,涉及一种转C4光合关键基因提高C3植物光合作用的方法,特别涉及利用多基因转化系统,改造获得一个含5个光合基因的载体,利用不同的启动子驱动这些基因,创制可以显著提高光合作用效率的水稻材料的方法。
背景技术
:随着工业化、城镇化的发展以及人口的增加,我国粮食需求将呈刚性增长,而耕地减少、水资源短缺、气候变化等对粮食生产的约束日益突出,我国粮食供需将长期处于“紧平衡”状态,保障粮食安全面临严峻挑战。农作物的株型育种和杂种优势利用为我国粮食作物产量的提高,保障国民经济的快速发展做出了巨大贡献,但粮食产量的进一步提高难度越来越大。在当前保障粮食安全作为我国长期国策的背景下,如何实现粮食作物产量水平的持续提升是横亘在我国社会经济、农业科技前进道路上的一道难题。作物收获指数是衡量作物产量的主要指标之一,目前水稻、小麦的收获指数都接近0.5,这意味着这两种作物的粮食产量占全部生物量的近50%,通过传统育种实现粮食产量的大跨度提高已不可能。幸运的是,世界上还有另一些植物(C4植物,如玉米、高粱和甘蔗等),它们的光合效率要比C3植物高出50%。利用转基因的手段,理论上转化可以显著地提高C4光合作用。尽管C4途径的酶及转运蛋白在C3植物中的过表达研究取得了很大进展,在C3植物叶片的特定区域高水平表达C4途径酶的技术也很成熟,由于光合系统是一个复杂、分工明细的系统,转单个基因并不能明显提高C3植物的光合效率。一些研究表明,高水平表达C4基因对C3植物光合效率提高和生理性状改良的作用有限,有时甚至是不利的。Agarie等测定了PEPC活性高于对照50倍的转基因水稻株系,结果是其光合速率均略低于对照。Ku等发现部分转PEPC水稻植株显示了不良的农艺性状,如株高矮化、叶片变窄、不育等。而试图将PEPC、NADP-ME、PPDK、NADP-MDH全部转入水稻中,以期构成一个完整C4循环的实验也以失败告终,该转多基因水稻同样表现生长受抑制,说明体内所建立的是一个无效的循环,对C4特有的CO2浓缩机制没有贡献。技术实现要素:针对现有技术的不足,本发明采用了不同的启动子策略,将5个C4光合关键基因导入水稻。第一,本发明要求保护一种增强植物光合作用和/或提高植物产量和/或提高植物生物量的方法。本发明所提供的增强植物光合作用和/或提高植物产量和/或提高植物生物量的方法(或称为培育光合作用增强和/或产量提高和/或生物量提高的转基因植物方法),具体可包括如下步骤:向受体植物中导入5个C4光合关键基因,得到转基因植物;所述转基因植物与所述受体植物相比,光合作用增强和/或产量提高和/或生物量提高。其中,所述5个C4光合关键基因为磷酸丙酮酸双激酶(ZmPPDK1)的编码基因、PEP羧化酶(ZmPEPC1)的编码基因、苹果酸脱氢酶(ZmMDH6)的编码基因、NADP-苹果酸酶(ZmME2)的编码基因,以及碳酸酐酶(ZmCA1)的编码基因。在所述转基因植物中,所述磷酸丙酮酸双激酶(ZmPPDK1)的编码基因由所述磷酸丙酮酸双激酶(ZmPPDK1)的编码基因的内源启动子驱动转录;所述PEP羧化酶(ZmPEPC1)的编码基因由所述PEP羧化酶(ZmPEPC1)的编码基因的内源启动子驱动转录;所述苹果酸脱氢酶(ZmMDH6)的编码基因由35CaMVS启动子驱动转录;所述NADP-苹果酸酶(ZmME2)的编码基因由Ubiqutin启动子驱动转录;所述碳酸酐酶(ZmCA1)的编码基因由OsActin启动子驱动转录。其中,所述5个C4光合关键基因是通过重组载体的形式导入所述受体植物中的。所述重组载体为下文任一所述的重组载体。在上述方法中,将所述重组载体导入所述受体植物具体可为:通过使用Ti质粒、Ri质粒、植物病毒载体、直接DNA转化、显微注射、电导、农杆菌介导等常规生物学方法转化植物细胞或组织,并将转化的植物组织培育成植株。第二,本发明要求保护一种重组载体或含有所述重组载体的重组菌。所述重组载体为含有DNA片段1、DNA片段2、DNA片段3、DNA片段4和DNA片段5的环形质粒。