背景技术
:普鲁兰酶,又名普鲁兰多糖-6-葡聚糖水解酶(pullulan-6-glucanohydrolase),是一种能够专一性切割普鲁兰多糖(普鲁兰多糖是由α-1,4-糖苷键连接的麦芽三糖重复单元经α-1,6-糖苷键聚合而成的直链状多糖)、支链淀粉、以及支链糊精中的α-1,6葡萄糖苷建的水解酶。普鲁兰酶根据作用糖苷键类型的差异主要被分为2类:ⅰ型普鲁兰酶和ⅱ型普鲁兰酶。其中,ⅰ型普鲁兰酶(ec3.2.1.41)具有水解普鲁兰多糖和分支糊精中α-1,6-葡萄糖苷键活力,不具有α-1,4-葡萄糖苷键水解活力;ⅱ型普鲁兰酶,又被称为淀粉普鲁兰酶(amylopullulanase,ec3.2.1.1/41),其既可以作用于普鲁兰多糖中的α-1,6-葡萄糖苷键,也能水解支链淀粉或一些糊精中的α-1,4-糖苷键,但是不能作用于普鲁兰多糖中的α-1,4-葡萄糖苷键。因此,普鲁兰酶常与如糖化酶、β-淀粉酶等的其他淀粉酶协同作用或单独作用,用于葡萄糖浆、麦芽糖浆、超高麦芽糖浆、海藻糖等的制备中,在食品、医药、发酵等领域均占据重要的地位。但是,由于普鲁兰酶蛋白分子普遍偏大,一般含有900个以上的氨基酸,单亚基分子量约在106kda;此外,普鲁兰酶单亚基通常含有6个结构域,并含有一些特有的氨基酸序列,这导致了普鲁兰酶极易形成包涵体,难以分泌到胞外,胞外表达水平低。因此,目前为止,国内外在普鲁兰酶开发中仍然存在很多问题,其中,普鲁兰酶可溶性分泌水平不足导致的发酵单位低是影响其规模化制备的关键因素之一。例如,邓毅等人将来源于b.naganoensis的酸性普鲁兰酶在芽孢杆菌中表达后,其酶活只有26.5u·ml-1;甄杰等人将来源于anoxybacillussp.lm18-11中的普鲁兰酶基因在枯草芽孢杆菌中表达后,虽然相比初始菌种表达活力有所提高,但是胞外酶活也只有42u·ml-1;吴丽双等人将来源于b.acidopullulyticus中的普鲁兰酶基因进行密码子优化后,转入毕赤酵母中进行表达,最终胞外酶活只有0.4u·ml-1,远远达不到工业生产的要求。并且,普鲁兰酶胞外表达水平低的问题,会大大提高发酵生产发酵普鲁兰酶时后续产物分离提取的难度,进一步提高工业生产成本,阻碍了普鲁兰酶的进一步推广应用。技术实现要素:[技术问题]本发明要解决的技术问题是得到一种具有高分泌能力,尤其是高胞外分泌能力的普鲁兰酶,以提高普鲁兰酶的产量,同时,降低普鲁兰酶分离提取的难度。[技术方案]为解决上述问题,本发明提供了一种具有高分泌能力的普鲁兰酶,所述普鲁兰酶的氨基酸序列如seqidno.1所示。在本发明的一种实施方式中,所述普鲁兰酶来源于阿氏芽孢杆菌(bacillusaryabhattai)gel-09;所述阿氏芽孢杆菌(bacillusaryabhattai)gel-09已于2017年6月9日保藏于中国典型培养物保藏中心,保藏编号为cctccno:m2017320,保藏地址为中国,武汉,武汉大学。所述阿氏芽孢杆菌(bacillusaryabhattai)gel-09已经记载于公开号为cn107760623a的专利申请文本中。本发明还提供了编码上述普鲁兰酶的基因。在本发明的一种实施方式中,所述基因的核苷酸序列如seqidno.2所示。本发明还提供了携带上述基因的重组质粒。在本发明的一种实施方式中,所述重组质粒的载体为pet载体、puc载体、pt7-7载体或pgex载体。在本发明的一种实施方式中,所述重组质粒的载体为pet-20b(+)载体。本发明还提供了携带上述基因或上述重组质粒的宿主细胞。在本发明的一种实施方式中,所述宿主细胞为大肠杆菌、芽孢杆菌或或酵母菌。在本发明的一种实施方式中,所述宿主细胞为大肠杆菌e.colibl21(de3)。本发明还提供了上述普鲁兰酶的制备方法,所述方法为使用上述宿主细胞,将上述宿主细胞接种至发酵培养基中进行发酵。本发明还提供了应用上述方法制备得到的普鲁兰酶。