微环DNA表达连接人与动物靶细胞与效应细胞的桥接分子及其应用的制作方法
本发明属于生物医药领域,具体涉及一种微环(minicircle,mc)dna载体,更具体为一种用于在体内表达人-动物交叉反应性双特异性抗体(bispecificantibodycross-reactivebetweenhumanandanimal,crbsab)的微环dna载体。
背景技术
:双特异性抗体(bispecificantibody,简称bsab)是基因工程抗体的一种,是一类具有双功能的抗体杂交分子,两价抗体中的可变结构域(或者fab段)分别具有不同的特异性,能够与两种不同的配体结合。研究人员在设计bsab分子时,考虑到bsab能够与靶细胞上的抗原分子结合,又可以与效应细胞上的功能分子结合。这样,bsab分子就在靶细胞(抗原分子)和效应细胞(功能分子)之间架起桥梁,激发具有导向性的免疫反应。抗肿瘤和抗免疫活性细胞(比如t细胞或nk细胞)的双特异性抗体,不仅具有激活免疫活性细胞的作用,而且可以通过抗肿瘤的fab段特异性结合肿瘤细胞而发挥作用,提高局部免疫活性细胞的浓度,增强效应分子杀伤肿瘤的能力。bsab现已成为抗体工程领域的研究热点,在肿瘤的免疫治疗中具有广阔的应用前景,是未来抗体药物市场的发展方向。目前双特异性抗体bsab的制备主要包括三种方法:化学偶联法、杂交-杂交瘤法、基因工程抗体制备法。其中基因工程的方法是目前最常用和最先进的工艺技术。微环dna载体表达方法属于基因工程制备法。利用微环dna载体可以表达微环双特异性抗体mc.bsab,所表达的双特异性抗体可以连接靶细胞与t细胞或nk细胞,并触发t细胞或nk细胞杀灭靶细胞。与该发明最接近的相关技术是我们之前发明的微环dna载体表达双特异性抗体的技术mc.bsab(cn201710245146.6)。在药物临床前研究中,动物模型的相关试验是必不可少的。猴和鼠类模型是常见的模式动物,常用来评价药物临床前的安全性与有效性。目前研发中的双特异性抗体大多是人化或人源化的,仅对来源于人的抗原具有特异性的,与猴或鼠类的抗原无交叉反应,难以利用猴或鼠类评估其安全性与有效性。相反,少数只对猴与鼠类有效的抗体,实验结果也不一定等同于人。然而,本发明解决了这个难题,因为存在人-动物的交叉反应,只一个版本的微环基因药就能够同时用于动物实验、人的临床试验和上市的产品,避免了不同版本的生物药可能存在的差异及潜在的安全隐忧。由此可见,开发一种人-动物交叉反应性双特异性抗体(bispecificantibodycross-reactivebetweenhumanandanimal,crbsab),有利于借助动物模型(如猴或鼠类)评价双特异性抗体类药物的安全性与有效性,以促进药物的开发进程,是一个重要的进步。技术实现要素:本发明提供一种表达人-动物交叉反应性靶细胞-效应细胞桥接分子(bridgebetweentargetcellandeffectorcellofbothhumanandanimals,crbtec)的重组基因载体,所述桥接分子包含:与靶细胞作用靶点特异性结合的部分a,和与效应细胞作用靶点特异性结合的部分b。在一个实施方案中,所述重组基因载体选自非病毒基因载体。优选的,所述重组基因载体选自微环dna载体。其中,所述桥接分子选自蛋白或多肽。优选的,所述桥接分子选自双特异性抗体。其中,所述动物选自猴或鼠类。优选的,所述猴选自食蟹猴,鼠类选自大鼠或小鼠。在一个实施方案中,所述重组基因载体为重组表达载体,选自原核表达载体或真核表达载体;优选真核表达载体;更优选用于哺乳动物细胞真核表达的重组表达载体。在一个实施方案中,所述部分a和所述部分b分别选自蛋白分子或多肽分子。优选的,所述部分a和部分b分别选自fab、fab’、单链抗体(scfv)、单域抗体(vhh)、单链t细胞受体(sctcr)和其它。在一个实施方案中,所述靶细胞选自癌细胞、受感染细胞或其它。所述效应细胞选自t细胞、nk细胞或其它。优选的,所述部分a特异性结合的作用靶点选自:cd19、cd20、epcam、egfr、her2、vegfr、pd-1、pd-l1、il-17r、tnf-αr、病毒抗原或其它。所述部分b特异性结合的作用靶点选自:cd3、cd16、cd28、4-1bb、ox40、tcr、cd56、nkg2d、ncr或其它。在一个实施方案中,所述部分a包含:重链可变区,其氨基酸序列如seqidno:1、5或9所示,和轻链可变区,其氨基酸序列如seqidno:3、7或11所示;以及,所述部分b包含:重链可变区,其氨基酸序列如seqidno:13或17所示,和轻链可变区,其氨基酸序列如seqidno:15或19所示。