水稻类NAP1编码基因DS8及其用途的制作方法

本发明涉及植物基因工程领域。主要涉及水稻类nap1蛋白基因ds8的核苷酸序列,该核苷酸序列编码的蛋白质及其功能衍生物,以及含有该核苷酸的载体。另外,本发明还涉及该基因对水稻叶片表皮发育和干旱胁迫下气孔闭合的调控,为水稻耐旱性的调控机理研究提供基础。



背景技术:

微丝是细胞骨架的主要成分之一,在植物表皮细胞形态发育过程中起着十分重要的作用。scar/wave-arp2/3复合体参与微丝成核过程,其中scar/wave复合体有5个在进化中十分保守的亚基组成,分别是abi,abi、brick1/hspc300、nap1/nap125、pir/sra1和scar[1,2]。arp2/3复合体则由七个亚基组分,分别是arp2、arp3、arpc1、arpc2、arpc3、arpc4和arpc5,scar/wave复合体通过协助arp2/3参与微丝的成核[3-4]。体内和体外实验都证明scar/wave复合体能促进arp2/3复合体介导的微丝成核速率增长[5-7]。植物scar/wave复合体亚基的突变往往会造成微丝形态异常,进而产生表皮发育异常,主要表现在异常的毛状体(拟南芥中)或乳突(水稻中)发育。另外,也有报道影响表皮扁平细胞的发育[8-13]。植物的叶片表皮对于植物的水分调控起着十分重要的作用。水稻的表皮细胞壁外覆盖有一层疏水的角质层,内部则镶嵌有二氧化硅,两者都具有防止水稻叶片水分过度蒸发的功能[14-22]。此外,叶片上的气孔结构作为水分和气体交换的场所,在水分调节上扮演着十分重要的角色,表皮的水分丧失90%是通过气孔进行的,气孔的密度、气孔开度以及对外界环境的反应速度都直接影响植物的耐旱性[23]。目前部分研究发现微丝参与气孔闭合,在气孔的闭合过程中,原本张开状态的气孔保卫细胞中的放射状的微丝解聚呈现随机分布形态,之后再次聚合形成闭合状态的气孔中纵向分布的形态[24-26]。已证明这一过程发生于脱落酸(aba)和过氧化氢(h2o2)调控气孔闭合通路的下游,微丝成核或者解聚的受限制都会造成气孔闭合的异常或者对aba和h2o2的反应迟缓[27-29]

目前植物中对scar/wave-arp2/3复合体的研究大部分集中于叶片表皮细胞的形成和发育方面,缺少关注于其在植物抵御环境胁迫上的作用。

技术实现要素:

本发明要解决的技术问题是提供一种与水稻表皮发育和气孔闭合相关的蛋白质及其编码基因,以及该基因的用途。

为了解决上述技术问题,本发明提供一种水稻类nap1蛋白基因ds8编码的蛋白质,该蛋白质具有seqidno:3所示的氨基酸序列。

作为本发明的水稻类nap1蛋白基因ds8编码的蛋白质的改进:该蛋白质为在seqidno:3所示的氨基酸序列中通过缺失、替换或者插入一个或多个氨基酸而获得的功能类似物。

本发明还提供了编码上述蛋白质的基因,该基因具有seqidno:1、seqidno:2所示的序列。

作为本发明的基因的改进:该基因为在seqidno:1、seqidno:2所示的核苷酸序列中通过缺失、替换或者插入一个或多个核苷酸而生成的等位基因或衍生物。

本发明还同时提供了一种载体,含有上述基因。

本发明还同时提供了一种宿主细胞,该宿主细胞含有上述载体;:该细胞为大肠杆菌细胞、农杆菌细胞或植物细胞。

本发明还同时提供了上述ds8基因的用途:提高水稻耐旱性。

作为本发明的ds8基因的用途的改进:利用具有seqidno:1和2所示的核苷酸序列的基因转化水稻愈伤,再将转化后的水稻愈伤培育成植株。

本发明利用ds8突变体和正常籼稻品种93-11构建定位群体,利用图位克隆技术首先克隆了ds8基因,该基因编码水稻类nap1蛋白,影响微丝形成并影响水稻的耐旱性。本发明还进行了改基因功能的验证。