本发明还提供了上述普鲁兰酶或上述基因或上述重组质粒或上述宿主细胞或上述制备方法或上述制备得到的普鲁兰酶在水解淀粉方面的应用。本发明还提供了一种水解淀粉的方法,所述方法为使用上述普鲁兰酶或上述宿主细胞或上述制备得到的普鲁兰酶,将上述普鲁兰酶或上述宿主细胞或上述制备得到的普鲁兰酶与其他淀粉酶同时加入淀粉中进行酶解;所述其他淀粉酶包含糖化酶、α-淀粉酶、β-淀粉酶或淀粉葡萄糖苷酶中的一种或多种。[有益效果](1)本发明的普鲁兰酶胞外分泌能力高,将携带本发明普鲁兰酶的大肠杆菌摇瓶发酵48h,可使发酵液中的总酶活力达212.0u·ml-1,其中,胞外酶活达200.5u·ml-1,占了总酶活力的94.5%,因此,本发明的普鲁兰酶容易发酵制备,可以显著降低发酵工艺控制难度及成本;(2)本发明的普鲁兰酶作用条件温和,能够在温度40~60℃,ph6.0~7.5的条件下水解淀粉中的α-1,6-葡萄糖苷键,因此,本发明的普鲁兰酶在工业转化中能够降低成本,在食品、淀粉糖等行业中具有潜在利用价值。附图说明图1:重组菌pul937-pet20b(+)/e.colibl21(de3)发酵液的胞外上清组分、胞内上清组分以及胞内沉淀组分的sds-page电泳结果;其中,m为蛋白分子量标准;1为胞外上清组分;2为胞内上清组分;3为胞内沉淀组分。图2:本发明普鲁兰酶在不同ph条件下的酶活变化。图3:本发明普鲁兰酶在不同ph条件下保存24h后的酶活变化。图4:本发明普鲁兰酶在不同温度条件下的酶活变化。图5:本发明普鲁兰酶在不同温度条件下保存12h期间的酶活变化。具体实施方式下面结合具体实施例,对本发明进行进一步的阐述。下述实施例中涉及的大肠杆菌e.colibl21(de3)购自北京索莱宝科技有限公司,阿氏芽孢杆菌cctccno:m2017320购自中国典型培养物保藏中心,pet-20b(+)载体购自novagen公司。(上述菌株大肠杆菌e.colibl21(de3)、阿氏芽孢杆菌cctccno:m2017320均可以购买得到,不需要进行用于专利程序的保藏)下述实施例中涉及的培养基如下:lb液体培养基:酵母粉5.0g·l-1、胰蛋白胨10.0g·l-1、nacl10.0g·l-1、氨苄青霉素100μg·ml-1。lb固体培养基:酵母粉5.0g·l-1、胰蛋白胨10.0g·l-1、nacl10.0g·l-1、琼脂粉15g/l、氨苄霉素30μg·ml-1。tb培养基:甘油5.0g·l-1、胰蛋白胨12.0g·l-1、酵母粉24.0g·l-1、k2hpo4·3h2o16.4g·l-1、kh2po42.3g·l-1、甘氨酸7.5g·l-1、氨苄青霉素100μg·ml-1。下述实施例中涉及的检测方法如下:普鲁兰酶酶活的测定方法:分别取1ml的底物(1.0%普鲁兰溶液)和0.9ml的浓度为50mmol·l-1、ph为6.5的磷酸缓冲液于试管中,50℃水浴预热10min左右;加入0.1ml稀释的酶液样品,震荡混匀,50℃孵育10min,加入3mldns终止反应,沸水浴7min,冰水浴中冷却;向上述反应体系中加入10ml的蒸馏水,颠倒混匀,在540nm下测量其吸光值,同样条件下用以灭活的酶液作为酶液样品的反应体系作为空白。酶活力(u)定义为:在上述分析测定条件下,每分钟催化产生相当于1μmol葡萄糖还原力的酶量定义为一个活力单位(1u)。实施例1:编码本发明普鲁兰酶的基因的提取以及含有编码本发明普鲁兰酶的基因的重组菌的构建具体步骤如下:(1)阿氏芽孢杆菌cctccno:m2017320基因组dna的提取挑取阿氏芽孢杆菌cctccno:m2017320单菌落接种至lb液体培养基中,于37℃、200rpm的条件下振荡培养12h后,于12000rpm的条件下离心2分钟,收集菌体;用去离子水洗涤菌体后,再次离心并收集菌体并将收集的菌体悬浮于200μltris-edta(三羟甲基氨基甲烷-乙二胺四乙酸)缓冲液中,得到重悬液;在重悬液中先加入20μl溶菌酶,于37℃下