技术领域
本发明涉及用于疫苗组合物中的多肽佐剂。
佐剂是一种物质或方法,它可以通过调节免疫细胞的活性来特异地增强对抗原的免疫反应。佐剂的例子包括弗氏佐剂,胞壁酰二肽,脂质体。佐剂也可以是具有激动作用或拮抗作用的由免疫细胞表达的抗体直至受体。佐剂与载体的区别在于载体通常被用来增强对抗原的免疫反应。
载体是一种具有免疫原性的分子,当它与另一个分子相结合时可以增强后者的免疫反应。有一些本身不具有免疫原性的抗原(即在它们直接存在时不具有免疫原性),但当它们与诸如匙孔血蓝素(keyhole-limpethemocyanin)或破伤风类毒素的外源蛋白分子相连时则可具有产生抗体反应的能力。这些抗原含有B细胞表位而没有T细胞表位。这些轭合物(“载体”蛋白)的蛋白部分提供T细胞表位来刺激辅助性T细胞,从而刺激抗原特异性B细胞分化为浆细胞,并产生针对于该抗原的抗体。为免疫细胞受体的蛋白配基的佐剂可以同时具有佐剂和载体的性质,而后者依赖于可被免疫系统T细胞识别的“外源”多肽序列的组成。本发明中的新型佐剂具有以上所述的两种性质。
多蛋白抗原或多梯状(polyladder)蛋白是由多种寄生性或自由生活的线虫产生的。这些多蛋白通常包括许多直接串联排列的多亚基,这些蛋白通常作为大前体蛋白合成,其可被蛋白酶剪切产生大小间隔约为15KD的、“梯状”的小片段,反应单个功能域的平均分子量(denominatormolecular mass)的增加。每一个这样亚基的最后4个氨基酸都具有一种易被蛋白酶消化的RX(K/R)R(或有时是RXFR)基序。另外,在这些基序上游(基序的N端)的7,8或9残基位置所述基序前面有半胱氨酸残基(McReynolds et al.(1993)Parasitology today 9 403-406),该残基的作用在于可以使得蛋白酶的剪切位点远离该蛋白质功能域的主体。
某些寄生虫的多蛋白(例如犬恶丝虫的DiAg蛋白)是很强的免疫刺激剂,能够产生抗原非特异性IgE,并通过干扰寄生虫特异IgE的产生,在寄生虫引起的免疫反应规避中起到重要作用。
产生对众多传染性疾病起到预防保护作用的疫苗,是近代医学中最重要的进步之一。许多预防和治疗包括自身免疫性、神经退行性疾病以及癌症的其它种类疾病的疫苗目前尚在研发中。本发明也可以应用于这些其它种类的疾病。在多数情况下,预防传染性疾病的疫苗来自于引起该疾病的介质(病原体)的灭活或减毒形式,为了预防病原体天然形式所引起的感染,可对受体进行该疫苗的注射或其它形式的给药。受体个体可以通过产生体液(抗体)反应,细胞(例如细胞溶解性T细胞,CTL)反应或两者同时反应来实现免疫反应。
对所谓亚单位疫苗(其中的免疫原被定义为传染性介质(agent)或肿瘤抗原的分子片段或亚单位)的开发一直都是众多医学研究关注的焦点。用于亚单位疫苗的开发中的确定候选分子(例如,蛋白或多糖)的需求最初是基于提高安全性的需要,以及在针对细菌性感染的疫苗时,抗菌素耐受问题的增加也是一种原因。然而,与基于整个有机体获得的疫苗相比,亚单位疫苗的免疫原性越低,而为了产生有效的免疫反应(或产生有效的抗肿瘤免疫反应)以保护目标有机体不受感染,亚单位疫苗对“佐剂”的依赖性也越强。
我们描述了一个蛋白家族,它们可以作为免疫佐剂提高针对于多种预防性和治疗型疫苗所产生的免疫反应。佐剂系统在刺激疫苗抗原产生免疫反应方面相当有效。新的佐剂系统尤其适用于亚单位疫苗,但对其他种类的疫苗也同样适用(包括基于整个有机体、核酸等所获得的疫苗)。