所述DNA片段1自上游到下游依次含有磷酸丙酮酸双激酶(ZmPPDK1)的编码基因的内源启动子、磷酸丙酮酸双激酶(ZmPPDK1)的编码基因。所述DNA片段2自上游到下游依次含有PEP羧化酶(ZmPEPC1)的编码基因的内源启动子、PEP羧化酶(ZmPEPC1)的编码基因。所述DNA片段3自上游到下游依次含有35CaMVS启动子、苹果酸脱氢酶(ZmMDH6)的编码基因。所述DNA片段4自上游到下游依次含有Ubiqutin启动子、NADP-苹果酸酶(ZmME2)的编码基因。所述DNA片段5自上游到下游依次含有OsActin启动子、碳酸酐酶(ZmCA1)的编码基因。进一步地,所述重组载体为将所述DNA片段1、所述DNA片段2、所述DNA片段3、所述DNA片段4和所述DNA片段5顺次连接后插入到pCambia1300载体的酶切位点HindIII和SmaI之间后得到的重组质粒。前文所述的磷酸丙酮酸双激酶(ZmPPDK1)可为如下任一所示蛋白质:(A1)氨基酸序列为SEQIDNo.11的蛋白质;(A2)将SEQIDNo.11所示的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加且具有相同功能的蛋白质;(A3)与(A1)-(A2)中任一所限定的氨基酸序列具有99%以上、95%以上、90%以上、85%以上或者80%以上同源性且具有相同功能的蛋白质;(A4)在(A1)-(A3)中任一所限定的蛋白质的N端和/或C端连接标签后得到的融合蛋白。前文所述的PEP羧化酶(ZmPEPC1)可为如下任一所示蛋白质:(B1)氨基酸序列为SEQIDNo.12的蛋白质;(B2)将SEQIDNo.12所示的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加且具有相同功能的蛋白质;(B3)与(B1)-(B2)中任一所限定的氨基酸序列具有99%以上、95%以上、90%以上、85%以上或者80%以上同源性且具有相同功能的蛋白质;(B4)在(B1)-(B3)中任一所限定的蛋白质的N端和/或C端连接标签后得到的融合蛋白。前文所述的苹果酸脱氢酶(ZmMDH6)可为如下任一所示蛋白质:(C1)氨基酸序列为SEQIDNo.13的蛋白质;(C2)将SEQIDNo.13所示的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加且具有相同功能的蛋白质;(C3)与(C1)-(C2)中任一所限定的氨基酸序列具有99%以上、95%以上、90%以上、85%以上或者80%以上同源性且具有相同功能的蛋白质;(C4)在(C1)-(C3)中任一所限定的蛋白质的N端和/或C端连接标签后得到的融合蛋白。前文所述的NADP-苹果酸酶(ZmME2)可为如下任一所示蛋白质:(D1)氨基酸序列为SEQIDNo.14的蛋白质;(D2)将SEQIDNo.14所示的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加且具有相同功能的蛋白质;(D3)与(D1)-(D2)中任一所限定的氨基酸序列具有99%以上、95%以上、90%以上、85%以上或者80%以上同源性且具有相同功能的蛋白质;(D4)在(D1)-(D3)中任一所限定的蛋白质的N端和/或C端连接标签后得到的融合蛋白。前文所述的碳酸酐酶(ZmCA1)可为如下任一所示蛋白质:(E1)氨基酸序列为SEQIDNo.15的蛋白质;(E2)将SEQIDNo.15所示的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加且具有相同功能的蛋白质;(E3)与(E1)-(E2)中任一所限定的氨基酸序列具有99%以上、95%以上、90%以上、85%以上或者80%以上同源性且具有相同功能的蛋白质;(E4)在(E1)-(E3)中任一所限定的蛋白质的N端和/或C端连接标签后得到的融合蛋白。前文所述的磷酸丙酮酸双激酶(ZmPPDK1)的编码基因可为如下任一所述的DNA分子:(a1)SEQIDNo.2所示的DNA分子;(a2)在严格条件下与(a1)限定的DNA分子杂交且编码所述磷酸丙酮酸双激酶(ZmPPDK1)的DNA分子;(a3)与(a1)-(a2)中任一限定的DNA序列具有99%以上、95%以上、90%以上、85%以上或者80%以上同源性且编码所述磷酸丙酮酸双激酶(ZmPPDK1)的DNA分子。前文所述的PEP羧化酶(ZmPEPC1)的编码基因可为如下任一所述的DNA分子:(b1)SEQIDNo.4所示的DNA分子;(b2)在严格条件下与(b1)限定的DNA分子杂交且编码所述PEP羧化酶(ZmPEPC1)的DNA分子;(b3)与(b1)-(b2)中任一限定的DNA序列具有99%以上、95%以上、90%以上、85%以上或者80%以上同源性且编码所述PEP羧化酶(ZmPEPC1)的DNA分子。前文所述的苹果酸脱氢酶(ZmMDH6)的编码基因可为如下任一所述的DNA分子:(c1)SEQIDNo.6所示的DNA分子;(c2)在严格条件下与(c1)限定的DNA分子杂交且编码所述苹果酸脱氢酶(ZmMDH6)的DNA分子;(c3)与(c1)-(c2)中任一限定的DNA序列具有99%以上、95%以上、90%以上、85%以上或者80%以上同源性且编码所述苹果酸脱氢酶(ZmMDH6)的DNA分子。前文所述的NADP-苹果酸酶(ZmME2)的编码基因可为如下任一所述的DNA分子:(d1)SEQIDNo.8所示的DNA分子;(d2)在严格条件下与(d1)限定的DNA分子杂交且编码所述NADP-苹果酸酶(ZmME2)的DNA分子;(d3)与(d1)-(d2)中任一限定的DNA序列具有99%以上、95%以上、90%以上、85%以上或者80%以上同源性且编码所述NADP-苹果酸酶(ZmME2)的DNA分子。前文所述的碳酸酐酶(ZmCA1)的编码基因可为如下任一所述的DNA分子:(e1)SEQIDNo.10所示的DNA分子;(e2)在严格条件下与(e1)限定的DNA分子杂交且编码所述碳酸酐酶(ZmCA1)的DNA分子;(e3)与(e1)-(e2)中任一限定的DNA序列具有99%以上、95%以上、90%以上、85%以上或者80%以上同源性且编码所述碳酸酐酶(ZmCA1)的DNA分子。上述严格条件可为用6×SSC,0.5%SDS的溶液,在65℃下杂交,然后用2×SSC,0.1%SDS和1×SSC,0.1%SDS各洗膜一次。前文所述的磷酸丙酮酸双激酶(ZmPPDK1)的编码基因的内源启动子的核苷酸序列如SEQIDNo.1所示。前文所述的PEP羧化酶(ZmPEPC1)的编码基因的内源启动子的核苷酸序列如SEQIDNo.3所示。前文所述的35CaMVS启动子的核苷酸序列如SEQIDNo.5所示。前文所述的Ubiqutin启动子的核苷酸序列如SEQIDNo.7所示。前文所述的OsActin启动子的核苷酸序列如SEQIDNo.9所示。更进一步地,所述DNA片段1的核苷酸序列为将SEQIDNo.1和SEQIDNo.2直接顺次连接后得到的序列。所述DNA片段2的核苷酸序列为将SEQIDNo.3和SEQIDNo.4直接顺次连接后得到的序列。所述DNA片段3的核苷酸序列为将SEQIDNo.5和SEQIDNo.6直接顺次连接后得到的序列。所述DNA片段4的核苷酸序列为将SEQIDNo.7和SEQIDNo.8直接顺次连接后得到的序列。所述DNA片段5的核苷酸序列为将SEQIDNo.9和SEQIDNo.10直接顺次连接后得到的序列。在本发明的一个实施例中,所述重组载体具体为将SEQIDNo.1-10所示的10个序列直接顺次连接后插入到pCambia1300载体的“GGCCAGTGCCAAGCTT”和“CCCGGGCTGCAAGTTCAA”处后得到的重组质粒。第三,本发明要求保护所述重组载体或重组菌在增强植物光合作用和/或提高植物产量和/或提高植物生物量中的应用。在本发明中,所述植物为C3植物。具体的,所述C3植物可为水稻;更加具体的,如水稻品种日本晴(OryzasativaL.ssp.Japonica)。