技术领域
本发明公开一种抗隐孢子虫重组免疫毒素蛋白及其制备工艺和医用用途,涉及预防和治疗隐孢子虫病的药物制备技术领域。
背景技术
隐孢子虫病(Cryptosporidiosis)是一种重要的人兽共患寄生虫病,该病具有以下特点该虫宿主范围极为广泛、感染率高、危害严重,隐孢子虫病可引起人和牛的严重腹泻,是艾滋病人的主要致死因素之一。犊牛隐孢子虫病死亡率高达53.5%,奶牛感染隐孢子虫后产奶量可降低3.2kg/d,耐过动物可成为带虫者和传染源,而且隐孢子虫可造成T细胞数量的急剧减少使机体免疫力降低。90%以上的隐孢子虫病人有腹泻症状,多呈水样便,并伴有腹痛、腹胀、呕吐、乏力和发热等。免疫功能缺陷者感染隐孢子虫后可引起严重的腹泻并危及生命。因此本病是今后医学和兽医界防治的重要寄生虫病。
迄今已有200余种药物包括所有抗生素、抗球虫药及磺胺类药等,被试用于治疗人和动物隐孢子虫病,但尚无一种药物对隐孢子虫有真正杀灭作用。长期以来人们期望的一种仅杀伤有害细胞而不损伤正常细胞的导向药物在肿瘤的治疗中发挥了重要作用。同时也显示出它在解决迄今缺乏有效治疗药物的感染性疾病方面的具大潜力,已成为一个活跃的研究领域。

发明内容
本发明提供一种抗隐孢子虫重组免疫毒素蛋白及其制备工艺,可适用于工业化生产。
本发明还提供了抗隐孢子虫重组免疫毒素蛋白在预防和治疗人和动物隐孢子虫病的医用用途。

本发明抗隐孢子虫重组免疫毒素蛋白制备工艺包括以下步骤利用单克隆抗体技术和基因重组技术,从分泌抗微小隐孢子虫(Cryptosporidium parvum)子孢子表膜抗体的杂交瘤细胞株中克隆其可变区基因,再连接成单链抗体基因,然后将其插入原核表达载体的相应位点上,构建了重组原核表达质粒,再分别采用酶切和PCR方法将去除受体结合区的细菌外毒素插入重组原核质粒中单链抗体基因的下游位点,即利用单链抗体基因代替细菌外毒素的受体结合区,构建重组表达质粒。将上述构建的重组表达质粒分别转化受体菌,经IPTG诱导,SDS-PAGE分析表达产物,后对表达产物包涵体进行纯化和复性,并对其进行体外抗虫活性测定和模型动物隐孢子虫病治疗。
通过以下试验例证明抗隐孢子虫重组免疫毒素蛋白对于对隐孢子虫疾病的预防和治疗作用实验例1.
60只小鼠每只经口接种1×106个隐孢子虫后,随机将其分为六组,一组至五组为治疗组,六组为对照组。治疗组每日经肛门分别灌入小肠不同剂量(0.5,1,1.5,2,2.5g)的重组毒素,连续3天,对照组以生理盐水代替重组毒素。7天后,取1克粪便计算每克粪便中的卵囊数。结果显示,治疗组卵囊数明显少于对照组(p<0.05)。其中治疗的最佳剂量为1g/只鼠。
实施例2.