已知线虫的多梯状蛋白对诱导IgE反应高度有效(Tomlinson et al.J.Immunol.143 2349-2356(1989);Paxton et al.Infect Immunol.612827-2833(1993)。然而,一些多梯状蛋白(例如犬恶丝虫的DiAg蛋白)则可以通过产生抗原非特异的IgE来扰乱正常的免疫反应,其不能与DiAg或寄生虫结合(Tezuka,H et al.Infection and Immunity,July 2003,3802-3811),并通过不相关IgE将寄生虫特异性的IgE排除在外,来干扰激活的肥大细胞和嗜伊红细胞对寄生虫去除。而且,因为IgE抗体常常与危险甚至是致命的过敏反应相关联(例如,如果在个体中含有预先存在的对于所述抗原特异的IgE抗体,该个体与该抗原相遇时就会发生过敏反应),因此IgE反应一般被认为是疫苗接种的非理想表现(至少是在针对不是寄生虫的介质的疫苗的情况下)。不仅如此,如果将其作为疫苗物质,那么可存在多梯状蛋白增强人个体内正在进行中的致敏原特异性的IgE反应、或干扰对于寄生虫预期的免疫反应的有限危险性。总之,通过多梯状蛋白诱导产生的IgE反应和可以干扰寄生虫去除或加重过敏性疾病的非特异性IgE反应,都是将寄生虫多梯状蛋白作为疫苗组分或是佐剂进行使用时的禁忌症。
令人惊奇的是,我们公开了在没有强烈非特异性IgE反应出现的时候,通过例如多梯状蛋白(或者优选与这些蛋白的单重复单元)的轭合或微粒共制剂的物理性连接是一种可较强地提高针对相联抗原产生免疫反应的方法。我们公开了多梯状蛋白(或者优选多梯状蛋白的个体单一功能域部分)和预期免疫反应所针对的抗原的物理性连接,能够将对于所述多梯状蛋白的潜在危险的非抗原特异性IgE反应转变为有益的佐剂效应,来产生针对所述抗原的预期抗原特异性抗体。我们还公开了所产生的抗原特异性免疫反应(针对多梯状-功能域-连接的疫苗抗原)是如何倾向于IgG的产生(适用于细菌病原体的去除示例),并倾向于适合去除诸如病毒(和某些寄生虫)的细胞内寄生物的Th1型T细胞反应的产生,而不是倾向于有潜在危险的IgE反应。我们还公开了多梯状蛋白或蛋白功能域是如何被用来产生针对相联疫苗抗原甚至是当该抗原以非微粒形式进行给药时的细胞溶解性T淋巴细胞(CTL)反应的。除此之外,我们还公开了多梯状蛋白功能域(甚至是DiAg多梯状蛋白的功能域)是如何被用来产生针对与其进行物理性连接的抗原(例如寄生虫抗原而并非DiAg和多梯状蛋白)的预期抗原特异性IgE反应。
DiAg及其相关蛋白已被证明在小鼠中能够将免疫系统偏离引起自身免疫I型(胰岛素依赖型)糖尿病的致病性Th1反应,并且提倡治疗性用于Th1为基础的自身免疫疾病的治疗(Imai,S.et al.Biochem.Biophys.Res.Comm.2861051-1058)。因此令人惊奇的是,在本发明的另一方面,我们现已公开了DiAg及其相关蛋白是如何进行过敏性疾病治疗的,它们与涉及过敏性疾病的发病机理中T细胞的作用(Th1)正好完全相反(Th2)。以下对产生DiAg蛋白这种新用途的必需组合物和方法进行了描述。
一方面,本发明提供作为免疫佐剂用途的多肽,所述多肽包括i)由RXK/RR氨基酸残基组成的氨基酸基序,R为精氨酸,X为任一氨基酸残基,及K为赖氨酸;和/或ii)由RXFR氨基酸残基组成的氨基酸基序,其中F为苯丙氨酸,且在所述基序氨基端前约7-9残基位置所述基序前面有半胱氨酸残基,所述多肽能够通过至少一个氨基酸残基的添加、缺失或取代来修饰。
