一种杀虫融合蛋白、编码基因及其应用的制作方法与工艺
本发明涉及一种杀虫融合蛋白、该杀虫融合的编码基因,及上述融合蛋白的应用。(二)
背景技术
:害虫给全球农业生产带来巨大的损失,目前害虫防治主要依靠化学农药,但是农药的使用加重了生产成本,并且农药残留对人体健康带来严重危害。因此,利用基因工程方法防治害虫具有重大经济、环境和社会价值。获得转基因抗虫作物的关键是克隆优良杀虫蛋白质。杀虫蛋白质有很多,使用得最为广泛的是苏云金芽孢杆菌(Bacillusthuringiensis,简称Bt)在产胞期间所分泌一种伴胞晶体蛋白(insecticidalcrystalproteins,ICP),如Cry1Ab,Cry1C等,这些蛋白对鳞翅目、双翅目、鞘翅目等昆虫(比如小菜蛾、玉米螟)有很强的杀伤作用(Schnepf,E.,Crickmore,N.,Van,R.J.,Lereclus,D.,Baum,J.,&Feitelson,J.,etal.1998,62(3),775-806.),目前已被深入研究和广泛使用,玉米、大豆、土豆、棉花等转Bt基因作物已得到大规模商业化种植。苏云金芽胞杆菌在营养生长期间,还会分泌一种与ICPs无氨基酸序列同源性、杀虫机理完全不同的蛋白,即营养期杀虫蛋白(vegetativeinsecticidalproteins,VIPs),如Vip3A、Vip3B、Vip3C、Vip3D、Vip3H,对一些对ICPs不敏感的害虫也有杀虫活性,并且不会发生交叉抗性(Estruch,J.J.,Warren,G.W.,Mullins,M.A.,Nye,G.J.,Craig,J.A.,&Koziel,M.G.1996,ProceedingsoftheNationalAcademyofSciencesoftheUnitedStatesofAmerica,93(11),5389-94.)。Vip3蛋白作为一种新型的杀虫蛋白,对目前使用最为广泛的ICP蛋白从杀虫机制、杀虫谱到杀虫活性上都是一个很好的补充。比如,ICPs对斜纹夜蛾和粘虫的杀伤作用很弱,Vip3则对斜纹夜蛾和粘虫有非常好的防治效果。尽管如此,在转基因作物研发的实际操作中,仍然存在一些急需解决的技术性问题。例如,研究人员发现高表达单独的Vip3基因的植株生长比较迟缓,容易出现白化现象。这可能是由于转基因植物中直接表达Vip3蛋白后,该蛋白由于信号肽的分泌作用大量聚集并结合在植物细胞膜上并形成孔道,从而对细胞造成损坏,影响其生长发育。玉米是全世界产量最高的粮食作物,同时也面临着诸多害虫的威胁。而在这些害虫当中,根萤叶甲的名字尤其让人胆寒——它们是破坏力最强的玉米害虫之一。根萤叶甲的幼虫侵入寄主的根茎后,会吞食组织,导致根腐烂、植株倒伏,使得植株减产乃至死亡。而成虫也会采食寄主的叶、花以及种子。在1955年以前,它们仅分布在美国有限的几个州内,但在这之后,它们侵入了本不太适合它们生存的美国北部。1990年,东欧激荡,这种害虫就在这个时候被意外引入塞尔维亚,于是,欧洲大陆尤其是东欧,也成了它们的地盘。而近些年也有蔓延至中国的境内。根萤叶甲是根萤叶甲属(Diabroticasp.)昆虫的统称。在美国,危害玉米的几种根萤叶甲被称作玉米根虫(CornRootworm),其中危害最大的三种被叫做“西方玉米根虫”、“北方玉米根虫”及“南方玉米根虫”。目前发现的具有玉米根虫杀灭效果的蛋白主要是来自苏云金杆菌中的Cry蛋白和Vip蛋白。其中,mCry3A、Cry3Bb1、eCry3.1Ab和蛋白复合体Cry34Ab1/Cry35Ab1已经在商业化的转基因品系中应用,并且已经产生了不同程度的抗性(LefkoSA,NowatzkiTM,ThompsonSD,etal.2008,JournalofAppliedEntomology,132(3),189–204;Zhao,J.Z.,Oneal,M.A.,Richtman,N.M.,Thompson,S.D.,Cowart,M.C.,&Nelson,M.E.,etal.2016,JournalofEconomicEntomology(3))。