在本发明中,所述增强植物光合作用(或所述光合作用增强)具体可体现为提高植物光合速率。所述提高植物产量(或所述产量提高)具体可体现为提高植物种子千粒重。本发明利用不同启动子驱动5个光合途径的关键蛋白的编码基因,5个基因来源于玉米(Zeamays.L),分别为磷酸丙酮酸双激酶(ZmPPDK1)、PEP羧化酶(ZmPEPC1)、苹果酸脱氢酶(ZmMDH6)、NADP-苹果酸酶(ZmME2)、碳酸酐酶(ZmCA1)。不同启动子分别为35CaMVS、Ubiqutin、OsActin、ZmPPDK1启动子、ZmPEPC1启动子。将这个多基因载体命名为MGTV(Multi-gene-transformation-vector)载体。将MGTV载体导入C3植物水稻中,创制出具有光合效率显著提高的材料,大辐提高植物的生物量,且没有附带其它不利性状。本发明对于培育C4水稻材料具有重大生产应用价值。附图说明图1为MGTV载体的构建流程图。图2为空载对照和转多基因阳性株系的T1代植株表型。pCambia1300为空载对照植株;MGTV-OV4为转多基因阳性株系OV4。具体实施方式下述实施例中所使用的实验方法如无特殊说明,均为常规方法。下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。下述实施例中的定量试验,均设置三次重复实验,结果取平均值。水稻品种日本晴(OryzasativaL.ssp.Japonica),又称野生型,用WT表示:中国水稻杂交育种中心(中国,长沙)。pCambia1300载体、pCambia1302载体、pCambia1390载体:pCAMBIA公司。农杆菌AGL1:中国质粒载体菌株细胞株基因保藏中心。实施例1、MGTV载体构建及转基因水稻的获得与功能鉴定一、5个光合元件的克隆及多基因载体构建C4植物玉米比C3植物水稻有着较高的光合效率、生物学产量等,主要原因是C4植物具有C4光合循环途径,本发明从玉米中克隆了这些光合途径关键基因:磷酸丙酮酸双激酶(ZmPPDK1)、PEP羧化酶(ZmPEPC1)、苹果酸脱氢酶(ZmMDH6)、NADP-苹果酸酶(ZmME2)、碳酸酐酶(ZmCA1)。利用不同启动子驱动,其中ZmPPDK1启动子驱动ZmPPDK1,ZmPEPC1启动子驱动ZmPEPC1,35CaMVS驱动ZmMDH6,Ubiqutin启动子驱动ZmME2,OsACTIN启动子驱动ZmCA1。磷酸丙酮酸双激酶(ZmPPDK1)的编码基因(CDS)序列如SEQIDNo.2所示,编码SEQIDNo.11所示蛋白质。PEP羧化酶(ZmPEPC1)的编码基因(CDS)序列如SEQIDNo.4所示,编码SEQIDNo.12所示蛋白质。苹果酸脱氢酶(ZmMDH6)的编码基因(CDS)序列如SEQIDNo.6所示,编码SEQIDNo.13所示蛋白质。NADP-苹果酸酶(ZmME2)的编码基因(CDS)序列如SEQIDNo.8所示,编码SEQIDNo.14所示蛋白质。碳酸酐酶(ZmCA1)的编码基因(CDS)序列如SEQIDNo.10所示,编码SEQIDNo.15所示蛋白质。ZmPPDK1启动子的核苷酸序列如SEQIDNo.1所示。ZmPEPC1启动子的核苷酸序列如SEQIDNo.3所示。35CaMVS启动子的核苷酸序列如SEQIDNo.5所示。Ubiqutin启动子的核苷酸序列如SEQIDNo.7所示。OsActin启动子的核苷酸序列如SEQIDNo.9所示。1、首先分别扩增5个光合基因元件的编码框(CDS)和启动子序列。采用TOYOBOKOD高保真酶扩增,按照说明书进行操作,反应系统如表1所示。表1扩增5个光合基因元件的编码框(CDS)和启动子序列的反应系统对分子量正确的扩增条带切胶回收,-20℃保存产物。2、拼接分别获得5个完整的元件序列。因为用于扩增每组元件CDS的F端引物,加有和启动子重叠的15-18bp序列,因而可以利用融合PCR把每组元件的启动子和CDS拼接为一条PCR产物。