本发明属于基因及其制备领域,特别涉及一种大腹园蛛梨状腺丝蛋白的基因及其制备方法。
背景技术:
园蛛科蜘蛛可分泌7种蜘蛛丝,分别为主壶腹腺丝、次壶腹腺丝、鞭状腺丝、管状腺丝、梨状腺丝、葡萄状腺丝及聚合腺丝;蛛丝纤维由不同的蛛丝蛋白组成,具有不同的材料学性能和生物学功能。蜘蛛丝拥有优异的机械性能以及力学性能,是许多天然或人工材料无法比拟的。获得蛛丝蛋白基因的全长基因序列是研究蜘蛛丝性能及其机理的基础,蛛丝蛋白序列由相对保守的非重复区nt,ct以及一个很大的重复区组成,蛛丝蛋白往往是很大的蛋白质,相对分子量可达200-350kda。从dna文库中获取蛛丝全长基因耗时耗力,直接pcr得到全长蛛丝基因方便快捷,但由于缺乏相关目的序列片段及长距离pcr结果的不确定性,只有少数全长的蛛丝蛋白基因被克隆出来,严重制约了对蜘蛛丝的进一步研究。
梨状腺丝由梨状腺所分泌,属于固着丝,梨状腺丝其实是干性丝纤维和湿胶的复合体,主要用于蛛丝与固体支持物之间的连接。第一条pysp1全长基因在2017年才被报道出来,其包括21个重复单元,每个重复单元包括234个氨基酸残基,其中由包含大量px和qq小模块。
蜘蛛丝外观光滑纤巧、丝滑闪亮、坚韧轻柔,且具有生物相容性以及较好的记忆性能,作为性能优异的天然蛋白类高分子材料具有极大的潜在应用价值,人工制备的蛛丝蛋白通过不同的方法可以制备成不同的形态,如纤维、膜、微球、胶囊等作为生物材料应用于军事、航天及生物医学领域,且其蛋白成分决定了它的可降解性,符合可持续发展的战略要求。
技术实现要素:
本发明所要解决的技术问题是提供一种大腹园蛛梨状腺丝蛋白的基因及其制备方法,克服了现有技术的大腹园蛛梨状腺丝蛋白基因空白的缺陷,该方法克隆了整个基因包含有完整的nt端,重复区以及完整的ct端,用于编码大腹园蛛梨状腺丝蛋白;而且对环境无污染;制备方法工艺简单,条件温和,易于操作,为在基因工程上大规模生产蛛蛋白奠定了基因基础。
本发明的一种大腹园蛛梨状腺丝蛋白的基因,所述基因序列如seqidno.1所示。
所述基因全长大小为11,931bp,共编码3976个氨基酸,具体序列如seqidno.1所示,蛋白分子量为400.0kda。
所述全长基因中nt端大小为450bp,编码150个氨基酸,具体序列如seqidno.2所示。
n-linker区大小为768bp,共编码256个氨基酸,具体序列如seqidno.3所示。
重复区大小为10,116bp,共编码3372个氨基酸,具体序列如seqidno.4所示。
c-linker区大小为327bp,共编码109个氨基酸,具体序列如seqidno.5所示
ct端大小为267bp,共编码89个氨基酸,具体序列如seqidno.6所示。
本发明一种大腹园蛛梨状腺丝蛋白的基因的制备方法,包括:
(1)根据蜘蛛梨状腺丝的pysp1蛋白基因,在nt端简并引物,在重复区设计2条特异性引物,通过简并pcr扩增得到部分的nt端基因序列;
(2)根据步骤(1)所获得的序列分别设计2条特异性引物及1条锚定引物,通过锚定pcr将nt端补齐;
(3)根据步骤(2)所获得的包含完整nt端序列,在nt端5’末端设计1条正向引物,在ct端3’末端设计1条反向引物,用于扩增全长pysp1基因;
(4)对步骤(3)所得pcr产物进行琼脂糖凝胶切胶回收后,与载体进行平末端连接反应,之后进行连接产物回收;
(5)对步骤(4)的连接产物进行平末端克隆,使用热击转化法,转入dh5α感受态细胞中进行转化,复苏时间1h,将转化后的菌液涂布在lb固体培养基上,过夜培养后,挑取单克隆进行pcr检测;