一方面,本发明提供的含有由核酸分子编码的多肽的佐剂,其中有至少一个RX(K/R)R(其中R为精氨酸,X为任一氨基酸,K/R为赖氨酸或精氨酸,R为精氨酸)或RXFR(其中F为苯丙氨酸)氨基酸基序,在此RX(K/R)R或RXFR基序N端前7,8或9残基位置该基序前面有半胱氨酸残基。
优选的所述多肽由线虫的核酸分子编码。
在本发明的另一实施方案中,提供了所述蛋白片段的佐剂,并优选为淋巴细胞结合片段。
在本发明的另一实施方案中,提供了与所述蛋白或与所述蛋白片段、并优选淋巴细胞结合片段具有至少70%同源性的佐剂,。
另一方面,本发明提供含有至少一种多肽的疫苗组合物,其中所述多肽包括i)由氨基酸残基RX(K/R)R组成的氨基酸基序,其中R为精氨酸,X为任一氨基酸,K为赖氨酸;和/或ii)由RXFR氨基酸残基组成的氨基酸基序,其中F为苯丙氨酸,且在所述基序氨基末端前约7-9残基位置所述基序前面有半胱氨酸残基,所述多肽能够通过至少一个氨基酸残基的添加、缺失或取代来修饰,以及至少一种可以引发预期免疫反应的抗原来修饰。
在本发明的优选实施方案中,所述多肽与所述抗原混合。
在本发明的另一优选实施方案中,所述多肽与所述抗原轭合,连接或交联。
在本发明的另一优选实施方案中,该多肽含有犬恶丝虫(Dirofilariaimmitis)蛋白,嗜中性白细胞趋化因子(NCF),或其淋巴细胞结合片段,或其同源结构,或其同源结构的淋巴细胞结合片段。
在本发明的优选实施方案中,所述多肽包括选自SEQ NO.1;2;3;4;5;6;7;8;9;10;11;12;13;14;15;16;17;18;19中的氨基酸序列,或与所选自的序列具有至少50%同源性、更优选具有70%的同源性、甚至更优选具有90%同源性的多肽。
在本发明的优选实施方案中,所述多肽的长度为至少20个连续的氨基酸,其序列相同于选自SEQ ID No1;2;3;4;5;6;7;8;9;10;11;12;13;14;15;16;17;18或19序列中的至少20个氨基酸部分。
在本发明的优选实施方案中,在本发明的优选实施方案中,所述多肽是这种蛋白的淋巴细胞结合片段。
在本发明的另一实施方案中,所述多肽由包括选自SEQ ID No20;21;22;23;24;25;26;27;28;29;30;31;32;33或34的核酸序列的核酸分子;或由与所述核酸分子杂交的编码具有免疫佐剂活性多肽的核酸分子所编码。
优选的所述核酸序列与所选自的序列具有至少50%的同源性,或更优选与所选自的核酸序列具有至少70%的同源性,或更优选的与所选自的序列具有至少85%的同源性。
在本发明的另一实施方案中,所述多肽由选自SEQ ID No20;21;22;23;24;25;26;27;28;29;30;31;32;33或34中的核酸序列的60个核苷酸部分所编码。
在本发明的另一实施方案中,佐剂蛋白由与线虫梯状蛋白部分同源的蛋白组成,该蛋白部分含有但不包括全部的RX(K/R)R或RXFR序列,并最优选避开该序列中的R,K(有时为F)残基。
在本发明的另一实施方案中,RX(K/R)R蛋白酶剪切基序(序列(1)中下划线部分)发生了突变,或未包括在佐剂蛋白中。
这种突变序列的应用例子是其中的R和K残基被甘氨酸残基“G”所取代的序列。第二个例子是其中R和K残基被丝氨酸残基“S”所取代的序列。第三个例子是其中R和K残基被任意置换的G或S所取代(例如GXGS,SXSG,GXSG等)。
可用的连接序列可能比天然多梯状蛋白中的长(例如至多达30个残基),最优选5-20个氨基酸残基,且通常缺乏较强的形成二级结构(例如形成螺旋或形成β-折叠)的倾向以及通常缺乏可被胰蛋白酶样酶剪切的残基(原则上是K和R)。
