄嘌呤,次黄嘌呤,腺嘌呤,鸟嘌呤等)进入细胞内部的功能,是某些微生物摄取尿酸作为氮源代谢的基础。不同微生物来源的尿酸透过酶,分子量有差异,但是跨膜结构和功能区域具有较高的相似度,研究结果显示,如果缺失尿酸透过酶的功能,尿酸的摄入量会显著下降(diallinasandscazzocchio,1989)。在许多益生菌的基因组测序中,有预测为尿酸透过酶的基因,但是本专利发明人在降尿酸测试过程中,均未发现天然益生菌能高效吸收外源尿酸的能力,少数益生菌虽然可以降低外源尿酸,但效果非常弱,不具有实际应用价值(内部数据,未公开)。天然益生菌不具有高效降低尿酸能力的可能原因有:(1)尿酸透过酶基因在天然益生菌中不表达或表达效率低;(2)预测的尿酸透过酶无尿酸透过功能。至今,尚未有研究将尿酸透过酶应用到治疗高尿酸血症和痛风的工程益生菌领域。技术实现要素:为了解决上述问题,本发明的目的是提供一种重组高效表达尿酸透过酶或尿酸透过酶和尿酸氧化酶组合的工程益生菌及其制备方法与应用,所述方法能够方便快捷的制备工程益生菌,所述工程益生菌能够高效组成型表达尿酸透过酶或尿酸透过酶和尿酸氧化酶的组合,较仅表达尿酸氧化酶的降尿酸工程益生菌,其降尿酸能力显著增强,且动物降尿酸的效果试验显示,具有明显的降尿酸效果。作为降低血尿酸和痛风治疗的新手段,具有广阔的应用前景。为了实现上述目的本发明采用如下技术方案:第一方面,提供一种工程益生菌,所述工程益生菌的基因组上整合了完整的尿酸透过酶表达盒,包括强启动子、尿酸透过酶基因和强串联终止子。优选的,所述强启动子,包括但不限于p59,p32,pslpa,hcepromotor等,优选hcepromotor,核苷酸序列如seqidno.20所示,所述强串联终止子,包括但不限于大肠杆菌rrnbt1t2,核苷酸序列如seqidno.21所示。优选的,所述的尿酸透过酶基因来源于原核生物,包括枯草芽孢杆菌pucj,其氨基酸序列如seqidno.11所示、枯草芽孢杆菌puck,其氨基酸序列如seqidno.12所示、嗜铬单胞菌puck,其氨基酸序列如seqidno.13所示、链霉菌puck,其氨基酸序列如seqidno.14所示、拟杆菌pucj,其氨基酸序列如seqidno.15所示、格氏链球菌pbux,其氨基酸序列如seqidno.16所示、副干酪乳杆菌puck,其氨基酸序列如seqidno.17、大肠杆菌ygfu,其氨基酸序列如seqidno.18、大肠杆菌uact,其氨基酸序列如seqidno.19中的任一种。更优选的,所述的尿酸透过酶具有如seqidno.11,seqidno.17,seqidno.18所示的氨基酸序列中的任一种。第二方面,提供一种工程益生菌,所述工程益生菌的基因组上整合了完整的尿酸氧化酶和尿酸透过酶的表达盒,包括强启动子、尿酸氧化酶、尿酸透过酶和强串联终止子。优选的,所述强启动子,包括但不限于p59,p32,pslpa,hcepromotor等,优选hcepromotor,核苷酸序列如seqidno.20所示,所述强串联终止子,包括但不限于大肠杆菌rrnbt1t2,核苷酸序列如seqidno.21所示。优选的,所述的尿酸氧化酶基因来源于原核生物、真核生物或动物,包括枯草芽孢杆菌uox,其氨基酸序列如seqidno.1所示、产朊假丝酵母uox,其氨基酸序列如seqidno.2所示、黄曲霉uox,其氨基酸序列如seqidno.3所示、黑曲霉uox,其氨基酸序列如seqidno.4所示、里氏木霉uox,其氨基酸序列如seqidno.5所示、球形节杆菌uox,其氨基酸序列如seqidno.6所示、微杆菌uox,其氨基酸序列如seqidno.7所示、蛹虫草uox,其氨基酸序列如seqidno.8所示、克鲁维酵母uox,其氨基酸序列如seqidno.9所示、猪uox,其氨基酸序列如seqidno.10所示任一种。更优选的,所述的尿酸氧化酶具有如seqidno.2,seqidno.3,seqidno.5所示的氨基酸序列中的任一种。优选的,所述的尿酸透过酶基因来源于原核生物,包括枯草芽孢杆菌pucj,其氨基酸序列如seqidno.11所示、枯草芽孢杆菌puck,其氨基酸序列如seqidno.12所示、嗜铬单胞菌puck,其氨基酸序列如seqidno.13所示、链霉菌puck,其氨基酸序列如seqidno.14所示、拟杆菌pucj,其氨基酸序列如seqidno.15所示、格氏链球菌pbux,其氨基酸序列如seqidno.16所示、副干酪乳杆菌puck,其氨基酸序列如seqidno.17、大肠杆菌ygfu,其氨基酸序列如seqidno.18、大肠杆菌uact,其氨基酸序列如seqidno.19中的任一种。更优选的,所述的尿酸透过酶具有如seqidno.11,seqidno.17,seqidno.18所示的氨基酸序列中的任一种。优选的,上述第一方面、第二方面所述的工程益生菌,其宿主细胞包括但不限于乳杆菌属(lactobacillus)、芽孢杆菌属(bacillus)、乳球菌属(lactococcus)。更优选的,所述工程益生菌的宿主细胞为干酪乳杆菌(lactobacilluscasei)、枯草芽孢杆菌(bacillussubtilis)、乳酸乳球菌乳酸亚种(lactococcuslactissubsp.lactis)。第三方面,提供上述工程益生菌的制备方法,该方法包括以下步骤:(1)尿酸透过酶基因或尿酸氧化酶和尿酸透过酶基因的克隆获得;(2)尿酸透过酶或尿酸透过酶和尿酸氧化酶重组表达整合载体的构建;(3)将(2)中所获得的重组整合载体电转入相应的宿主菌,筛选阳性转化子;(4)将(3)中阳性转化子通过提高培养温度至42℃,迫使其发生单交换,筛选阳性菌株;(5)将(4)中所获得的阳性单交换菌株,继续传代培养,筛选抗性丢失菌株,通过pcr鉴定筛选阳性整合菌株。本发明的原理:为提高肠道工程益生菌的尿酸降解能力,需增加其外源尿酸摄入能力。本专利发明人筛选了各种增强尿酸透过酶表达的启动子,包括p59,p32,pslpa,hcepromotor等,以及不同物种来源的尿酸透过酶和尿酸氧化酶,以期提升益生菌的尿酸摄取能力,直接有效的降低了肠道内游离尿酸的浓度,从而提升血中尿酸向肠道内排泄的比例。人体每日排泄粪便的50%是肠道菌,摄取了外源游离尿酸的益生菌,被排出体外的同时,也带走了尿酸,同样有助于降低肠道中的游离尿酸浓度。构建了尿酸透过酶的益生菌与原始益生菌相比,可显著的增加外源尿酸的摄入,从而降低胞外尿酸的浓度。第四方面,提供上述工程益生菌在制备预防和/或治疗高尿酸血症、痛风或痛风并发症的产品中的应用。优选的,所述产品为药物、特殊医学用途配方食品、保健食品或食品。第五方面,提供一种能降低血尿酸的口服微生物制剂,其特征在于,所述口服微生物制剂含有一种或一种以上上述工程益生菌。第六方面,提供上述口服微生物制剂在制备预防和/或治疗高尿酸血症、痛风或痛风并发症的产品中的应用。优选的,所述产品为药物、特殊医学用途配方食品、保健食品或食品。优选的,所述口服微生物制剂单独应用或者与其他药物联合应用。本发明与现有技术相比,具有如下优点:本申请将尿酸透过酶,尿酸透过酶和尿酸氧化酶用强启动子元件分别构建入不同的肠道益生菌株,比较其降尿酸的能力,惊奇的发现,构建了高效表达尿酸透过酶的益生菌与原始益生菌相比,可显著的增加外源尿酸的摄入,从而降低胞外尿酸的浓度,并且,同时构建了高效表达尿酸透过酶与尿酸氧化酶的菌种,其降低胞外尿酸的能力更强,显著高于原始益生菌种,也优于只含有尿酸透过酶的工程菌种,体外孵育20h降尿酸能力最高达到96%。本发明专利的降尿酸工程益生菌,较仅表达尿酸氧化酶的降尿酸益生菌,其降尿酸能力显著增强,且在高尿酸血症动物模型中降尿酸的效果试验显示,具有明显的降血尿酸效果,血清尿酸浓度下降最高达到45.4%,尿尿酸浓度下降最高达到73.3%。作为降低血尿酸和痛风治疗的新手段,具有广阔的应用前景。附图说明图1为载体pe