(6)将步骤(5)中得到的阳性单克隆的质粒进行完整测序,最终得到大腹园蛛梨状腺丝蛋白全长基因。
上述制备方法的优选方式如下:
所述步骤(1)中用于扩增部分nt端的引物:正向简并引物:atgttycaratgagttcaatg;
如seqidno.7所示。
反向重复区特异性引物:ctgttgtgtttgagcggattg。如seqidno.8所示。所述步骤(1)中简并pcr扩增的条件为:95℃预变性5min,95℃变性30s,55℃退火30s,72℃延伸30s,循环30次。
所述步骤(2)中用于补全nt端的特异性引物:
引物1:tttcaatagctgccgcctgc;如seqidno.9所示。
引物2:gtggcaatagcttgggttacgc;如seqidno.10所示。
锚定引物:actcctgtggaaccatcggacgggggg。如seqidno.11所示。
所述步骤(2)中锚定pcr条件为:在第一轮pcr中,使用特异性引物1进行单引物扩增,条件:95℃预变性5min,95℃变性30s,60℃退火30s,72℃延伸30s,循环30次;之后将pcr产物进行琼脂糖凝胶切胶回收,用tdt酶进行加多聚dctp反应,产物回收后进行第二轮pcr;使用特异性引物2以及锚定引物进行扩增,条件同上。
所述步骤(3)中用于扩增全长pysp1基因的引物:
正向引物:atgtcttggaccctggggcttcagtttttattc;如seqidno.12所示
反向引物:ctatccaagtgctgcaagtacgac。如seqidno.13所示。
1对引物分别位于pysp基因的5’末端以及3’末端,以获得完整的pysp1基因。
所述步骤(3)中用于扩增全长pysp1基因的酶为phanta高保真聚合酶。
所述步骤(3)中扩增条件为:98℃预变性1min,98℃变性5s,60℃退火15s,72℃延伸10min,循环30次。
所述步骤(4)中载体为:peasy-bluntzerocloningvector。
所述步骤(4)中平末端连接反应为:16℃反应过夜。
所述步骤(5)中连接产物和dh5α感受态细胞的体积比为1:10;lb固体培养基含有氨苄青霉素amp和卡那霉素kan,氨苄青霉素和卡那霉素在培养基中的浓度均为100μg/ml。
本发明的一种大腹园蛛梨状腺丝蛋白的基因的应用,如作为纤维材料。
大腹园蛛梨状腺丝蛋白的基因的功能为:梨状腺丝蛋白是最近才被发现的一种蛛丝蛋白,它是组成附着盘的主要蛋白。附着盘用于连接网的交界处、附着拖丝到其它介质的表面,而且也被用来捕捉和移动猎物。扫描电子显微镜研究发现:在附着盘中,小直径蛋白纤维被埋在像胶的粘合物中,形成类似于树根形状的网络,而且里面还有大直径拖丝,是一种黏性极强的材料,在蜘蛛生命活动中扮演重要角色。
有益效果
本发明首先通过简并pcr扩增出pysp1的nt端部分序列,然后利用锚定pcr将其补全,之后采用长距离pcr方法扩增得到完整的pysp1基因并进行平末端克隆,得到含有全长pysp1基因的克隆;
本发明克隆了整个基因包含有完整的nt端,重复区以及完整的ct端,用于编码大腹园蛛梨状腺丝蛋白;而且对环境无污染;
首次尝试构建具有天然大腹园蛛梨状腺丝蛋白主要特征模块的迷你型蛛丝蛋白基因,(av.pyspnt-pysprp-pyspct)及pysp的一个完整的重复区域(av.pysprp),但迷你型pysp蛛丝蛋白基因av.pyspnt-pysprp-pyspct在e.colibl21(de3)菌株中的表达量极低,单纯pysp完整的重复区(av.pysprp)基因在e.colibl21(de3)菌株中可以正常表达,得到的大腹园蛛梨状腺丝蛋白,经过静电纺丝可将该蛋白纺成纳米纤维膜;
本发明制备方法工艺简单,条件温和,易于操作,为在基因工程上大规模生产蛛蛋白奠定了基因基础。

具体实施方式
下面结合具体实施例,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。此外应理解,在阅读了本发明讲授的内容之后,本领域技术人员可以对本发明作各种改动或修改,这些等价形式同样落于本申请所附权利要求书所限定的范围。
实施例1
(1)根据其它蜘蛛的pysp1的nt端的保守序列,分别在nt端和ct端各设计1条简并引物:nt端简并引物:atgttycaratgagttcaatg,并在重复区设计1条特异性反向引物,特异性反向引物:ctgttgtgtttgagcggattg;然后进行pcr,条件为95℃预变性5min,95℃变性30s,55℃退火30s,72℃延伸30s,循环30次。
(2)根据所获得的序列分别在nt端和ct端各设计2条特异性引物以及1条锚定引物用于补全nt端和ct端。
nt端特异性引物1:tttcaatagctgccgcctgc;
nt端特异性引物2:gtggcaatagcttgggttacgc;
:锚定引物:actcctgtggaaccatcggacgggggg;
锚定pcr条件为:在第一轮pcr中,使用特异性引物1进行单引物扩增,条件:95℃预变性5min,95℃变性30s,60℃退火30s,72℃延伸30s,循环30次。之后将pcr产物进行琼脂糖凝胶切胶回收,用tdt酶进行加c反应,产物回收后进行第二轮pcr。使用特异性引物2以及锚定引物进行扩增,条件同上。
(3)之后设计1对特异性引物用于扩增全长pysp基因片段,
正向引物:atgtcttggaccctggggcttcagtttttattc;
反向引物:ctatccaagtgctgcaagtacgac;
pcr条件为98℃预变性1min,98℃变性5s,60℃退火15s,72℃延伸10min,循环30次。之后琼脂糖凝胶切胶回收。(phanta高保真聚合酶添加在pcr反应液中)
(4)将回收产物与载体peasy-bluntzerocloningvector进行平末端连接反应,条件为:16℃反应过夜。
(5)之后转入dh5α感受态细胞,体积比为1:10,复苏时间为1h。之后将菌液涂布在氨苄青霉素和卡那霉素的固体培养基上,浓度均为100μg/ml,37℃培养12h。然后对菌落进行筛选,测序。
使用signalp4.1、expasytools、psipred等在线软件以及geneious、dnastar软件对该基因进行分析,signalp4.1用于寻找基因的信号肽,expasytools用来对pysp1的疏水性进行分析,psipred对pysp1的二级结构进行预测,geneious用来进行多序列比对,dnastar用来对pysp1基因的碱基组成和氨基酸组成进行分析。

图1为pysp1蛋白的结构域以及序列图谱,从图1中可以看出,整个pysp1基因划分为五个区域,nt端,n-linker,重复区,c-linker以及ct端。其中重复区内由16个在序列上极其相似的重复单元组成,并占据了整个蛋白序列的85%以上。其中前15个重复单元大小为213个氨基酸,第16个重复单元为177个氨基酸。在图1中可以发现蛋白的信号肽存在于nt端中(粗体标记),并且存在2个cys残基。
本发明大腹园蛛pysp1蛋白的氨基酸序列,同时如seqidno.16所示。