这种形成二级结构倾向的缺乏可用于间隔地连接脯氨酸残基的工程化,该间隔例如每第三个残基或者每第十个残基,更优选每第4个,第5个或第6个残基。可选择地所述脯氨酸可随机的分布于融合蛋白序列的融合区域中,使得5个残基的较小的连接序列含有1个脯氨酸,而15个残基的较大的连接序列中含有3个或4个脯氨酸。
连接序列的目的在于首先将融合蛋白(也就是多梯状蛋白半族和抗原半族)中的两个蛋白部分,以一种对蛋白酶(与天然的多梯状蛋白序列情况不同,其中所述的功能域间边界对蛋白酶是非常稳定的)相对稳定的方式,连接在一起。所述序列的第二个功能是允许蛋白部分在生物合成过程中发生折叠以形成它们功能属性(佐剂性,抗原性/免疫原性)所依赖的天然结构域。当使用的连接多肽采用多种形式时,所述连接中不能含有来源于人自身抗原(或欲免疫动物的自身抗原)的序列是非常重要的。因此,通常应在数据库中筛选假定的连接序列,以确保连接序列与人类蛋白不具有显著的同源性,尤其是已知蛋白,或那些怀疑在自身免疫疾病病原学中起重要作用的诸如谷氨酸脱羧酶、胰岛素、甲状腺球蛋白、甲状腺过氧化物酶、胰岛细胞自身抗原、壁细胞自身抗原、肾自身抗原、髓鞘基础蛋白、髓鞘相关糖蛋白、髓鞘少突胶质糖蛋白的蛋白。
这项普遍规则的一个例外情况是当这些蛋白以非器官特异性分布于机体,或高丰度表达-例如血液白蛋白和免疫球蛋白的时候。例如,IgG的铰链区因为其并不是蛋白酶特异不稳定的,因此可为所述的融合蛋白提供很好的连接序列。类似地,尽管此序列暴露在外(以IgA的三维结构存在)且易于弯曲,由于仅有少数几种蛋白酶(例如脑膜炎球菌IgA蛋白酶)可以剪切这段序列,所以IgA的铰链区可为所述融合蛋白提供很好的连接序列。由于作为佐剂使用的蛋白开始于一个剪切基序的下游(羧基端),并中止在下一个基序的上游(氨基端),所以RX(K/R)R或RXFR基序可不包括在佐剂蛋白中。佐剂蛋白的起点和终点也可以位于蛋白酶基序的内部。
在本发明的一个优选实施方案中,所述多肽可与所述抗原可通过诸如戊二醛或EDC(乙基碳化二亚胺-一种水溶性的碳化二亚胺)的蛋白交联剂或优选通过诸如MBS或文献中以及Pierce Chemical Company ofRockford,Illinois,USA或Molecular Probes Inc.of Eugene,Oregon,USA目录中所述的其他类型的异型双功能试剂进行轭合或交联。
在本发明的另一优选实施方案中,采用本领域中公知的体外DNA重组技术和克隆技术方法,通过将编码抗原和佐剂的核酸进行读码框融合,产生含有所述抗原的融合蛋白形式的所述佐剂。
在本发明的另一优选实施方案中,采用合成的微粒或纳米微粒(例如聚交酯-乙交酯或“PLG”),脂质体或免疫刺激复合物(ISCOMs)将所述多肽和所述抗原包被起来。
微粒制剂是所期望的,因为它引导抗原至抗原提呈细胞,该细胞偏好针对所述抗原的Th1形式的免疫反应,而且对抗所述多梯状蛋白部分朝向Th2形式的表现和IgE产生的任何倾向。这种制剂形式在针对抗原产生的所期望细胞介导的和IgG抗体反应的刺激是有用的。微粒制剂还能容易地整合用来将免疫反应偏移至Th1方向上的附加材料。这些材料通常包括抗IL10和IL4,诸如IFN-γ的Th1细胞因子和CpG DNA的抗体。
最好的情况是,微粒制剂包含所述的多梯状蛋白部分和抗原,针对该抗原在同一个微粒中形成预期的免疫反应,从而使制剂中的每一个微粒都有效负载地携带两个实体。这种制剂可以保证两种物质被任何给定的单一抗原体呈细胞摄取并最大化对于所述有效负载抗原的微粒制剂的Th1倾向作用,甚至当此抗原和多梯状功能域(以及上一段中描述的任意的Th1倾向性物质)并没有相连时,同时存在于同一微粒中除外。