本发明的积极效果在于蛋白基因易于修饰和改造、稳定性好、渗透性好、制备简单、特异性强、杀灭作用强,对隐孢子虫疾病具有很高的预防和治疗作用。

实施例1从人工感染隐孢子虫的犊牛粪便中用蔗糖密度梯度离心法分离纯化微小隐孢子虫卵囊(CSO),经超声粉碎(100W×5min)后免疫BALB/C小鼠,应用杂交瘤技术,制备出1株能稳定分泌抗隐孢子虫McAb的杂交瘤细胞株3D6,并通过免疫荧光法对其进行了鉴定,其在子孢子表面呈弥慢性荧光反应,提示其针对子孢子表膜。从分泌抗隐孢子虫子孢子单克隆抗体的杂交瘤细胞3D6中,提取其mRNA,在反转录酶的作用下,合成cDNA,经PCR扩增(反应条件94℃1min,55℃2min,72℃2min,30个循环)获得抗隐孢子虫子孢子表膜单克隆抗体的轻、重链可变区基因VL和VH。在这两个基因中加入连接肽基因后,经进一步PCR扩增、组装合成出单链抗体(ScFv)基因,将该基因克隆入pMD-18T载体后,进行了序列测定。测序结果表明,获得的单克隆抗体轻、重链可变区基因VL和VH符合小鼠抗体可变区基因特征,为功能性重排的鼠抗体可变区基因序列,分离得到的单链抗体基因ScFv由重链可变区基因VH-连接肽基因-轻链可变区VL基因构成,这种构成模式较VL-Linker-VH基因结构具有更强的抗体亲和活性。ScFv基因全长为720bp,其中VH基因为351bp,编码117个氨基酸,VL基因全长为324bp,编码108个氨基酸。
将获得的ScFv基因分别代替绿脓杆菌外毒素PE的受体结合区,利用原核表达载体pET28,构建了用于表达重组毒素的原核表达质粒3D6pET28-ScFv-PE(单链重组免疫毒素基因序列附后)。将上述质粒转化受体菌BL21(DE3)后,经IPTG诱导,成功地表达了目的蛋白。重组毒素3D6ScFv-PE40的表达量占菌体蛋白总量的14%,表达产物以包涵体和上清两种形式存在。将低温诱导获得的3D6ScFv-PE可溶性表达产物,加入含有脱囊后的子孢子的1640培养液中,观察18h内活动的子孢子数目和活动性的变化,结果与对照组相比,加入重组毒素组活动的子孢子数目明显减少(P<0.05),活动性明显降低,提示重组毒素对隐孢子虫子孢子具有较强的杀灭作用,其治疗隐孢子虫病模型动物的剂量为1μg/只鼠。
实施例2从人工感染隐孢子虫的犊牛粪便中用蔗糖密度梯度离心法分离纯化微小隐孢子虫卵囊(CSO),经超声粉碎(110W×6min)后免疫BALB/C小鼠,应用杂交瘤技术,制备出1株能稳定分泌抗隐孢子虫McAb的杂交瘤细胞株4G4,并通过免疫荧光法对其进行了鉴定发发现针对子孢子表膜。从分泌抗隐孢子虫子孢子单克隆抗体的杂交瘤细胞4G4中,提取mRNA,在反转录酶的作用下,合成cDNA,经PCR扩增(反应条件95℃1min,56℃2min,72℃2min,30个循环),获得抗隐孢子虫子孢子表膜单克隆抗体的轻、重链可变区基因VL和VH。在这两个基因中加入连接肽基因后,经进一步PCR扩增、组装合成出单链抗体(ScFv)基因,将该基因克隆入pMD-18T载体后,进行了序列测定。测序结果表明,获得的单克隆抗体轻、重链可变区基因VL和VH符合小鼠抗体可变区基因特征,为功能性重排的鼠抗体可变区基因序列,分离得到的单链抗体基因ScFv由重链可变区基因VH-连接肽基因-轻链可变区VL基因构成。4G4 ScFv基因全长为717bp,其中VH基因为348bp,编码116个氨基酸,VL基因全长为324bp,编码108个氨基酸。
将获得的ScFv基因分别代替绿脓杆菌外毒素PE的受体结合区,利用原核表达载体pET28,构建了用于表达重组毒素的原核表达质粒4G4pET28-ScFv-PE(单链重组免疫毒素基因序列附后)。将上述质粒转化受体菌BL21后,经IPTG诱导,成功地表达了目的蛋白。重组毒素4G4ScFv-PE40的表达量占菌体蛋白总量的12%,表达产物以包涵体和上清两种形式存在。将低温诱导获得的4G4ScFv-PE可溶性表达产物,加入含有脱囊后的子孢子的1640培养液中,观察20h内活动的子孢子数目和活动性的变化,结果与对照组相比,加入重组毒素组活动的子孢子数目明显减少(P<0.05),活动性明显降低,提示重组毒素对隐孢子虫子孢子具有较强的杀灭作用,其治疗隐孢子虫病模型动物的剂量为1.5μg/只鼠。

4.根据权利要求3所述的抗隐孢子虫重组免疫毒素蛋白制备工艺,包括以下步骤从人工感染隐孢子虫的犊牛粪便中用蔗糖密度梯度离心法分离纯化微小隐孢子虫卵囊,经超声粉碎后免疫BALB/C小鼠,应用杂交瘤技术,制备出1株能稳定分泌抗隐孢子虫McAb的杂交瘤细胞株3D6,并通过免疫荧光法对其进行了鉴定,其在子孢子表面呈弥慢性荧光反应,提示其针对子孢子表膜,从分泌抗隐孢子虫子孢子单克隆抗体的杂交瘤细胞3D6中,提取其mRNA,在反转录酶的作用下,合成cDNA,经PCR扩增,获得抗隐孢子虫子孢子表膜单克隆抗体的轻、重链可变区基因VL和VH,在这两个基因中加入连接肽基因后,经进一步PCR扩增、组装合成出单链抗体ScFv基因,将该基因克隆入pMD-18T载体后,进行了序列测定;将获得的ScFv基因分别代替绿脓杆菌外毒素PE的受体结合区,利用原核表达载体pET28,构建了用于表达重组毒素的原核表达质粒3D6pET28-ScFv-PE,将上述质粒转化受体菌BL21后,经IPTG诱导,成功地表达了目的蛋白。