也有研究表明,RNAi技术可以用于玉米根虫等鞘翅目害虫的防治(Baum,J.A.,Bogaert,T.,Clinton,W.,Heck,G.R.,Feldmann,P.,&Ilagan,O.,etal.2007,NatureBiotechnology,25(11),1322-6.)。最近,杜邦公司从假单胞菌(Pseudomonas)中克隆的一个基因IPD072Aa具有杀灭玉米根虫的效果(Schellenberger,U.,Oral,J.,Rosen,B.A.,Wei,J.Z.,Zhu,G.,&Xie,W.,etal.2016,Science,354(6312),635-637.)。第一代转基因作物大多只具有抗鳞翅目害虫的特性,而第二代转基因作物正在向同时抗多种害虫的复合性状发展。获得具有复合性状的转基因作物有多种方法,比如通过对具有单一性状的转基因作物的杂交;通过将多个基因表达框构建在同一个表达框里面;通过共转化,将多个含有单基因的农杆菌混在一起,进行混合转化,筛选出多个基因同时整合了两个或多个质粒T-DNA的转基因植株。但是上述方法在实际操作和应用中都还存在一些问题。因此,获得更为简单高效的培育具有复合性状的转基因作物的方法是当下需要切实解决的问题。(三)技术实现要素:本发明目的是提供一种通过基因融合使得同一个杀虫融合蛋白同时对鳞翅目和鞘翅目害虫具有杀伤效果的方法。本发明采用的技术方案是:本发明提供一种杀虫融合蛋白,所述杀虫融合蛋白从N端到C端分别为IPD072蛋白和Vip3毒素。进一步,所述IPD072蛋白为IPD072Aa蛋白、IPD072Bb蛋白、IPD072Ca蛋白或IPD072Da蛋白之一;所述Vip毒素为Vip3A毒素、Vip3B毒素、Vip3C毒素、Vip3D毒素或Vip3H毒素中的一种。进一步,所述杀虫融合蛋白由IPD072Aa蛋白和Vip3A毒素融合而成,氨基酸序列为SEQIDNO:2所示。进一步,所述杀虫融合蛋白由IPD072Aa蛋白和Vip3H毒素融合而成,氨基酸序列为SEQIDNO:4所示。本发明还提供一种所述杀虫融合蛋白的编码基因,所述的基因从5’-3’依次为编码IPD072蛋白的核苷酸序列和编码Vip3毒素的核苷酸序列;且上述2个核苷酸序列位于同一个开放阅读框内。进一步,所述的编码IPD072蛋白的基因为IPD072Aa、IPD072Bb、IPD072Ca、IPD072Da之一;所述编码Vip3毒素的基因为Vip3A、Vip3B、Vip3C、Vip3D或Vip3H之一。更优选,所述编码基因的核苷酸序列为SEQIDNO:1或SEQIDNO:3所示。本发明还提供一种所述的杀虫融合蛋白在制备转基因抗虫作物、转基因杀虫微生物或抗体中的应用。进一步,所述转基因抗虫作物是由融合蛋白联合BT晶体毒素转化制备而成。与现有技术相比,本发明有益效果主要体现在:本发明所设计的IPD072蛋白与一个Vip3毒素融合成的人工蛋白质分子,与原先的IPD072蛋白和Vip3蛋白相比,具有如下优点:杀虫谱广,能同时防治鳞翅目和鞘翅目中多种重要害虫(如甜菜夜蛾、粘虫、玉米根虫),杀虫效率高达50%-100%;有利于具有上述不同功能的蛋白和其它抗虫蛋白(如ICPs)联合使用,进一步扩大杀虫谱,延缓害虫抗性产生。(四)附图说明图1是本发明的杀虫融合蛋白的结构图,杀虫融合蛋白中N端为IPD072蛋白,C端为Vip3毒素。图2是本发明中将杀虫融合蛋白基因导入植物中的表达框结构图;pUBI为玉米泛素启动子,杀虫融合蛋白基因为IPD072-Vip3毒素融合基因。图3是本发明中将杀虫融合蛋白和ICPs联合导入植物中的表达框结构图;pUBI为玉米泛素启动子,杀虫融合蛋白基因为IPD072-Vip3毒素融合基因,p35S为花椰菜花叶病毒35S启动子,ICPs为伴胞晶体蛋白编码基因。(五)具体实施方式下面结合具体实施例对本发明进行进一步描述,但本发明的保护范围并不仅限于此:实施例1、IPD072Aa-Vip3A杀虫融合蛋白表达载体的构建IPD072Aa和Vip3A杀虫蛋白的基因均由上海生工合成,其DNA序列分别为SEQIDNO:5和中国专利200610049611.0中的SEQIDNO:7,并克隆在pET28a表达载体的限制性内切酶BamHI和SacI位点之间。构建好的载体分别命名为pET28a-IPD072Aa和pET28a-Vip3A。IPD072Aa-Vip3A合成具体步骤如下:1、以Vip3A(中国专利200610049611.0中的SEQIDNO:7)作为模板进过PCR获得Vip3A基因片段。引物为:Vip3A-F:5’CCCGGGAAGGGTGGAGGAATGAACAAGAACAACACCAAG和Vip3A-R:5’CGAGCTCCTACTTGATGCTCACGTCGTAGAACTTCACGA。这两个引物分别包含限制性内切酶位点XmalI和ScaI的位点。2、用XmalI和ScaI处理PCR产物,并与用相同的酶处理过的pET28a-IPD072Aa载体连接。3、转入大肠杆菌,获得质粒为pET28a-IPD072Aa-Vip3A。融合蛋白基因IPD072Aa-Vip3A(SEQIDNO:1)被克隆到了pET28a表达载体中,融合蛋白基因编码的融合蛋白IPD072Aa-Vip3A(SEQIDNO:2)分子量大约为98kD。实施例2、IPD072Aa-Vip3H融合蛋白表达载体的构建IPD072Aa和Vip3H杀虫蛋白的基因均由上海生工合成,其DNA序列分别为SEQIDNO:5和SEQIDNO:6,并克隆在pET28a表达载体的限制性内切酶BamHI和SacI位点之间。构建好的载体分别命名为pET28a-IPD072Aa和pET28a-Vip3H。IPD072Aa-Vip3H合成具体步骤如下:4、以Vip3H(SEQIDNO:6)作为模板进过PCR获得Vip3A基因片段。引物为:Vip3H-F:5’CCCGGGAAGGGTGGAGGAATGAACAAGAACAACAGTAAG和Vip3H-R:5’CGAGCTCCTACTTGATGCTGAAGTCCCTGAAGGTGATGT。这两个引物分别包含限制性内切酶位点XmalI和ScaI的位点。5、用XmalI和ScaI处理PCR产物,并与用相同的酶处理过的pET28a-IPD072Aa载体连接。6、转入大肠杆菌,获得质粒为pET28a-IPD072Aa-Vip3H。融合蛋白基因IPD072Aa-Vip3H(SEQIDNO:3)被克隆到了pET28a表达载体中,融合蛋白基因编码的融合蛋白IPD072Aa-Vip3H(SEQIDNO:4)分子量大约为98kD。实施例3、杀虫蛋白的表达利用通用标准方法将上述含融合蛋白基因的表达载体(pET28a-IPD072Aa-Vip3A、pET28a-IPD072Aa-Vip3H、pET28a-IPD072Aa、pET28a-Vip3A和pET28a-Vip3H)分别转入大肠杆菌BL21star。挑取单克隆菌落接种到100mlLB细菌培养液中,37℃震动培养至OD600=0.6,加入IPTG到0.5mM,继续在相同条件下培育4小时。收集培养液5000g离心10分钟沉淀大肠杆菌细胞,然后弃上清收集沉淀。沉淀中加30毫升pH7.5、20mMTris-HCL缓冲液,超声粉碎后即可用于杀虫活性的测定。实施例4、IPD072Aa-Vip3A和IPD072Aa-Vip3H对多种鳞翅目和鞘翅目害虫具有杀伤作用实施例3所获得的杀虫蛋白(IPD072Aa-Vip3A、IPD072Aa-Vip3H、IPD072Aa、Vip3A和Vip3H)分别涂在昆虫人工饲料的表面,新生一龄幼虫用来进行杀虫性测定。阴性对照为pET28a空载体,准备方法与实施例3相同。杀虫活性结果如表1所示:表1、融合蛋白的杀虫效率粘虫甜菜夜蛾西方玉米根虫IPD072Aa0%0%100%Vip3A100%100%0%Vip3H100%100%0%IPD072Aa-Vip3A100%100%100%IPD072Aa-Vip3H100%100%100%阴性对照0%0%0%实施例5、IPD072Aa-Vip3A植物转化载体构建以IPD072Aa-Vip3A(SEQIDNO:1)作为模板经过PCR获得IPD072Aa-Vip3A基因片段。引物为:IVA-F:5’AGGATCCAACAATGGGCATCACCGTGACCAAC和IVA-R:5’CGAGCTCCTACTTGATGCTCACGTCGTAGAACTTC。这两个引物分别包含限制性内切酶位点BamH1和ScaI的位点。人工合成终止子序列ter(SEQIDNO:8),5’端和3’端分别设置有ScaI和KpnI位点。把用BamH1和ScaI酶切后的IPD072Aa-Vip3A和用ScaI和KpnI酶切后的终止子连入经BamH1和KpnI酶切的pCambia1300载体中,获得pCambia1300-IPD072Aa-Vip3A-ter。pUBI为玉米泛素蛋白启动子,通过PCR获得。以商业玉米品种郑单958的基因组DNA为模板,通过PCR扩增获得pUBI。PCR反应条件为:95℃3分钟;95℃15秒,68℃15秒,72℃2分钟,重复32个循环;然后72℃10分钟。将获得的大约2.0Kb的PCR产物克隆到T-载体pMD19中。获得序列正确的克隆用于下面的试验。PCR引物为pUBI-F(5’GAAGCTTGCATGCCTACAGTGCAGCGTGACCC)和pUBI-R(5’GGGTGGATCCTCTAGAGTCGACCTGCAGAAGTAAC),引物上分别设置了HindIII和BamHI限制性内切酶酶切位点。用HindIII和BamHI酶切pUBI,把酶切后的片段连入用相同酶切的pCambia1300-IPD072Aa-Vip3A-ter载体中,获得终载体。这个载体命名为:pCambia1300-pUBI-IPD072Aa-Vip3A。最后,通过电转的方法把上述T-DNA质粒转入农杆菌LBA4404中,通过含有15μg/ml四环素和50μg/mL的卡那霉素的YEP固体培养基筛选出阳性克隆,并保菌,用于接下来的植物转化。实施例6、IPD072Aa-Vip3H植物转化载体构建以IPD072Aa-Vip3H(SEQIDNO:3)作为模板经过PCR获得IPD072Aa-Vip3H基因片段。引物为:IVH-F:5’AGGATCCAACAATGGGCATCACCGTGACCAAC和IVH-R:5’CGAGCTCCTACTTGATGCTGAAGTCCCTGAAGG。这两个引物分别包含限制性内切酶位点BamH1和ScaI的位点。人工合成终止子序列ter(SEQIDNO:8),5’端和3’端分别设置有ScaI和KpnI位点。把用BamH1和ScaI酶切后的IPD072Aa-Vip3H和用ScaI和KpnI酶切后的终止子连入经BamH1和KpnI酶切的pCambia1300载体中,获得pCambia1300-IPD072Aa-Vip3H-ter。pUBI为玉米泛素蛋白启动子的获得方法同实施例5。用HindIII和BamHI酶切pUBI,把酶切后的片段连入用相同酶切的pCambia1300-IPD072Aa-Vip3H-ter载体中,获得终载体。这个载体命名为:pCambia1300-pUBI-IPD072Aa-Vip3H。最后,通过电转的方法把上述T-DNA质粒转入农杆菌LBA4404中,通过含有15μg/ml四环素和50μg/mL的卡那霉素的YEP固体培养基筛选出阳性克隆,并保菌,用于接下来的植物转化。实施例7、IPD072Aa-Vip3A和ICPs蛋白植物转化载体构建以Cry1Ab(SEQIDNO:7)杀虫蛋白的基因由上海生工合成,5’端和3’端分别设置有BamH1和ScaI位点。人工合成终止子序列ter(SEQIDNO:8),5’端和3’端分别设置ScaI和EcoRI位点。