微粒制剂优选具有显著程度的多梯状蛋白部分和抗原部分的表面暴露(5-10%)。一般情况下,微粒制剂过程中就可默认形成这种暴露程度。如果这样的暴露程度不能实现,上述蛋白部分可以通过共价轭合的形式轭合到所述微粒表面。抗原部分的表面暴露有利于刺激抗原特异性B细胞,并且有助于对于抗原的抗体反应。
通常这些微粒的大小为150纳米最高至10微米直径。更优选它们为200纳米最高至2微米。
在本发明的一个优选实施方案中,所述多肽和抗原共吸收或共沉淀在诸如氢氧化铝凝胶或磷酸钙的铝或钙盐上。
在本发明的另一优选实施方案中,所述多肽由核酸分子编码,该核酸分子是载体的一部分,其中所述多肽的表达由启动子可操纵地控制。
在本发明的另一优选实施方案中,所述抗原由核酸分子编码,该核酸分子是载体的一部分,其中所述抗原的表达由启动子可操纵地控制。
在本发明的一个优选实施方案中,该多肽和抗原由相同的核酸分子编码。优选的所述核酸分子编码所述多肽和所述抗原的阅读框架融合形式。
在本发明的一个优选实施方案中,所述阅读框架融合形式包括编码柔性连接序列(例如编码寡丝氨酸或甘氨酸或一定残基数量的丝氨酸-甘氨酸组合)的连接核酸分子。
优选的所述载体是一种“穿梭载体”,可以在大肠杆菌中进行扩增,并通过适合的诸如CMV或其它真核启动子的启动子在哺乳动物细胞中表达融合蛋白。
根据本发明另一方面,佐剂蛋白功能域(例如来自于犬恶丝虫的NCF)可由几个拷贝(最优选1,2或3拷贝)代表,并作为同一条多肽链的部分与抗原(单一拷贝)形成共线性融合。可选择地,佐剂蛋白功能域以单拷贝存在,与抗原蛋白融合形成寡聚体(例如二聚体,三聚体,四聚体等)。在后者的构建中,一旦抗原蛋白发生寡聚化,佐剂蛋白功能域就成为寡聚结构。最方便的情况是,单拷贝的佐剂蛋白与来自于传染性介质的寡聚蛋白形成读码框融合,针对该介质进行预防疫苗设计,例如流感红细胞凝集素或HIV衣壳糖蛋白gp120。可选择地,单拷贝佐剂蛋白功能域与不发生寡聚化的抗原相融合。在这种情况下,寡聚体形式可以通过与具有天然寡聚化倾向的蛋白部分(例如卷曲化卷曲)结合来产生。适合的寡聚化部分的例子为链球菌中(例如M蛋白)配对的螺旋卷曲化卷曲结构,其可形成二聚体卷曲化卷曲;同样也可以使用三聚体螺旋蛋白部分。其中一个例子是诸如CD23的2型膜蛋白的茎结构。被设计用来研究卷曲化卷曲蛋白的聚合性质的人工卷曲化卷曲多肽也是合适的。最优选的寡聚化程度是三聚体,因为它反应出犬恶丝虫(D.immitis)蛋白的预测天然存在形式。在佐剂功能域以多个拷贝的形式出现的情况下,最优选的实施方案将是犬恶丝虫蛋白的天然重复结构。当结构域作为单一多肽链的部分进行重复时,最优选的是在DNA水平上发掘可替代的密码子的使用,以避免直接的DNA序列重复,该序列在编码DNA扩增过程中会另外产生同源重组的问题和缺失问题。
通过采用交联剂将佐剂和抗原相连以形成轭合物。可选择地轭合物也可以是佐剂和抗原的翻译融合物。
在本发明的另一优选实施方案中,所述组合物包含有载体。

这种治疗过敏性疾病方式的优点在于它可以应用于所有或任何一种过敏性疾病,而不需要考虑过敏原,甚至在对过敏原一无所知(例如过敏性哮喘的情况)的情况下依然有效。DiAg及其相关多梯状蛋白,可以被用来产生非同寻常的大量非抗原特异性IgE来竞争肥大细胞和嗜伊红细胞表面的位点(高亲和性IgE受体),剥夺这些细胞与过敏源特异性IgE的结合,防止它们在与过敏源接触后被激活以及释放出炎症介质。