保温30min,然后加入5μlrna酶,于37℃下保温30min,再加入30μl10%的sds(十二烷基硫酸钠)和15μl蛋白酶k,于37℃下保温60min,最后加入100μlnacl(5m)和80μlctab(十六烷基三甲基溴化铵),于65℃下保温20min,得到反应液;将反应液用700μl的酚:氯仿:异戊醇(25:24:1)进行抽提后,于12000rpm的条件下离心,得到上清液;将上清液用700μl的氯仿:异戊醇(24:1)抽提后,于12000rpm离心,再次得到上清液;将再次得到的上清液与1400μl体积的冰异戊醇混合后,于-20℃沉淀30min,并于12000rpm离心,得到沉淀;将沉淀用200μl70%的乙醇清洗后,于12000rpm离心,再次得到沉淀;将再次得到的沉淀用tris-edta缓冲液溶解,即得阿氏芽孢杆菌基因组dna;(2)编码阿氏芽孢杆菌cctccno:m2017320普鲁兰酶的基因的提取设计如下引物设计:pul937-f:5’-ccggcgatggccatggcggataccacaaaactcacg-3’(seqidno.3),pul937-r:5’-gtgcggccgcaagcttatcttactagtacaaatgtcg-3’(seqidno.4);利用上述引物,以提取的阿氏芽孢杆菌bacillusaryabhattaicctccno:m2017320基因组为模板,pcr扩增普鲁兰酶基因;pcr反应在50μl体系中进行,反应条件为94℃预热4min变性,随后进入循环,于94℃加热变性30s后于56℃降温退火30s,然后升温至72℃延伸3min,一共循环30次;随后72℃保存10min后置于12℃保存;将扩增得到的大小为2814bp的pcr片段割胶回收,将回收产物与pet20b(+)载体经双酶切(ncoi和hindiii)后进行连接,连接产物转化大肠杆菌jm109,转化产物涂布于含30μg·ml-1氨苄霉素的lb固体培养基,于37℃培养过夜,在lb固体培养基上挑取5个转化子,接入含有30μg·ml-1氨苄霉素的lb液体培养基培养,培养10h后提取质粒,将此质粒进行序列测定(序列为seqidno.4),结果表明此普鲁兰酶基因全长为2814bp个核苷酸,编码937个氨基酸。(3)编码阿氏芽孢杆菌cctccno:m2017320普鲁兰酶的基因的验证将测序正确的质粒(pul937-pet20b(+))热激转化e.colibl21(de3)宿主菌,于含30μg/ml氨苄青霉素的lb固体培养基上于37℃培养过夜,挑选转化子;将挑选得到的重组菌pul937-pet20b(+)/e.colibl21(de3)在lb液体培养基中于37℃培养过夜,然后接入tb培养基中于37℃培养4h,再用0.4mm异丙基硫代-d半乳糖苷(iptg)于30℃诱导培养44h,得到发酵液,将发酵液离心取上清,得到发酵上清液。将得到的发酵上清液作为酶液样品进行普鲁兰酶活力测定,检测结果为酶液样品中普鲁兰酶胞外酶活力为175.0u·ml-1,结果表明阿氏芽孢杆菌cctccno:m2017320普鲁兰酶表达成功,且具有普鲁兰酶活力。对比例1:含有编码其他来源普鲁兰酶的基因的重组菌的构建具体步骤如下:从ncbi上获取不同来源普鲁兰酶(分别为氨基酸序列如seqidno.5所示、核苷酸序列如seqidno.6所示的bacillusacidopullulyticus普鲁兰酶、氨基酸序列如seqidno.7所示、核苷酸序列如seqidno.8所示的bacillusmegaterium普鲁兰酶、氨基酸序列如seqidno.9所示、核苷酸序列如seqidno.10所示的pullulanibacillusnaganoensis普鲁兰酶)的核苷酸序列,通过人工合成得到这些序列后,然后将得到的序列分别连接到pet20b(+)载体上并转化宿主细胞e.colibl21(de3),得到重组菌pulbac-pet20b(+