实现本发明的具体技术步骤如下:

一、水稻耐旱性相关突变体ds8的分离和遗传分析:

本发明的水稻耐旱性相关突变体ds8来自粳稻品种日本晴ems(ethylmethylsulfonate)诱变产生的突变(图1)。通过ds8突变体与野生型籼稻品种的正、反交实验进行遗传分析,实验结果证明该突变体受隐性单基因控制。

二、ds8基因的图位克隆:

(1)ds8基因的初步定位:

为了分离ds8基因,本发明首先利用ds8和野生型籼稻品种93-11(indica)做杂交构建f2定位群体,再通过图位克隆的方法,利用sts、ssr等分子标记对ds8位点进行初步定位,将其初步定位在第8染色体上rm1345和rm3120两标记之间(图2)。

(2)ds8基因的精细定位与候选基因预测:

通过对初定位区间的bac序列进行分析,进一步发展多态性标记进行精细定位,最终将ds8精确定位于bac克隆ap004632上e1与e2标记之间,53.7-kb范围之内(图2),分析此区段开放阅读框(orf)推测出候选基因。

(3)ds8基因的鉴定和功能分析:

为了验证候选基因功能,构建了如图3所示的互补载体,通过农杆菌介导的转基因技术将互补载体转入到突变体ds8愈伤中,获得了使突变体ds8恢复正常表型的转基因水稻植株(图4),证明本发明正确克隆了ds8基因。氨基酸序列分析表明ds8编码类nap1蛋白。

综上所述,本发明利用水稻耐旱相关突变体ds8,通过图位克隆技术首次在水稻中克隆到了类nap1蛋白编码基因,该基因影响水稻叶片表皮的发育和气孔闭合。通过对ds8基因的克隆和功能解读,有助于阐明水稻的耐旱机制。

具体实施方式

下面结合具体实施例对本发明进行进一步描述,但本发明的保护范围并不仅限于此:

实施例1、水稻类nap1蛋白编码基因ds8的图位克隆

1、水稻材料:

水稻(oryzasatival.)突变体ds8(droughtsensitive8),原始背景材料为粳稻品种“日本晴”。

水稻耐旱相关突变体ds8获取于中国浙江省境内,由日本晴(npb)ems(ethylmethylsulfonate)诱变产生(图1),诱变过程为用清水将待处理npb种子浸种8h以上,自然晾干。之后用1%浓度的ems溶液将npb种子完全浸入,28℃处理10h。用5%硫代硫酸钠作为终止剂并用流水冲洗10-12h,然后催芽、播种。

水稻野生型wt(npb)和突变体ds8苗期表型、分蘖期表型、灌浆期表型分别如图1a~c所示;将水稻野生型wt和突变体ds8于苗期进行干旱处理,将浸种催芽处理过后的种子播种于装土的盆钵中,于30℃12h光照/25℃12h黑暗,70%湿度的环境下培养,定期浇水,到三叶期的时候停止浇水10天,之后再定期浇水观察幼苗的恢复状态,统计幼苗的存活率。水稻野生型wt和突变体ds8干旱处理前后表型如图1d所示,幼苗存活率如图1e所示。分别取抽穗期野生型wt和突变体ds8不同植株上的剑叶叶片,称量鲜重之后放于28℃,60%湿度的培养箱中,每10min称量一次叶片重量,最后统计叶片失水速率,叶片失水速率如图1f所示。

2、遗传分析和定位群体构建:

通过ds8突变体与野生型籼稻品种93-11、zf802和cj06的正反交实验,f1代都为野生型表型,f2中野生表型和突变表型的分离经卡方检测发现符合3:1,表明该突变体受隐性单基因控制。另外从ds8和93-11f2群体中挑选突变表型(对干旱敏感)个体作为定位群体。从定位群体中随机抽取100个单株提取总dna用于构建混池dna并进行初定位。

dna提取采用水稻微量dna的快速提取方法,剪取适量叶片于2.0mlep管中,放入2颗小钢珠(直径约为0.5cm)并用液氮迅速冷却,然后放入高通量植物组织研磨器中高速震荡使叶片成粉末状,加入dna提取液750μl并置于65℃烘箱20min,加入500μl氯仿,剧烈上下震荡后,室温静置并离心。吸取上层清液转移至1.5ml离心管中,加入等体积预冷的异丙醇溶液,-20℃放置20min后离心,弃上清,沉淀用70%酒精洗涤一次后晾干,加入200μl超纯水溶解。

3、ds8基因的初步定位

以bsa(混合分离分析法)法构建早衰突变体混池和正常表型混池:利用公布的粳稻和籼稻创建的分子遗传图谱,选取近似均匀分布于12条染色体上的ssr引物,并进行pcr扩增,经5%琼脂糖凝胶电泳分离和溴化乙啶(eb)染色,检测引物的多态性,选取多态引物将ds8初步定位在第8号染色rm1345和rm3120两个标记之间。

4、ds8基因的精细定位

利用ncbi(http://www.ncbi.nlm.nih.gov/)公布的npb序列以及本实验室自保存序列,提取第8染色体rm1345和rm3120两个标记之间区段;在rm1345及rm3120之间设计加密标记,使用primer3.0在线设计引物网站进行多态引物的设计,同时使用gramene(http://www.gramene.org/)网站上的blast功能,预测引物特异性并同时确认引物扩增片段的物理距离;通过以上步骤,设计了在双亲中多态性极佳的多态引物(表1),选取ds8与93-11组合的f2群体中共2319株隐性个体,进行染色体步移,将ds8定位于bac克隆ap004632上e1与e2标记之间,物理距离53.7kb(图2)。

5、基因预测和比较分析:

在精细定位的基础上,利用ricegenomeannotationproject(http://rice.plantbiology.msu.edu/)网站对53.7-kb范围内的预测,发现在此区间内共有7个候选基因(图2)。设计各候选基因的测序引物(表2),分别从ds8和野生型品种基因组中扩增出候选基因进行测序分析。最终发现ds8突变体在loc_os08g43130的第3个外显子第一个1个碱基的发生替换突变,由g突变为a(图2)。根据rgap基因注释信息,预测此基因编码一个类nap1蛋白。

该基因的cdna全长如seqidno:1所示,gdna全长如seqidno:2所示。该基因编码的蛋白质的氨基酸序列如序列表的seqidno:3所示。

实施例2、ds8基因功能

设计引物com-f:gacctgcaggcatgcaagctacctgtaaactgcgagacga,com-r:cgacggccagtgccaagcttcctctccatcaagcacctca,以粳稻品种“日本晴”基因组dna为模板,pcr扩增候选基因序列atg前2081bp及taa后826bp,pcr扩增体系如下:

pcr扩增程序如下:

atg前2081bp及tga后826bp,连同seqidno:2所述序列,共14554bp片段。

通过重组连接的方式连接到hindiii酶切过的pcambia1300双元表达载体中,获得了互补载体pcambia1300-ds8。这个质粒通过电击转化的方法转入农秆菌(agrobacteriumtumefaciens)株系eha105中。利用突变体种子诱导愈伤,作为转化的受体。用含有pcambia1300-ds8载体的eha105菌株侵染愈伤并在25℃黑暗条件下共培养3天,用含有50mg/l潮霉素的筛选培养基筛选抗性愈伤并将抗性愈伤转入分化培养基上培养,最后将分化的转基因苗转至生根培养基上培养。对最终获得的转基因植株进行鉴定和观察发现:
