一种抗虫融合基因、编码蛋白及其应用的制作方法与工艺
本发明涉及一种抗虫融合基因、该抗虫融合基因编码的融合蛋白,及上述融合蛋白的应用。(二)
背景技术
:害虫给全球农业生产带来巨大的损失,目前害虫防治主要依靠化学农药,但是农药的使用加重了生产成本,并且农药残留对人体健康带来严重危害。因此,利用基因工程方法防治害虫具有重大经济、环境和社会价值。获得转基因抗虫作物的关键是克隆优良杀虫蛋白质。杀虫蛋白质有很多,使用得最为广泛的是苏云金芽孢杆菌(Bacillusthuringiensis,简称Bt)在产胞期间所分泌一种伴胞晶体蛋白(insecticidalcrystalproteins,ICP),如Cry1Ab,Cry1C等,这些蛋白对鳞翅目、双翅目、鞘翅目等昆虫(比如小菜蛾、玉米螟)有很强的杀伤作用(Schnepf,E.,Crickmore,N.,Van,R.J.,Lereclus,D.,Baum,J.,&Feitelson,J.,etal.1998,62(3),775-806.),目前已被深入研究和广泛使用,玉米、大豆、土豆、棉花等转Bt基因作物已得到大规模商业化种植。苏云金芽胞杆菌在营养生长期间,还会分泌一种与ICPs无氨基酸序列同源性、杀虫机理完全不同的蛋白,即营养期杀虫蛋白(vegetativeinsecticidalproteins,VIPs),如Vip3A、Vip3B、Vip3C、Vip3D、Vip3H,对一些对ICPs不敏感的害虫也有杀虫活性,并且不会发生交叉抗性(Estruch,J.J.,Warren,G.W.,Mullins,M.A.,Nye,G.J.,Craig,J.A.,&Koziel,M.G.1996,ProceedingsoftheNationalAcademyofSciencesoftheUnitedStatesofAmerica,93(11),5389-94.)。Vip3蛋白作为一种新型的杀虫蛋白,对目前使用最为广泛的ICP蛋白从杀虫机制、杀虫谱到杀虫活性上都是一个很好的补充。比如,ICPs对斜纹夜蛾和粘虫的杀伤作用很弱,Vip3则对斜纹夜蛾和粘虫有非常好的防治效果。尽管如此,在转基因作物研发的实际操作中,仍然存在一些急需解决的技术性问题。例如,研究人员发现高表达单独的Vip3基因的植株生长比较迟缓,容易出现白化现象。这可能是由于转基因植物中直接表达Vip3蛋白后,该蛋白由于信号肽的分泌作用大量聚集并结合在植物细胞膜上并形成孔道,从而对细胞造成损坏,影响其生长发育。半翅目害虫(如粉虱、飞虱)通过吮吸韧皮部汁液导致植物枯萎、落叶、发育迟缓、减产甚至绝收,对水稻、棉花等粮食和经济作物造成很大的危害。Bt抗虫基因对鳞翅目、鞘翅目等害虫有很好的杀伤作用。但是,目前还没有发现任何Bt蛋白对半翅目害虫有很好防治作用。最近,有科学家从蕨类植物中发现了一种对半翅目害虫具有很好的防治作用的蛋白Tma12,该蛋白是一种具有几丁质酶活性的几丁质结合蛋白(ShuklaAK,UpadhyaySK,MishraM,etal.2016,NatureBiotechnology。第一代转基因作物大多只具有抗鳞翅目害虫的特性,而第二代转基因作物正在向同时抗多种害虫的复合性状发展。获得具有复合性状的转基因作物有多种方法,比如通过对具有单一性状的转基因作物的杂交;通过将多个基因表达框构建在同一个表达框里面;通过共转化,将多个含有单基因的农杆菌混在一起,进行混合转化,筛选出多个基因同时整合了两个或多个质粒T-DNA的转基因植株。但是上述方法在实际操作和应用中都还存在一些问题。因此,获得更为简单高效的获得具有复合性状的转基因作物的方法是当下需要切实解决的问题。(三)技术实现要素:本发明目的是提供一种通过基因融合使得同一个融合蛋白同时对鳞翅目和半翅目具有杀伤效果的方法。本发明采用的技术方案是:本发明提供一种抗虫融合基因,所述的基因从5’-3’依次包括编码几丁质结合蛋白的核苷酸序列和编码Vip3毒素的核苷酸序列;且上述2个核苷酸序列位于同一个开放阅读框内。