)/e.colibl21(de3)、pulbm-pet20b(+)/e.colibl21(de3)以及pulbn-pet20b(+)/e.colibl21(de3);其中,bacillusacidopullulyticus普鲁兰酶基因与pet20b(+)载体是经ncoi和hindiii双酶切后进行连接的,bacillusmegaterium普鲁兰酶基因是经ncoi和ecori双酶切后进行连接的,pullulanibacillusnaganoensis普鲁兰酶基因是经ecorv和xhoi双酶切后进行连接的。实施例2:不同来源普鲁兰酶的制备及不同来源普鲁兰酶分泌能力的检测具体步骤如下:(1)不同来源普鲁兰酶的制备将实施例1与对比例1制备得到的重组菌pulbac-pet20b(+)/e.colibl21(de3)、pulbm-pet20b(+)/e.colibl21(de3)以及pulbn-pet20b(+)/e.colibl21(de3)分别接种到lb液体培养基中,于37℃振荡培养8h,得到种子液;再分别取2.5ml种子液添加到tb培养基中(含有100μg·ml-1氨苄青霉素),置于37℃、200r·min-1的条件下振荡培养4h,加入0.4mm诱导剂iptg,诱导培养48h,得到重组菌pul937-pet20b(+)/e.colibl21(de3)发酵得到的发酵液a、pulbac-pet20b(+)/e.colibl21(de3)发酵得到的发酵液b、pulbm-pet20b(+)/e.colibl21(de3)发酵得到的发酵液c以及pulbn-pet20b(+)/e.colibl21(de3)发酵得到的发酵液d。(2)不同来源普鲁兰酶的检测1、样品处理将得到的发酵液a、b、c、d置于50ml离心管中,于4℃、6000rpm离心20min,收集上清(胞外上清组分),并收集离心沉淀的菌体细胞;将离心后的菌体细胞用等体积的磷酸盐缓冲液重悬,置于冰上进行超声破碎,功率采用130kw,超声3s,间歇5s,共20min;将破碎后细胞悬液置于4℃、12000rpm离心5min,分别收集离心上清(胞内上清组分)和细胞碎片(胞内沉淀组分)。2、样品检测检测胞外上清组分、胞内上清组分以及胞内沉淀组分中的普鲁兰酶酶活,检测结果为发酵液a中的胞外上清、胞内上清和胞内沉淀部分的酶活力分别为200.5u·ml-1、11.3u·ml-1和0.2u·ml-1,其中,胞外酶活力占总酶活力的94.6%;发酵液b中的胞外上清、胞内上清和胞内沉淀部分的酶活力分别为26.3u·ml-1、35.6u·ml-1和15.7u·ml-1,其中,胞外酶活力占总酶活力的33.9%;发酵液c中的胞外上清、胞内上清和胞内沉淀部分的酶活力分别为98.2u·ml-1、65.4u·ml-1和0.1u·ml-1,其中,胞外酶活力占总酶活力的60.0%;发酵液d中的胞外上清、胞内上清和胞内沉淀部分的酶活力分别为17.6u·ml-1、48.9u·ml-1和36.7u·ml-1,其中,胞外酶活力占总酶活力的17.1%;以购自碧云天生物技术有限公司的蛋白质分子量标准p0061为标准品,采用sds-page电泳方法检测发酵液a中三种组分的重组普鲁兰酶蛋白的蛋白表达水平(见图1),发现发酵液上清中普鲁兰酶蛋白占主要组分,胞内上清重组蛋白含量极少。可见,本发明普鲁兰酶的胞外分泌能力好,在普鲁兰酶胞外发酵制备过程中具有明显的优势。实施例3:本发明普鲁兰酶的的分离纯化具体步骤如下:将重组菌pul937-pet20b(+)/e.colibl21(de3)接种至lb液体培养基中培养10h,得到种子液;将种子液按5%接种量转接至tb培养基中发酵4h后,加入0.4mm诱导剂iptg,诱导培养48h,得到发酵液;将发酵液于4℃、10000g离心20min收集含有普鲁兰酶的发酵上清液;将上清液采用70%的(nh4)2so4进行盐析,离心收集沉淀;将沉淀复溶于ph6.8、50mmol·l-1的磷酸缓冲液中透析20h,中间更换一次缓冲液,得到样品;将样品经过0.45μm膜过滤后制成上样样品;将上样样品经deae阴离子交换色谱柱进行纯化,280nm紫外在线监测收集目的组分,分部收集含普鲁兰酶酶活的洗脱液,得到纯化后的普鲁兰酶。实施例4:本发明普鲁兰酶的的酶学性质研究具体步骤如下:(1)本发明普鲁兰酶的最适ph及ph稳定性配制ph值分别为4.0、5.0、6.0、7.0、8.0的磷酸缓冲液代替普鲁兰酶活力测定方法中的缓冲液,在50℃下测定普鲁兰酶活力,以酶活最高的为100%,其余酶活与之相比计算相对酶活,以考察酶的最适作用ph(检测结果见图2);配制ph值分别为4.0、5.0、6.0、7.0、8.0的磷酸缓冲液代替普鲁兰酶活力测定方法中的缓冲液,将纯化得到的酶分别在上述缓冲体系下于25℃保存24h后,在50℃下测定普鲁兰酶活力,以初始酶活为100%,保存后酶活与之相比计算残留酶活,以考察其ph稳定性(检测结果见图3)。由图2可知,本发明普鲁兰酶的最适ph为6.5,在ph7.0酶活力保留率为91%,当ph低于6.5或高于7.0时,普鲁兰酶活力快速下降,即本发明普鲁兰酶在弱酸性环境中拥有良好的催化活性,而在中性和弱碱性催化活性较弱,而偏离这个范围后,酶活迅速降低,几乎没有酶活,这说明本发明普鲁兰酶的最适作用ph较为温和,但范围较窄。由图3可知,本发明普鲁兰酶在ph5.5~7.5的弱酸性到中性条件下皆具有较好的ph稳定性,酶活保存率都在80%以上,而在ph低于5.0以下残存酶活非常低,而在ph高于7.5以后,酶活的保存率开始下降,说明本发明普鲁兰酶适合保存于中性或弱酸性条件下。。(2)本发明普鲁兰酶的最适温度及温度稳定性分别在40℃、45℃、50℃、55℃、60℃的条件下测定普鲁兰酶酶活,以酶活最高的为100%,其余酶活与之相比计算相对酶活,以考察酶的最适作用温度(检测结果见图4);将纯化得到的酶分别在30℃和40℃的条件下保温12h,相隔一段时间后将部分酶液取出,迅速冷却,测定其普鲁兰酶活力,以初始酶活为100%,保温后酶活与之相比计算残留酶活,以考察其温度稳定性(检测结果见图5)。由图4可知,本发明普鲁兰酶的最适温度为50℃,在45℃到50℃之间保留90%以上的活性,当温度高于50℃酶活力迅速下降,这说明本发明普鲁兰酶作用条件温和,非常适合应用于中温条件下的催化过程。由图5可知,本发明普鲁兰酶普在30℃和40℃保存下的前20h酶活下降较快,在40℃保温30h后,酶活残留50%左右;而在30℃条件下,前期酶活下降较快,但是在后期酶活保持稳定,半衰期能达到90h以上。(3)金属螯合剂与金属离子对本发明普鲁兰酶的影响将1ml1.0%的普鲁兰多糖溶液(底物)和0.9ml50mmol·l-1、ph6.5的磷酸缓冲液加入试管中,将金属离子ca2+、fe2+、zn2+、mg2+、zn2+、cu2+、co2+及金属螯合剂edta母液分别加入试管中使金属离子ca2+、fe2+、zn2+、mg2+、zn2+、cu2+、co2+及金属螯合剂edta母液在试管中的终浓度分别为1mm和5mm,得到混合液;将混合液在50℃水浴预热10min左右,然后,在混合液中加入0.1ml的稀释的酶液样品,震荡混匀,50℃孵育10min进行反应,反应结束时迅速加入3mldns终止反应,沸水浴显色7min,冰水中冷却,最后,向上述反应体系中加入10ml的蒸馏水,混匀,在540nm下测量其吸光值;以不加入金属离子和金属螯合剂的样品(control组)测定的酶活力为100%,其它底物测定酶活力与该活力之比为其相对酶活力(结果见表1)。