把用BamHI和ScaI酶切后的Cry1Ab和用ScaI和EcoRI酶切后的终止子连入经BamH1和EcoRI酶切的pCambia1300载体中,获得pCambia1300-Cry1Ab-ter。p35S启动子为花椰菜花叶病毒CaMV35S启动子,由上海生工合成,序列如SEQIDNO:9,5’端和3’端分别设置有KpnI和BamHI位点。用BamH1和EcoRI酶切pCambia1300-Cry1Ab-ter,用KpnI和BamHI酶切p35S,把上述两个片段和经KpnI和EcoRI酶切的载体pCambia1300-IPD072Aa-Vip3A-ter进行三段连接,获得终载体。这个载体命名为:pCambia1300-pUBI-IPD072Aa-Vip3A-p35S-Cry1Ab。最后,通过电转的方法把上述T-DNA质粒转入农杆菌LBA4404中,通过含有15μg/ml四环素和50μg/mL的卡那霉素的YEP固体培养基筛选出阳性克隆,并保菌,用于接下来的植物转化。实施例8、IPD072Aa-Vip3H和ICPs蛋白植物转化载体构建以Cry1Ab(SEQIDNO:7)杀虫蛋白的基因由上海生工合成,5’端和3’端分别设置有BamH1和ScaI位点。人工合成终止子序列ter(SEQIDNO:8),5’端和3’端分别设置ScaI和EcoRI位点。把用BamHI和ScaI酶切后的Cry1Ab和用ScaI和EcoRI酶切后的终止子连入经BamH1和EcoRI酶切的pCambia1300载体中,获得pCambia1300-Cry1Ab-ter。p35S启动子为花椰菜花叶病毒CaMV35S启动子,由上海生工合成,序列如SEQIDNO:9,5’端和3’端分别设置有KpnI和BamHI位点。用BamH1和EcoRI酶切pCambia1300-Cry1Ab-ter,用KpnI和BamHI酶切p35S,把上述两个片段和经KpnI和EcoRI酶切的载体pCambia1300-IPD072Aa-Vip3H-ter进行三段连接,获得终载体。这个载体命名为:pCambia1300-pUBI-IPD072Aa-Vip3H-p35S-Cry1Ab。最后,通过电转的方法把上述T-DNA质粒转入农杆菌LBA4404中,通过含有15μg/ml四环素和50μg/mL的卡那霉素的YEP固体培养基筛选出阳性克隆,并保菌,用于接下来的植物转化。实施例9、转基因玉米玉米的转化技术已经比较成熟。参考文献如:VladimirSidorov&DavidDuncan(inM.PaulScott(ed.),MethodsinMolecularBiology:TransgenicMaize,vol:526;YujiIshida,YukohHiei&ToshihikoKomari(2007)Agrobacterium-mediatedtransformationofmaize.NatureProtocols2:1614-1622。基本方法如下:取授粉后8-10天的Hi-II玉米穗,收集所有的未成熟胚(大小为1.0-1.5mm)。将实施例3中构建的含有T-DNA载体转化农杆菌获得的重组农杆菌与未成熟胚在共培养培养基上(MS+2mg/L2,4-D+30g/L蔗糖+3g/L琼脂(sigma7921)+40mg/L乙酰丁香酮)共培养2-3天(22℃)。转移未成熟胚到愈伤诱导培养基上(MS+2mg/L2,4-D+30g/L蔗糖+2.5g/Lgelrite+5mg/LAgNO3+200mg/L乙酰丁香酮),28℃暗培养10-14天。将所有的愈伤转到带有2mM草甘膦的筛选培养基(与愈伤诱导培养基相同)上,28℃暗培养2-3周。转移所有的组织到新鲜含2mM草甘膦的筛选培养基上,28℃暗培养2-3周。然后,转移所有筛选后成活的胚性组织到再生培养基(MS+30g/L蔗糖+0.5mg/Lkinetin+2.5g/Lgelrite+200mg/L乙酰丁香酮)上,28℃暗培养10-14天,每皿一个株系。