expasytools分析结果:图2为整个pysp1蛋白的疏水性分析图以及nt端,repeatunit8(r8),ct端的疏水性图。从图2中可以看到,疏水性区域以及亲水性区域在整个蛋白序列中反复变化,整个蛋白分子表现为疏水性。ct端的平均疏水性值大于nt端,但nt端的疏水性峰值大于ct端。
psipred分析结果:图3为pysp蛋白二级结构预测图,从图3可以看出,在nt端中含有5个α螺旋结构,第一个cys残基处于第1个和第2个螺旋之间,第二个cys残基处于第4个螺旋中。在图3中可以发现ct端中只含有4个螺旋结构。图3中重复单元内有8个螺旋结构。
geneious分析结果:图4为pysp蛋白的对比图。从图4a,图4b,图4c中可以看出不同物种之间的pysp的nt端,重复单元以及ct端都有较高的相似度。
实施例2
1.根据pysp重复区序列设计引物
primer1:tattctgcagcatcaactgcc;如seqidno.14所示
primer2:agcgtttgagctttgttgagattg;如seqidno.15所示。
扩增条件为:95℃预变性5,95℃变性30s,57℃退火30s,72℃延伸1min,循环30次。
扩增全长pysp重复区基因片段得到pcr产物。将上述实验制备所得到的插入片段和pehs载体进行连接,在冰盒上解冻以下试剂,并按下列顺序在冰上加样,连接体系如下:

置于pcr管中,混匀,于室温下连接6h。连接产物热激转化入e.colidh5α感受态细胞。
抽取质粒转入bl21菌株,进行蛋白诱导表达:
(1)蛋白诱导表达5h后,使用低温高速离心机4000rpm,15min离心收集菌体,并用20mm的tris-hcl(ph8.0)以30ml/l菌液的比例溶解;
(2)将溶解得到的混合液使用机械细胞破碎机进行细胞破碎,菌体破碎后12000rpm,4℃,10min收集包涵体;
(3)将离心得到的包涵体全部溶于8m的尿素中,按照70ml/l的比例,将混合液套上保鲜膜于电磁加热搅拌器上孵育30~60min;
(4)孵育后的混合液,12000rpm,25℃条件下离心30min;
(5)离心后,收集上清液,取出部分使用sds-page检测蛋白表达的裂解的情况;
(6)剪一段合适长度的透析袋,用去离子水将透析袋内部冲洗干净,待用;
(7)将透析袋的一端用透析夹夹紧,倒入蛋白液之前可先向其中加入少量去离子水以检查密封性;
(8)将处理过的含尿素的蛋白溶液缓慢倒入透析袋中,最后,将透析袋的另一端同样用透析夹夹紧;
(9)将透析袋放入装有超纯水的大的容器中,放入转子并置于电磁搅拌器上搅拌,约2h更换一次容器中的透析液,整个透析过程进行约48h。
透析结束后,将透析袋内的蛋白液全部倒入到500ml冻干杯内,用冷冻干燥机冻干,至杯中出现粉状的蛋白。取蛋白粉进行静电纺:
(1)取0.24g提取的蛋白加入3ml的六氟异丙醇(hfip)制成8%的纺丝液;
(2)另取一干净的针管,将混合均匀的纺丝液倒入针管中,并将处理好的针头插好,固定在注射棒上;
(3)准备一张约17cm*17cm的正方形纸板,用铝箔纸包好,使一面光滑完整;
(4)设置注射棒的参数,5cc,0.80ml/h,4.00ml;
(5)待针头处有液滴流出时,接通高压电源(16kv),开始静电纺丝,可以观察到,在电场力的作用下,枕头出的液滴由球型变为圆锥形(taylor锥);
(6)静电纺丝结束后,关闭电源,将用过的针头取下重新置于六氟异丙醇中循环使用,纺得的纳米纤维膜放于干燥处储存待用。
将纺得的膜于扫描电子显微镜(sem)下拍照,参见附图5。
由此可以说明,利用纯pysp蛋白进行静电纺丝制备纳米纤维膜是可行的,为蛛丝纤维应用提供了基础材料。