进一步,优选所述的基因从5’-3’依次由编码几丁质结合蛋白的核苷酸序列和编码Vip3毒素的核苷酸序列连接组成。进一步,所述几丁质结合蛋白为Tma12,所述Vip3毒素为Vip3A、Vip3B、Vip3C、Vip3D或Vip3H。更进一步,所述Vip3毒素为Vip3A或Vip3H。进一步,所述抗虫融合基因的核苷酸序列为SEQIDNO:1(几丁质结合蛋白为Tma12,Vip毒素为Vip3A)或SEQIDNO:2所示(几丁质结合蛋白为Tma12,Vip毒素为Vip3H)。本发明还提供一种所述抗虫融合基因编码的融合蛋白,该融合蛋白从N端至C端依次为几丁质结合蛋白和Vip3毒素。进一步,所述融合蛋白的氨基酸序列为SEQIDNO:3(几丁质结合蛋白为Tma12,Vip3毒素为Vip3A)或SEQIDNO:4(几丁质结合蛋白为Tma12,Vip毒素为Vip3H)所示。本发明还提供一种所述的融合蛋白在制备转基因抗虫作物、转基因杀虫微生物或该融合蛋白的抗体中的应用。进一步,所述转基因抗虫作物是由融合蛋白联合BT晶体毒素转化制备而成。与现有技术相比,本发明有益效果主要体现在:本发明所设计的用一个几丁质结合蛋白与一个Vip3毒素融合成的人工蛋白质分子,与原先的几丁质结合蛋白和Vip3蛋白相比,具有如下优点:杀虫谱广,能同时防治鳞翅目和半翅目中多种重要害虫(如甜菜夜蛾、粘虫、粉虱),杀虫效率高达50%-100%;有利于具有上述不同功能的蛋白和其它抗虫蛋白(如ICPs)联合使用,进一步扩大杀虫谱,延缓害虫抗性产生。(四)附图说明图1是本发明的融合杀虫蛋白的结构图。融合蛋白中N端为几丁质结合蛋白,C端为Vip3毒素。图2是本发明中把杀虫融合蛋白基因导入植物中的T-DNA结构图。pUBI为玉米泛素启动子,融合蛋白基因为几丁质结合蛋白-Vip3毒素融合基因。图3是本发明中把杀虫融合蛋白和ICPs联合导入植物中的T-DNA结构图。pUBI为玉米泛素启动子,融合蛋白基因为几丁质结合蛋白-Vip3毒素融合基因,p35S为花椰菜花叶病毒35S启动子,ICPs为伴胞晶体蛋白编码基因。(五)具体实施方式下面结合具体实施例对本发明进行进一步描述,但本发明的保护范围并不仅限于此:实施例1、Tma12-Vip3A融合蛋白表达载体的构建Tma12和Vip3A杀虫蛋白的基因均由上海生工合成,其DNA序列分别为SEQIDNO:5和中国专利200610049611.0中的SEQIDNO:7,并克隆在pET28a表达载体的限制性内切酶BamHI和SacI位点之间。构建好的载体分别命名为pET28a-Tma12和pET28a-Vip3A。Tma12-Vip3A合成具体步骤如下:1、以Vip3A(中国专利200610049611.0中的SEQIDNO:7)作为模板经过PCR获得Vip3A基因片段。2、引物为:Vip3A-F:5’CCCGGGAAGGGTGGAGGAATGAACAAGAACAACACCAAG和Vip3A-R:5’CGAGCTCCTACTTGATGCTCACGTCGTAGAACTTCACGA。这两个引物分别包含限制性内切酶位点XmalI和ScaI的位点。3、用XmalI和ScaI处理PCR产物,并与用相同的酶处理过的pET28a-Tma12载体连接。4、转入大肠杆菌,获得质粒为pET28a-Tma12-Vip3A。融合蛋白基因Tma12-Vip3A(SEQIDNO:1)被克隆到了pET28a表达载体中,融合蛋白基因编码的融合蛋白Tma12-Vip3A(SEQIDNO:2)分子量大约为116kD。实施例2、Tma12-Vip3H融合蛋白表达载体的构建Tma12和Vip3H杀虫蛋白的基因均由上海生工合成,其DNA序列分别为SEQIDNO:5和SEQIDNO:6,并克隆在pET28a表达载体的限制性内切酶BamHI和SacI位点之间。构建好的载体分别命名为pET28a-Tma12和pET28a-Vip3H。