本发明涉及生物技术领域,更具体地,公开了一种新型的含mar核心片段的动物细胞表达载体。
背景技术:
目前,很多用于生产重组蛋白的载体已被开发,重组蛋白在以细菌、真核微生物、昆虫细胞等为宿主的表达中能获得较高的产量,但这些表达系统缺少类似于哺乳动物细胞的蛋白质修饰机制(例如糖基化修饰等),表达的动物蛋白易缺少生物活性或因折叠错误而产生包涵体。而以哺乳动物细胞表达系统来生产重组蛋白,其产量往往较低,而且导入基因的稳定性也会有一定的问题。后来人们发现,中国仓鼠卵巢细胞(cho)是生产单克隆抗体等复杂大分子的最佳系统,利用cho表达系统生产单克隆抗体,其产率有了很大的提高,但是筛选和维持高表达细胞株仍是件耗时耗力且结果不确定的工作。细胞转染后,融合基因通常随机整合入宿主细胞的基因组中,大多数的重组发生于“静默区”,例如非转录区。通常需要筛选上千个克隆才能得到几个产量尚可的细胞株。
核基质附着区(matrixattachmentregion,简称mar)元件是一种能帮助产生并维持开放的染色质域的dna序列。开放的染色质域能促进转录,表达及转基因的多拷贝整合。将mar元件放入载体中能增加细胞株的产量及高产细胞株的比例(kwaksth,otteap2006trendsbiotechnol24:137–142),但mar元件较大(3kb以上),而质粒转染细胞的效率随着质粒大小的增加而降低(yinwetalanalbiochem.2005nov15;346(2):289-94),从而限制了mar元件在双亚基共表达载体中的应用。
因此,开发一种更为有效的哺乳动物细胞表达载体,一直是本领域的技术人员急待解决的问题。
技术实现要素:
本发明的目的是提供一种可以提高导入基因在哺乳动物细胞中表达的表达载体,所述表达载体两端含有人工转录因子结合位点的核基质附着区的mar核心片段。该表达载体可有效提高蛋白在哺乳动物细胞中的产量并降低生产成本。
具体地,发明公开了:
1、一种分离的重组表达载体的mar核心片段,其核苷酸碱基序列如seqidno:1、seqidno:2或seqidno:3所示。
2、一种重组表达载体,所述载体两端含有上述1所述的mar核心片段。
3、上述2所述载体,其所述mar核心片段插入在启动子的上游及聚腺苷酸化位点的下游。
4、上述3所述载体,所述启动子为cmv、sv40或ef-1a启动子。优选地,所述mar核心片段插在载体上游fspi位点,或sv40pa下游afei位点,或pef-1a上游nhei位点,或bghpa下游ecorv位点。最优选上游fspi位点及pef-1a上游nhei位点。
5、上述2-4任一所述载体,所述载体为pbudce4.1、pdr1、pcdna3.1(+)、pdhff或pcho1.0,或者上述载体的改造载体。
6、上述5所述载体,所述载体为pbudce4.1改造的载体,所述载体核苷酸序列如seqidno:4、seqidno:5或seqidno:6所示。
7、上述2-6任一所述载体,其中所述载体包含一个或多个编码一种或多种重组蛋白的功能重组基因。
8、一种宿主细胞,其包含上述7所述的载体。
9、上述8所述的细胞,其为哺乳动物细胞,包括但不限于选自以下所述的组:中国仓鼠卵巢(cho)细胞、cos细胞、vero细胞、sp2/0细胞、ns/0骨髓瘤细胞、人类胚肾(hek293)细胞、幼仓鼠肾(bhk)细胞、hela细胞、人类b细胞、cv-1/ebna细胞、l细胞、3t3细胞、hepg2细胞、perc6细胞和mdck细胞。
10、一种制备重组蛋白的方法,所述方法包括转录和翻译存在于上述任一所述表达载体上的一个或多个编码所述重组蛋白的基因。
本发明的表达载体尤其适合应用于哺乳动物细胞,特别是中国仓鼠卵巢细胞cho的表达。
本发明的两端加有人工转录因子结合位点的mar核心片段dna序列可插入哺乳动物细胞表达载体,从而大大提高导入的哺乳动物基因在哺乳动物细胞中表达,且稳定性很好。
本发明提供了一种哺乳动物细胞表达载体,所述表达载体含有两端加有人工转录因子结合位点的mar核心片段dna序列.如本发明实例列举的,所述表达载体为改造的pbudce4.1载体,所述改造的pbudce4.1载体,在sv40聚腺苷酸化位点及bgh聚腺苷酸化位点下游分别插入afei和ecorv限制酶切位点。用kanamycin抗性基因及dhfr基因替换pbudce4.1中的zeocin抗性基因。所述改造载体命名为pbcm1.1,pbcm1.2,pbcm1.3,其dna序列分别为seqidno.4,5,6所示。不含上述核基质结合区元件片段及人工转录因子结合位点dna的改造载体命名为pbcm1.0,dna序列如seqidno7所示。
本发明的两端加有人工转录因子结合位点的mar核心片段dna序列可用常规的合成方法制备。
将本发明的两端加有人工转录因子结合位点的mar核心片段的质粒与不含两端加有人工转录因子结合位点的mar核心片段的质粒做比较。通过测定外源蛋白在上述两种质粒的表达量,结果表明,本发明的两端加有人工转录因子结合位点的mar核心片段的质粒特别适用于cho细胞,与不含两端加有人工转录因子结合位点的核基质结合区元件mar核心片段的质粒相比,动物细胞中的外源蛋白的表达量获得大幅提高,表达量提高了10-17倍。高产细胞株的比例提高了25-40倍。
将外源蛋白(例如治疗性蛋白、抗体)的基因插入本发明的表达载体中,然后转染至哺乳动物细胞,可用于外源蛋白(例如治疗性蛋白、抗体)的生产。

具体实施方式
以下实施例、实验例是对本发明进行进一步的说明,不应理解为对本发明的限制。
本发明实施例所列举的表达载体,是由pbudce4.1载体(购自:lifetechnologies)改造获得:包括在sv40聚腺苷酸化位点及bgh聚腺苷酸化位点下游分别插入afei和ecorv限制酶切位点;将卡那霉素抗性基因(kanamycinresistancegene),pgk启动子,二氢叶酸还原酶(dhfr),及sv40pa从pcho1.0(购自lifetechnologies)中pcr出,并通过nhei和afei位点装入pbudce4.1中,并同时去除zeocin抗性基因;将氯霉素乙酰转移酶基因(chloramphenicolacetyltransferasegene,cat基因)从pbudce4.1/lacz/cat(购自lifetechnologies)中pcr出,并通过kpni和bglii位点装入pbudce4.1中;将绿色荧光蛋白基因(greenfluorescentproteingene,gfp基因)从pcdna3.1/ct-gfp-top9(购自lifetechnologies)中pcr出,并通过psti和bamhi位点装入pbudce4.1。
实施例1pbudce4.1中引入afei和ecorv限制酶切位点
通过点突变的方法将afei和ecorv限制酶切位点引入sv40聚腺苷酸化位点及bgh聚腺苷酸化位点下游。用quickchange点突变试剂盒(购自安捷伦科技公司)进行点突变。引入afei酶切位点。
正向引物:5’-ggaaaacgattccgaagcgctac-3’
反向引物:5’-ccttctatgaaaggtagcgcttcg-3’
定点突变pcr扩增条件如下:1、95℃30秒;2、95℃30秒;3、55℃1分钟;4、68℃5分钟;5、2-4重复18循环。
加入dpni酶将模板质粒消化,然后转化xl1-blue感受态大肠杆菌。具体步骤如下:1.新鲜制备的或-20℃下保存的100ul感受态细胞(购自安捷伦科技),置于冰上,完全解冰后轻轻地将细胞均匀悬浮。2.加入dpni酶(购自:neb公司)消化后的质粒,轻轻混匀。3.冰上放置30分钟。4.42℃水浴热激60秒。5.冰上放置2分钟。6.加400ullb培养液(购自sigma-aldrich),37℃250转/分振荡培养30分钟。7、将细菌涂布在amp/lb琼脂平板上8.平皿在37℃下正向放置1小时,待接种的液体吸收进琼脂后,将平皿倒置,培养过夜。9、经37℃培养过夜的、在氨苄青霉素/lb琼脂平板上出现的菌落即为质粒转化的大肠杆菌,10、挑取单克隆菌并抽提质粒。得到突变的质粒后,用afei酶切鉴定正确并送上海生工生物工程有限公司测序,结果正确。
然后在已引入afei的载体中引入ecorv酶切位点:

定点突变pcr扩增条件如下:1、95℃30秒。2、95℃30秒;3、55℃1分钟;4、68℃5分钟;5、2-4重复18循环
加入dpni酶将模板质粒消化,然后转化xl1-blue感受态大肠杆菌。得到突变的质粒后用ecorv酶切鉴定正确并送上海生工生物工程有限公司测序,结果正确。
实施例2pbudce4.1中去除zeocin抗性基因并加入卡那霉素抗性基因,pgk启动子,dhfr及sv40pa
将卡那霉素抗性基因、pgk启动子、dhfr及sv40pa从pcho1.0(购自lifetechnologies)中pcr出,并通过nhei和afei位点装入实施例1所获得的pbudce4.1,同时去除zeocin抗性基因。

反向引物:5'-gtggctagctaagatacattgatgagtttg-3'
pcr用primestarhs高保真dna聚合酶(购自takara公司),
具体步骤如下:1、95℃3分钟;2、98℃10秒;3、47℃15秒;4、72℃3分钟;5、2-4重复30循环;6、72℃5分钟。
改造后的新载体命名为pbcm1.0,其结构图见附图5,其中,nhei、afei、fspi及ecorv位点为两端加有人工转录因子结合位点的mar核心片段的插入位点。
实施例3构建含有两端加有人工转录因子结合位点的mar核心片段的表达载体两端加有人工转录因子结合位点的mar核心片段(序列如seqidno:1、seqidno:2或seqidno:3所示)由上海生工生物工程有限公司合成得到。
两端加有人工转录因子结合位点的mar核心片段可插在pbcm1.0上游fspi位点,或sv40pa下游afei位点,或pef-1a上游nhei位点,或bghpa下游ecorv位点。
在以下的实例中,两端加有人工转录因子结合位点的mar核心片段插在pbcm1.0上游fspi位点及pef-1a上游nhei位点,mar核心片段序列如seqidno:1、seqidno:2或seqidno:3所示。
将两端加有人工转录因子结合位点的mar核心片段通过平端连接于pbcm1.0上游fspi位点。并测序正确。
将两端加有人工转录因子结合位点的mar核心片段通过粘末端连接于pbcm1.0上游nhei位点。并测序正确。
构建得到的两端加有人工转录因子结合位点的mar核心片段的表达载体分别命名为pbcm1.1,pbcm1.2,pbcm1.3,dna序列分别为seqidno.4,5,6。
实施例4含有两端加有人工转录因子结合位点的mar核心片段的表达载体中插入外源蛋白基因
cat基因插入pef-1a下游的kpni和bglii位点
将cat基因从pbudce4.1/lacz/cat(lifetechnologies)中pcr出并通过kpni和bglii位点装入上述实施例3所构建的表达载体。

cat基因的表达量用氯霉素乙酰基转移酶检测试剂盒(购自lifetechnologies)测定。
gfp基因从pcdna3.1/ct-gfp-top9(购自lifetechnologies)中pcr出并通过下游psti,bamhi位点装入实施例3所构建的表达载体。

gfp的表达量可通过cytofluorometricanalyses(bdfacscaliburflowcytometer)(购自bd公司)测定并通过winmdi2.8软件分析。
实施例5含有两端加有人工转录因子结合位点的mar核心片段的载体表达gfp及cat
两端加有人工转录因子结合位点的mar核心片段(序列如seqidno:1、seqidno:2或seqidno:3所示)对增加绿色萤光蛋白及氯霉素乙酰转移酶在转染的cho细胞中表达水平的影响。
利用freestylemax转染试剂(购自lifetechnologies)将实施例4中构建好的表达载体pbcm1.1,pbcm1.2,pbcm1.3导入cho-s细胞(购自lifetechnologies)中。将上述转染后细胞置37℃,8%co2,130-150rpm悬浮培养。48小时后开始稳定表达筛选。同时以不含mar和含mar全长序列的表达载体作为对照。实验结果如附图1-4所示,结果表明,含本发明的mar核心片段的表达载体在动物细胞中的外源蛋白的表达量获得大幅提高,gfp表达量提高了10-17倍(见附图1)。cat表达量提高了14-15倍(见附图3)。gfp高产细胞株的比例提高了25-40倍(见附图2)。cat高产细胞株的比例提高了25-40倍(见附图4)。