例如,因为DiAg在人寄生性感染中并不产生DiAg特异性的IgE,所以使用DiAg进行治疗产生过敏反应的危险是最小的。根据本发明的这一方面,可以使用单个多梯状(例如DiAg)功能域。这样的功能域能够以药物可接受的盐赋形剂制成蛋白溶液进行给药,或者在用于哺乳动物表达的质粒或病毒载体中由DNA或RNA编码,或者存在于作为能够在体内表达的质粒构建的疫苗脂质体载体中。
本发明另一方面提供增强针对多价疫苗的免疫反应的方法,尤其是通过载体蛋白-多梯状轭合物或嵌合蛋白与诸如相同载体蛋白与多糖抗原的多种抗原轭合物共形成的多价多糖疫苗。
在这种情况下,这两种载体蛋白通常可以是完全相同的蛋白,或者是相互之间至少具有一个相同的T辅助细胞表位的不同蛋白。其中的一种或两种都可以是合成的多肽。其中的一个例子是多价肺炎球菌(pneumoccal)轭合物疫苗,它含有数个不同的多糖,每个多糖都与突变的白喉(diphtheria)类毒素相轭合。通过向这种多梯状蛋白-白喉类毒素轭合物的混合轭合物中简单添加,针对该轭合物中所有多糖抗原的免疫反应都将被强烈的提升。

本发明所用“载体”可以是所期望序列可插入其中的任意一些核酸。载体包括但不限于质粒、噬菌粒和病毒基因组。克隆载体是指可以在宿主细胞中复制,且通常具有一个或多个限制性内切酶识别位点,并在此位点以确定的方式进行该载体剪切,并将期望的DNA序列连接进来,从而在宿主细胞中保留重组载体的复制能力。在质粒的情况下,在宿主通过有丝分裂进行繁殖前,在宿主细菌体内质粒以拷贝数增加的时候,期望序列的复制可以发生很多次或在每个宿主中仅发生一次。在噬菌体的情况下,在裂解期可以主动发生复制,而在溶原期被动发生。

适应性还包括自我复制序列,其同时适合所述载体在真核细胞或原核寄主中的维持,因此称为“穿梭载体”。维持自我复制的载体被称为游离型载体。因为这些分子能够连接较大的DNA片段(30-50kb DNA),所以游离型载体是所需要的。这类游离型载体的如国际公开第WO98/07876号所述。
促进编码基因载体病毒的适应性包括转录终止/多聚腺苷酸序列本表达的基因。其还也包括内部核糖体进入位点(IRES),其功能是最大化以双顺反子或多顺反子表达盒排列的编码基因载体的表达。

本领域中公知载体中的核酸序列被称为CpG基序或者是ISSs(免疫刺激序列)。尽管与这些二核苷酸相邻的序列可以影响诱导的刺激程度,在这些序列最低限度由作为中心的非甲基化CG二核苷酸组成。这些ISSs可以通过toll-样受体(TLR9)来激活抗原体呈细胞(APCs)。考虑到它们可以通过激活APCs来增强免疫反应,接种DNA疫苗的总的目的就应该将这些基序包含载体中。

参考以下的材料、方法和序列现仅以实施例的方式对本发明的实施方案进行描述。
材料与方法1.抗原-佐剂轭合物的制备a.重组佐剂蛋白的制备采用多聚酶链式反应(PCR),利用引物(5_引物,包含NdeI限制性内切酶识别位点5_GCATATGAATGATCATAATTTAGAAAGC-3,3_引物,包含BamHI限制性内切酶识别位点5_-CTAAAGGATCCTATCACCGCTTACGCCGTTCATTCATTG-3),从犬恶丝虫的cDNA文库对DiAg基因的V1功能域(一个重复单元)进行扩增。用NdeI和BamHI消化扩增的DNA后克隆到pET3a载体上(Stratagene),在大肠杆菌HMS174(DE3)中进行表达。根据以下所述进行rDiAg的纯化。用30ml 50mM HCl和5mMEDTA在4℃重悬5g细胞团,然后12,000 g离心10min。采用60-80%的饱和硫酸铵对上清液中的重组DiAg进行沉淀后过Superdex 200柱。采用固定化的多粘菌素B移去浓缩rDiAg溶液中大肠杆菌热源污染物。纯化后的佐剂蛋白经过冻干处理后储存于-20℃直至使用。