同时,为了下一步把5个元件重组到目的载体上,这一轮PCR扩增需要在各自的F端引物增加重叠序列,扩增体系如表2所示。表2拼接分别获得5个完整的元件序列的扩增体系使用Clontech的高效重组酶In-FusionHDCloningKit(货号639648),利用每个元件之间的15-18bp重复序列、元件1至元件5和pCambia1300载体之间的重复序列,按照表3中所示的对应比例进行重组反应、转化、鉴定、测序,最终完成载体的构建。表3元件1至元件5和pCambia1300载体的反应比例元件编号片段大小所需量体积元件14844bp216ng1.5μl元件24913bp219ng1.5μl元件31837bp82ng0.5μl元件43982bp177ng1μl元件52784bp124ng1μlpCambia1300载体8958bp200ng2μl补2.5μl重组酶至10μl以上所涉及的各引物序列具体如表4所示。表4载体构建所用的引物通过对在筛选培养基上长出的900多个克隆进行PCR鉴定,获得6个重组完全正确的克隆。对该5个克隆进行测序验证,发现构建载体和预期序列一致,并无碱基突变,可以用于下一步转化水稻。将这个多基因载体命名为MGTV(Multi-gene-transformation-vector)载体,见图1。MGTV载体的结构描述:将SEQIDNo.1-10所示的10个序列分别拼接为5个元件,然后利用其中间的重合序列,在重组酶作用下把该5个元件同时重组到pCambia1300载体的“GGCCAGTGCCAAGCTT”和“CCCGGGCTGCAAGTTCAA”处(位于酶切位点HindIII和SmaI之间)后得到的重组质粒。另外,本发明还构建了如下5个用于转单基因的重组载体:(1)pCambia1300-PPDK1载体:将SEQIDNo.1和SEQIDNo.2所示的2个序列直接顺次连接后插入到pCambia1300载体的“GGCCAGTGCCAAGCTT”和“CCCGGGCTGCAAGTTCAA”处后得到的重组质粒。(2)pCambia1300-PEPC1载体:将SEQIDNo.3和SEQIDNo.4所示的2个序列直接顺次连接后插入到pCambia1300载体的“GGCCAGTGCCAAGCTT”和“CCCGGGCTGCAAGTTCAA”处后得到的重组质粒。(3)pCambia1300-MDH6载体:将SEQIDNo.5和SEQIDNo.6所示的2个序列直接顺次连接后插入到pCambia1300载体的“GGCCAGTGCCAAGCTT”和“CCCGGGCTGCAAGTTCAA”处后得到的重组质粒。(4)pCambia1300-ME2载体:将SEQIDNo.7和SEQIDNo.8所示的2个序列直接顺次连接后插入到pCambia1300载体的“GGCCAGTGCCAAGCTT”和“CCCGGGCTGCAAGTTCAA”处后得到的重组质粒。(5)pCambia1300-CA1载体:将SEQIDNo.9和SEQIDNo.10所示的2个序列直接顺次连接后插入到pCambia1300载体的“GGCCAGTGCCAAGCTT”和“CCCGGGCTGCAAGTTCAA”处后得到的重组质粒。二、转基因材料的创制及鉴定1、将步骤一构建的重组质粒MGTV导入农杆菌AGL1,得到重组农杆菌。2、取步骤1得到的重组农杆菌,重悬于液体共培养培养基(YEP液体培养基+100mg/L乙酰丁香酮,pH5.2),得到OD600nm=0.14的菌液。3、取日本晴的胚性愈伤组织,用步骤2得到的菌液浸泡30min,然后经共培养、筛选、生根、壮苗,得到T0代植株。4、T0代植株自交获得T1代种子,T1代种子长成的植株即为T1代植株。5、用pCambia1300载体进行上述转化,得到转空载体植株。6、用pCambia1300-PPDK1、pCambia1300-PEPC1、pCambia1300-MDH6、pCambia1300-ME2和pCambia1300-CA1载体进行上述转化,得到5种转单基因植株。