转移胚性组织到新鲜的再生培养基上,26℃光照培养10-14天。转移所有发育完全的植株到生根培养基(1/2MS+20g/L蔗糖+2.5g/Lgelrite+200mg/L乙酰丁香酮)上,26℃光照培养直到根发育完全。获得含上述转化载体的转基因玉米植株。实施例10、转基因玉米可以杀虫将实施例9制备的转基因玉米植株的T0代植株移栽到温室中,用商业化品种“郑丹958”的母本,“郑58”(Z58)的花粉进行授粉,收获T0代种子。然后将这些品系与商业化品种“郑丹958”的母本,“郑58”(Z58)进行回交转育,获得Z58近等位基因系。再对这些近等位基因系的除草剂抗性进行比较分析。我们对获得的98个转pCambia1300-IPD072Aa-Vip3A-ter载体的转基因株系(命名为IV3A)和105个转pCambia1300-IPD072Aa-Vip3H-ter载体的转基因株系(命名为IV3H)进行抗虫性测定,其杀虫效果效果如表2所示:表2*:*注:表2为对播种后30天的玉米叶片提取物进行不同害虫的杀虫效果检测。每个实验都至少设置3个重复。对我们获得的69个转pCambia1300-IPD072Aa-Vip3A-p35S-Cry1Ab载体的转基因株系(命名为IV3A1Ab)和73个转pCambia1300-IPD072Aa-Vip3H-p35S-Cry1Ab载体的转基因株系(命名为IV3H1Ab)进行抗虫性测定,其杀虫效果效果如表3所示:表3*:害虫杀虫率玉米螟100%棉铃虫100%粘虫100%甜菜夜蛾100%西方玉米根虫100%*注:表3为对播种后30天的玉米叶片提取物进行不同害虫的杀虫效果检测。每个实验都至少设置3个重复。最后,还需要注意的是,以上列举的仅是本发明的具体实施例。显然,本发明不限于以上实施例,还可以有许多变形。本领域的普通技术人员能从本发明公开的内容直接导出或联想到的所有变形,均应认为是本发明的保护范围。SEQUENCELISTING<110>浙江大学<120>一种杀虫融合蛋白、编码基因及其应用<130><160>9<170>PatentInversion3.5<210>1<211>2652<212>DNA<213>unknown<220><223>人工序列<400>1atgggcatcaccgtgaccaacaacagcagcaacccgatcgaggtggccatcaaccactgg60ggcagcgacggcgacaccagcttcttcagcgtgggcaacggcaagcaggagacctgggac120aggagcgacagcaggggcttcgtgctgagcctgaagaagaacggcgcccagcacccgtac180tacgtgcaggccagcagcaagatcgaggtggacaacaacgccgtgaaggaccagggcagg240ctgatcgagccgctgagccgaggccccgggaagggtggaggaatgaacaagaacaacacc300aagctgagcaccagggccctgccgagcttcatcgactacttcaacggcatctacggcttc360gccaccggcatcaaggacatcatgaacatgatcttcaagaccgacaccggcggcgacctg420accctggacgagatcctgaagaaccagcagctgctgaacgacatcagcggcaagctggac480ggcgtgaacggcagcctgaacgacctgatcgcccagggcaacctgaacaccgagctgagc540aaggagatcctgaagatcgccaacgagcagaaccaggtgctgaacgacgtgaacaacaag600ctggacgccatcaacaccatgctgagggtgtacctgccgaagatcaccagcatgctgagc660gacgt