Tma12-Vip3H合成具体步骤如下:1、以Vip3H(SEQIDNO:6)作为模板经过PCR获得Vip3H基因片段。引物为:Vip3H-F:5’CCCGGGAAGGGTGGAGGAATGAACAAGAACAACAGTAAG和Vip3H-R:5’CGAGCTCCTACTTGATGCTGAAGTCCCTGAAGGTGATGT。这两个引物分别包含限制性内切酶位点XmalI和ScaI的位点。2、用XmalI和ScaI处理PCR产物,并与用相同的酶处理过的pET28a-Tma12载体连接。3、转入大肠杆菌,获得质粒为pET28a-Tma12-Vip3H。融合蛋白基因Tma12-Vip3H(SEQIDNO:3)被克隆到了pET28a表达载体中,融合蛋白基因编码的融合蛋白Tma12-Vip3H(SEQIDNO:4)分子量大约为112kD。实施例3、杀虫蛋白的表达通用标准方法将上述含融合蛋白基因的表达载体(pET28a-Tma12-Vip3A、pET28a-Tma12-Vip3H、pET28a-Tma12、pET28a-Vip3A和pET28a-Vip3H)分别转入大肠杆菌BL21star。挑取单克隆菌落接种到100mlLB细菌培养液中,37℃震动培养至OD600=0.6,加入IPTG到0.5mM,继续在相同条件下培育4小时。收集培养液5000g离心10分钟沉淀大肠杆菌细胞,然后弃上清收集沉淀。沉淀中加30毫升20mMTris-HCL缓冲液,超声粉碎后即可用于杀虫活性的测定。实施例4、Tma12-Vip3A和Tma12-Vip3H对多种鳞翅目和半翅目害虫具有杀伤作用实施例3所获得的杀虫蛋白(Tma12-Vip3A、Tma12-Vip3H、Tma12、Vip3A和Vip3H)分别涂在昆虫人工饲料的表面,新生一龄幼虫用来进行杀虫性测定。阴性对照为pET28a空载体,准备方法与实施例3相同。杀虫活性结果如表1所示:表1、融合蛋白的杀虫效率粘虫甜菜夜蛾烟粉虱Tma120%0%100%Vip3A100%100%0%Vip3H100%100%0%Tma12-Vip3A100%100%100%Tma12-Vip3H100%100%100%阴性对照0%0%0%实施例5、Tma12-Vip3A植物转化载体构建以Tma12-Vip3A(SEQIDNO:1)作为模板经过PCR获得Tma12-Vip3A基因片段。引物为:TVA-F:5’AGGATCCAACAATGGGCAGGAGCTGGGGCGT和TVA-R:5’CGAGCTCCTACTTGATGCTCACGTCGTAGAACTTC。这两个引物分别包含限制性内切酶位点BamH1和ScaI的位点。人工合成终止子序列ter(SEQIDNO:8),5’端和3’端分别设置有ScaI和KpnI位点。把用BamH1和ScaI酶切后的Tma12-Vip3A和用ScaI和KpnI酶切后的终止子连入经BamH1和KpnI酶切的pCambia1300载体中,获得pCambia1300-Tma12-Vip3A-ter。pUBI为玉米泛素蛋白启动子,通过PCR获得。以商业玉米品种郑单958的基因组DNA为模板,通过PCR扩增获得pUBI。PCR反应条件为:95℃3分钟;95℃15秒,68℃15秒,72℃2分钟,重复32个循环;然后72℃10分钟。将获得的大约2.0Kb的PCR产物克隆到T-载体pMD19中。获得序列正确的克隆用于下面的试验。PCR引物为pUBI-F(5’GAAGCTTGCATGCCTACAGTGCAGCGTGACCC)和pUBI-R(5’GGGTGGATCCTCTAGAGTCGACCTGCAGAAGTAAC),引物上分别设置了HindIII和BamHI限制性内切酶酶切位点。用HindIII和BamHI酶切pUBI,把酶切后的片段连入用相同酶切的pCambia1300-Tma12-Vip3A-ter载体中,获得终载体。这个载体命名为:pCambia1300-pUBI-Tma12-Vip3A。