用表2中对应的引物序列扩增再生植株DNA,电泳确定阳性株系。从鉴定阳性的转多基因株系中随机选择4个,分别编号为OV-1、OV-2、OV-3和OV-4。三、光合参数测定取生长状态一致的日本晴、步骤二得到的4个转基因阳性系(OV-1、OV-2、OV-3和OV-4)、空载对照、转单个基因的T1代植株,在盛花期,利用LI-COR6400进行光合参数的测定,并统计表5中所示的其他农艺性状。结果表明,与空载对照、野生对照、转单个基因(PPDK1、PEPC1、MDH6、ME2、CA1)相比,4个阳性株系(OV-1、OV-2、OV-3和OV-4)均不同程度地大辐度提高光合效率,进而提高千粒重,且没有呈现不利的农艺性状表型。见图2和表5。表5转基因与空对照的各个农艺性状比较注:*表明与空对照相比,差异显著(P<0.05)。<110>中国农业科学院生物技术研究所<120>转C4光合关键基因提高C3植物光合作用的方法<130>GNCLN180019<160>15<170>PatentInversion3.5<210>1<211>2000<212>DNA<213>玉米(ZeamaysL.)<400>1gagattttggggttatggtcgcaagggttatgaatgaagacttaccaggtaaatgggttc60agaatattttgaaagtccgagaggatgatccatgctttgctttaatcaaggttaagtata120atactaaaggtaaattttgctctcaattttggaagggaatactatatgcaaggaacttgg180taaatggggattatcaacaacgattaacaatgagaaaatgtactattttgggaggaggta240tgggttttggacatgccactaatattagaattcccattttttgttgtttgtcaaaataat300cattgtttggtcagtggttgttaggaaggaggtggatagaaagttaaatttagattttcc360ctggggtggaggacatgaaagagtgggaaaggttgctggacattttggaaggagtgataa420ttactaaacgagaatatatgttatcttcgtcgttagagaaatctagacagtatacaacaa480gatccacgtactacaggtaaacttttaggggtattgtgaacaagaggatgagtaaactct540aaaagaacaaagctccaatgaaaatttaggtttttatgtggttagtcatagggcaagttg600caaacaggtgttgatctaaaaaggaagtagtagggaaatgtgaagtgtctttgcgaggaa660ttggaaaatgaagatcacattttctttgggtgcatcatgggaagaaccatttgggactct720tttaaggaggcctaagaatgccataaagtttgcaagatctttttgaagagtgtctaccta780taaacaatagtaaatatcatgtcaaaattttcatcttcgccattattctttaggagaatt840tagaatgttccgaataaaatatggatagaaaagaagttcccaaagtcatccaattttcta900caaaatcttcaactttaagattgagagtgggtgttgtaaagttcttggaagatgagttga960accccatggaggcgttggctaaagtactgaaagcaatctaaagacatggaggtggaaggc1020ctgacgtagatagagaagatgctcttagctttcattgtctttcttttgtagtcatctgat1080ttacctctctcgtttatacaactggttttttaaacactccttaacttttcaaattgtctc1140tttctttaccctagac