由表1可知,1mmol·l-1的edta就对本发明普鲁兰酶具有很强的抑制作用,几乎使酶失活,残余酶活为4.8%;5mmol·l-1的edta对本发明普鲁兰酶处理后,残余酶活力为3.2%,这说明本发明普鲁兰酶发挥作用需要金属离子的参与;而在1mmol·l-1的金属离子中,mn2+、co2+对本发明普鲁兰酶具有中度抑制作用;fe2+、zn2+、cu2+对本发明普鲁兰酶的抑制作用非常明显;ca2+对本发明普鲁兰酶具有一定的促进作用;在5mmol·l-1的金属离子中,所有金属离子对于本发明普鲁兰酶均没有促进作用,其中,高浓度的mn2+离子对于酶的抑制作用最小,次之是ca2+离子,其它高浓度的金属离子对酶的抑制作用都很明显。表1金属离子和金属螯合剂对酶活力的影响金属离子或螯合剂(1mm)相对活性*金属离子或螯合剂(5mm)相对活性control100.0±2.2control100.0±3.97ca2+107.50±3.2ca2+93.35±3.66fe2+17.45±0.52fe2+6.62±0.33zn2+12.01±0.36zn2+6.54±0.32mg2+93.20±2.8mg2+95.97±2.79mn2+72.35±2.2mn2+71.62±3.58cu2+1.40±0.14cu2+0.94±0.04co2+52.85±1.59co2+19.71±0.98edta4.75±0.14edta3.17±0.98(4)本发明普鲁兰酶的动力学参数分别以浓度为0.1、0.25、0.5、1.0、1.5、2.0、3.0、4.0、5.0、6.0、8.0、10.0mg/ml的普鲁兰多糖溶液为底物,在50℃、ph6.5条件下测定普鲁兰酶初始水解活力,采用graphpadprism7.0软件中的非线性回归方法对数据进行拟合,分别得到米氏(michaelis-menten)方程的km和vmax值,再计算得到kcat和kcat/km值;kcat值的计算公式为:kcat=vmax/m/106124.74/60;其中,m为反应中加入的酶的质量,单位为mg。结果显示,本发明普鲁兰酶对普鲁兰底物的最大反应速率(vmax)、催化常数(kcat)、底物亲和力(km)和催化效率(kcat/km)分别为489.6u/mg-1、844.9s-1、0.19mg·ml-1、4371.0s-1·mg-1·ml,这说明本发明普鲁兰酶活力较高。虽然本发明已以较佳实施例公开如上,但其并非用以限定本发明,任何熟悉此技术的人,在不脱离本发明的精神和范围内,都可做各种的改动与修饰,因此本发明的保护范围应该以权利要求书所界定的为准。序列表<110>南京林业大学<120>一种具有高分泌能力的普鲁兰酶及其应用<160>10<170>patentinversion3.3<210>1<211>937<212>prt<213>人工序列<400>1alaaspthrthrlysleuthrilehistyrglnproalaserasnasp151015thrlysglutrpglyleutrpvalpheprogluglyglygluglylys202530protyralaphethrglygluaspglnpheglylysvalalagluval354045gluleuproglythrtyrasplysvalglypheilevalargthrglu505560sertrpglulysaspglyglyaspargphevalservalgluasngly65707580gluglygluvaltrpvallysserglyaspgluhisthrtyrthrser859095proproaspglyglutyrargaspleuproglupheasplysvalasn100105110valthrleuasntyrhisargtyraspglyasptyrserglytrpasn115120125iletrpthrtrpproglyaspglulysgluglylysglnilegluphe130135140thrgluaspthrasnpheglylyslysalathrtyrthrileasnser145150155160glyserglyasnleupheasplysileglypheile