最后,通过电转的方法把上述T-DNA质粒转入农杆菌LBA4404中,通过含有15μg/ml四环素和50μg/mL的卡那霉素的YEP固体培养基筛选出阳性克隆,并保菌,用于接下来的植物转化。实施例6、Tma12-Vip3H植物转化载体构建以Tma12-Vip3H(SEQIDNO:2)作为模板进过PCR获得Tma12-Vip3H基因片段。引物为:TVH-F:5’GGATCCAACAATGGGCAGGAGCTGGGGCGT和TVH-R:5’CGAGCTCCTACTTGATGCTGAAGTCCCTGAAGG。这两个引物分别包含限制性内切酶位点BamH1和ScaI的位点。人工合成终止子序列ter(SEQIDNO:8),5’端和3’端分别设置有ScaI和KpnI位点。把用BamH1和ScaI酶切后的Tma12-Vip3H和用ScaI和KpnI酶切后的终止子连入经BamH1和KpnI酶切的pCambia1300载体中,获得pCambia1300-Tma12-Vip3H-ter。pUBI为玉米泛素蛋白启动子的获得方法同实施例5。用HindIII和BamHI酶切pUBI,把酶切后的片段连入用相同酶切的pCambia1300-Tma12-Vip3H-ter载体中,获得终载体。这个载体命名为:pCambia1300-pUBI-Tma12-Vip3H。最后,通过电转的方法把上述T-DNA质粒转入农杆菌LBA4404中,通过含有15μg/ml四环素和50μg/mL的卡那霉素的YEP固体培养基筛选出阳性克隆,并保菌,用于接下来的植物转化。实施例7、Tma12-Vip3A和ICPs蛋白植物转化载体构建以Cry1Ab(SEQIDNO:7)杀虫蛋白的基因由上海生工合成,5’端和3’端分别设置有BamH1和ScaI位点。人工合成终止子序列ter(SEQIDNO:8),5’端和3’端分别设置ScaI和EcoRI位点。把用BamHI和ScaI酶切后的Cry1Ab和用ScaI和EcoRI酶切后的终止子连入经BamH1和EcoRI酶切的pCambia1300载体中,获得pCambia1300-Cry1Ab-ter。p35S启动子为花椰菜花叶病毒CaMV35S启动子,由上海生工合成,序列如SEQIDNO:9,5’端和3’端分别设置有KpnI和BamHI位点。用BamH1和EcoRI酶切pCambia1300-Cry1Ab-ter,用KpnI和BamHI酶切p35S,把上述两个片段和经KpnI和EcoRI酶切的载体pCambia1300-Tma12-Vip3A-ter进行三段连接,获得终载体。这个载体命名为:pCambia1300-pUBI-Tma12-Vip3A-p35S-Cry1Ab。最后,通过电转的方法把上述T-DNA质粒转入农杆菌LBA4404中,通过含有15μg/ml四环素和50μg/mL的卡那霉素的YEP固体培养基筛选出阳性克隆,并保菌,用于接下来的植物转化。实施例8、Tma12-Vip3H基因和BT晶体毒素基因(ICPs蛋白)植物转化载体构建以Cry1Ab(SEQIDNO:7)杀虫蛋白的基因由上海生工合成,5’端和3’端分别设置有BamH1和ScaI位点。人工合成终止子序列ter(SEQIDNO:8),5’端和3’端分别设置ScaI和EcoRI位点。把用BamHI和ScaI酶切后的Cry1Ab和用ScaI和EcoRI酶切后的终止子连入经BamH1和EcoRI酶切的pCambia1300载体中,获得pCambia1300-Cry1Ab-ter。p35S启动子为花椰菜花叶病毒CaMV35S启动子,由上海生工合成,序列如SEQIDNO:9,5’端和3’端分别设置有KpnI和BamHI位点。用BamH1和EcoRI酶切pCambia1300-Cry1Ab-ter,用KpnI和BamHI酶切p35S,把上述两个片段和经KpnI和EcoRI酶切的载体pCambia1300-Tma12-Vip3H-ter进行三段连接,获得终载体。