b.佐剂蛋白与抗原的轭合下面仅对从多种将抗体和多肽偶联的方法选出的一种作为示例进行描述将犬恶丝虫多梯状蛋白V1结构域佐剂(YFQTYLSWLTDAQKDEIKKMKEEGKSKMIQKKIFDYFESLTGDKKKKAAEELQQGCLMALSEIIGNEKMLMLKEIKDSGADPEQIEDMLKLVVDKEKKKRIDEYPPVCRKIYAAMNERRK)以3-30mg/ml的浓度溶解于pH 7.2的0.1M磷酸钠、0.15M NaCl溶液中。向其中加入6mg的硫代-SMCC(Pierce),将该混合物于室温孵育30min。采用pH 7.2的0.1M磷酸钠,0.15M NaCl作为层析缓冲液,以脱盐柱(Sephadex G-25)或超滤的方法可随即去除过量的交联物。将含有多肽(经OD280测定)的组分合并,并将马来酰亚胺激活的多肽浓缩到大约10mg/ml。
与此同时,抗原例如,纯化的重组HSV糖蛋白D抗原(gD)以1-5mg/ml的浓度溶解或交换于pH 7.2的0.1M磷酸钠、0.15M NaCl溶液中。以为1mg/ml浓度,在每ml gD中加入10-40μl SATA(Pierce)储存液(8mg/ml溶于DMSO或DMF)。室温反应30min。通过透析、凝胶过滤或超滤从未反应的SATA中纯化gD。
位于SATA修饰gD上的乙酰巯基化基团通过以下描述进行脱保护制备pH7.2的含有10mM EDTA的0.1M磷酸钠中的0.5M羟胺溶液。每毫升抗体中加入100μl上述溶液,室温放置2小时。将硫醇化的gD超滤纯化到pH7.2的0.1M磷酸钠、0.1M氯化钠、10mM EDTA溶液,并随即与经马来酰亚胺处理的DiAg以gD∶DiAg的1∶10的摩尔比混合。37C反应持续两小时。采用超滤的方法从未发生反应的DiAg中纯化轭合的gD-DiAg。
2.抗原佐剂融合蛋白的制备采用多聚酶链式反应(PCR),利用引物(5_引物,包含HindII限制性内切酶识别位点5_GAAGCTTAATGATCATAATTTAGAAAGC-3,3_引物,包含BamHI限制性内切酶识别位点5_-CTAAAGGATCCTATCACCGCTTACGCCGTTCATTCATTG-3),从犬恶丝虫的cDNA文库扩增犬恶丝虫多梯状蛋白的V1功能域(一个重复单元)。用HindIII和BamHI消化扩增的DNA。
与此同时,从HSV感染的细胞,使用结合附加的核酸的引物和在5’引物具有BamHI识别序列的引物,通过扩增得到gD编码DNA,从而通过BamHI消化并与DiAg的编码片段相连,使编码DiAg和gD的cDNA是相互在共同的读码框中,使得所述DNA连续地转录为可翻译成由DiAg和gD组成的融合蛋白的mRNA。将这两个DNA片段连入哺乳动物表达载体(pcDNA3.1(Invitrogen)),并将该质粒转化感受态大肠杆菌以产生足量的质粒(使用Qiagen柱进行纯化)来对COS-7细胞进行电穿孔转染。3-5天后,采用抗-DiAg亲和柱对转染的COS-7细胞的上清液进行纯化,在经过凝胶过滤的进一步纯化后,融合蛋白就可以用来进行免疫了。
3.编码抗原-佐剂融合蛋白的DNA疫苗的制备含有插入片段并如上所述制备的pcDNA3.1表达载体,可以直接作为DNA疫苗通过肌肉注射来使用。