这个载体命名为:pCambia1300-pUBI-Tma12-Vip3H-p35S-Cry1Ab。最后,通过电转的方法把上述T-DNA质粒转入农杆菌LBA4404中,通过含有15μg/ml四环素和50μg/mL的卡那霉素的YEP固体培养基筛选出阳性克隆,并保菌,用于接下来的植物转化。实施例9、转基因水稻转基因水稻的获得方法是采用现有技术(卢雄斌,龚祖埙(1998)生命科学10:125-131;刘凡等(2003)分子植物育种1:108-115)。选取成熟饱满的“秀水-134”种子去壳,诱导产生愈伤组织作为转化材料。分别取实施例5-8中构建的载体和pCambia1300空载体农杆菌划板。挑单菌落接种,准备转化用农杆菌。将待转化的愈伤组织放入OD为0.6左右的农杆菌菌液中(农杆菌菌液的制备:将农杆菌接种至培养基,培养至OD为0.6左右;培养基组成:3g/LK2HPO4、1g/LNaH2PO4、1g/LNH4Cl、0.3g/LMgSO4·7H2O、0.15g/LKCl、0.01g/LCaCl2、0.0025g/LFeSO4·7H2O、5g/L蔗糖、20mg/L乙酰丁香酮,溶剂为水,pH=5.8),让农杆菌结合到愈伤组织表面,然后把愈伤组织转移到共培养基(MS+2mg/L2,4-D+30g/L葡萄糖+30g/L蔗糖+3g/L琼脂(sigma7921)+20mg/L乙酰丁香酮)中,共培养2-3天。用无菌水冲洗转化后的愈伤,转移到筛选培养基(MS+2mg/L2,4-D+30g/L蔗糖+3g/L琼脂(sigma7921)+20mg/L乙酰丁香酮+2mM草甘膦(Sigma))上,筛选培养两个月(中间继代一次)。把筛选后,生长活力良好的愈伤转移到预分化培养基(MS+0.1g/L肌醇+5mg/LABA+1mg/LNAA+5mg/L6-BA+20g/L山梨醇+30g/L蔗糖+2.5g/Lgelrite)上培养20天左右,然后将预分化好的愈伤组织移到分化培养基上,每天14小时光照分化发芽。2-3周后,把抗性再生植株转移到生根培养基(1/2MS+0.2mg/LNAA+20g/L蔗糖+2.5g/Lgelrite)上壮苗生根,最后将再生植株洗去琼脂移植于温室,选择产量高、种子大或者生物量高等能够提高水稻产量的转基因株系,培育新品种。分别获得含上述转化载体和只含有筛选标记基因EPSPS的空载体的转基因水稻植株。实施例10、转基因水稻可以杀虫将实施例9方法制备的转基因水稻植株的T0代植株移栽到温室中,对转基因水稻植株和非转基因受体对照植株的杀虫性能进行比较分析。我们对获得的102个转pCambia1300-Tma12-Vip3A-ter载体的转基因株系(命名为TV3A)和83个转pCambia1300-Tma12-Vip3H-ter载体的转基因株系(命名为TV3H)进行抗虫性测定,其杀虫效果如表2所示:表2:对播种后30天的水稻叶片提取物进行不同害虫的杀虫效果检测害虫杀虫率粘虫100%甜菜夜蛾100%烟粉虱100%*注:每个实验都至少设置3个重复。对我们获得的137个转pCambia1300-Tma12-Vip3A-p35S-Cry1Ab载体的转基因株系(命名为TV3A1Ab)和142个转pCambia1300-Tma12-Vip3H-p35S-Cry1Ab载体的转基因株系(命名为TV3H1Ab)进行抗虫性测定,其杀虫效果如表3所示:表3:对播种后30天的水稻叶片提取物进行不同害虫的杀虫效果检测害虫杀虫率玉米螟100%棉铃虫100%粘虫100%甜菜夜蛾100%烟粉虱100%*注:每个实验都至少设置3个重复。实施例11、转基因大豆这里使用的获得转基因大豆的步骤来自于已有的技术(Dengetal.,1998,PlantPhysiologyCommunications34:381-387;Maetal.,2008,ScientiaAgriculturaSinica41:661-668;Zhouetal.,2001,JournalofNortheastAgriculturalUniversity32:313-319)。选取健康、饱满、成熟的大豆,用80%乙醇消毒2分钟,再用无菌水清洗,然后放置在充满氯气(由50mlNaClO与2ml浓HCl反应生成)的干燥器中灭菌4-6个小时。灭菌后的大豆在超净工作台里被播撒到B5培养基中,25℃条件下培养5天,同时光密度在90-150μmol光子/m2·s水平。当子叶变绿并顶破种皮,无菌的豆芽就会长出。去掉了下胚轴的豆芽在长度上被切成五五开,使得两片外植体都具有子叶和上胚轴。在子叶和上胚轴的节点处切外植体大约7-8处,即可用作被侵染的目标组织。分别取实施例5-8中构建的载体和pCambia1300空载体的农杆菌被分开培养待用。准备好的外植体浸没在农杆菌悬浮液中共培养30分钟左右。然后,将侵染的组织上多余的细胞悬浮液用吸水纸吸收干净,再转移到1/10B5共培养基里,25℃暗培养3-5天。共培养的植物组织用B5液体培养基清洗,以除去多余的农杆菌,然后放置到B5固体培养基中25℃下培养5天,待其发芽。诱导发生的胚芽组织转移到含有0.1-0.5mM草甘膦的B5筛选培养基中,25℃光照培养4周,期间每两周更换一次培养基。筛选出来的胚芽组织再转移到固体培养基中,25℃培养,待其长成小苗。随后,将转基因植株苗转移到1/2B5培养基中进行生根诱导。最后,长成的小植株经清洗去除琼脂后栽种在温室中。实施例12、转基因大豆可以杀虫将实施例11方法制备的转基因水稻植株的T0代植株移栽到温室中,对转基因水稻植株和非转基因受体对照植株的杀虫性能进行比较分析。我们对获得的76个转pCambia1300-Tma12-Vip3A-ter载体的转基因株系(命名为TV3A)和68个转pCambia1300-Tma12-Vip3H-ter载体的转基因株系(命名为TV3H)进行抗虫性测定,其杀虫效果效果如表4所示:表4:对播种后24天的大豆叶片提取物进行不同害虫的杀虫效果检测害虫杀虫率粘虫100%甜菜夜蛾100%烟粉虱100%*注:每个实验都至少设置3个重复。对我们获得的62个转pCambia1300-Tma12-Vip3A-p35S-Cry1Ab载体的转基因株系(命名为TV3A1Ab)和77个转pCambia1300-Tma12-Vip3H-p35S-Cry1Ab载体的转基因株系(命名为TV3H1Ab)进行抗虫性测定,其杀虫效果如表5所示:表5:对播种后24天的大豆叶片提取物进行不同害虫的杀虫效果检测害虫杀虫率玉米螟100%棉铃虫100%粘虫100%甜菜夜蛾100%烟粉虱100%*注:每个实验都至少设置3个重复。最后,还需要注意的是,以上列举的仅是本发明的具体实施例。显然,本发明不限于以上实施例,还可以有许多变形。本领域的普通技术人员能从本发明公开的内容直接导出或联想到的所有变形,均应认为是本发明的保护范围。SEQUENCELISTING<110>浙江大学<120>一种抗虫融合基因、编码蛋白及其应用<130><160>9<170>PatentInversion3.5<210>1<211>3042<212>DNA<213>unknown<220><223>人工序列<400>1atgggcaggagctggggcgtggtggccgtgatggtgctgtgcgccagcggcctgctgggc60atcgtgaggggccacggcagcatggaggacccgatcagcagggtgtacaggtgcaggctg120gagaacccggagaggccgaccagcccggcctgccaggctgccgtggccctgagcggcacc180caggccttctacgactggaacgaggtgaacatcccgaacgctgccggcaggcacagggag240ctgatcccggacggccagctgtgcagcgccggcaggttcaagttcaggggcctggacctg300gccaggagcgactggatcgccaccccgctgccgagcggag