在一个实施方案中,所述桥接分子选自人-动物交叉反应性双特异性抗体(crbsab)。所述crbsab能够结合人或动物的靶细胞作用靶点,也能够结合人或动物的效应作用靶点。其中,所述动物选自猴或鼠类。优选的,所述猴选自食蟹猴,鼠类选自大鼠或小鼠。在一个实施方案中,所述部分a包含的重链可变区具有与seqidno:1所示序列具有至少90%、95%、98%或99%同源性的氨基酸序列,包含的轻链可变区具有与seqidno:3所示序列具有至少90%、95%、98%或99%同源性的氨基酸序列。在一个实施方案中,所述部分b包含的重链可变区具有与seqidno:5所示序列具有至少90%、95%、98%或99%同源性的氨基酸序列,包含的轻链可变区具有与seqidno:7所示序列具有至少90%、95%、98%或99%同源性的氨基酸序列。本发明提供了一种表达人-动物交叉反应性靶细胞-效应细胞桥接分子的重组基因载体,所述重组基因载体包含所述桥接分子的编码基因。在一个实施方案中,所述重组基因载体包含:部分a的编码基因,和/或包含部分b的编码基因。优选的,部分a的编码基因包含:重链可变区的编码基因,其核苷酸序列如seqidno:2、6或10所示,和/或轻链可变区的编码基因,其核苷酸序列如seqidno:4、8或12所示。优选的,部分b的编码基因包含:重链可变区的编码基因,其核苷酸序列如seqidno:14或18所示,和/或轻链可变区的编码基因,其核苷酸序列如seqidno:16或20所示。在一个实施方案中,所述重组基因载体包含:与上述部分a和/或部分b的编码基因的核苷酸序列具有至少90%、95%、98%或99%同源性的核苷酸序列,且编码得到的桥接分子具有与之前实施方案中所述的桥接分子相同的功能。本领域技术人员公知,在不改变所编码的氨基酸的情况下,所述编码基因序列中的一个或多个密码子可以进行等义替换,如一个或几个密码子,如1、2、3、4、5、6、8、9、10、15、20、30、40、50个密码子。在一个实施方案中,所述重组基因载体包含所述桥接分子的编码基因,其核苷酸序列如seqidno:21-38中任一项所示。在一个实施方案中,所述重组基因载体包含:与上述桥接分子的编码基因的核苷酸序列具有至少90%、95%、98%或99%同源性的核苷酸序列,且编码得到的桥接分子具有与之前实施方案中所述的桥接分子相同的功能。本领域技术人员公知,在不改变所编码的氨基酸的情况下,所述编码基因序列中的一个或多个密码子可以进行等义替换,如一个或几个密码子,如1、2、3、4、5、6、8、9、10、15、20、30、40、50个密码子。本发明提供了一种由之前实施方案中所述的重组基因载体所表达的桥接分子。优选的,所述重组基因载体选自非病毒基因载体。更优选的,所述重组基因载体选自微环dna载体。本发明提供了一种之前实施方案中所述的重组基因载体的制备方法,具体步骤包括:(1)从现有技术中分别获取人-猴交叉反应性抗体的重链可变区(vh)序列、轻链可变区(vl)序列。(2)根据上述vh、vl序列设计成多种组合形式的桥接分子,并构建表达所述桥接分子的重组基因载体。任选的,(3)在体内外鉴定所述桥接分子的表达水平,并检测表达产物介导效应细胞对靶细胞的杀伤效果。在一个实施方案中,所述重组基因载体选自非病毒基因载体。优选的,所述重组基因载体选自微环dna载体。优选的,所述桥接分子选自双特异性抗体。更优选的,所述重组基因载体选自表达双特异性抗体的微环dna载体(mc.bsab)。其中,所述猴选自食蟹猴。本发明提供了一种宿主细胞,包含之前实施方案中所述的重组基因载体,或由之前实施方案中所述的制备方法所得到的重组基因载体。在一个实施方案中,所述宿主细胞包括细菌细胞、酵母菌细胞、昆虫细胞或哺乳动物细胞。本发明提供了一种之前实施方案中所述的双特异性抗体的制备方法,具体步骤包括:(1)构建所述重组基因载体;(2)将所述重组基因载体导入宿主细胞;(3)在适合表达的条件下,培养宿主细胞,诱导表达,分离纯化,获得所述双特异性抗体。本发明提供了一种药物组合物,包含之前实施方案中所述的重组基因载体,或由之前实施方案中所述的制备方法所得到的重组基因载体,或由之前实施方案中所述的重组基因载体所表达的桥接分子,以及药学上可接受的载体。在一个实施方案中,所述药物组合物可根据常规方法制成药物制剂。制剂过程中,优选将重组基因载体或双特异性抗体与药学上可接受的载体混合或用载体稀释。当载体作为稀释剂时,其可以为固体、半固体或液体。制剂选自片剂、丸剂、粉剂、胶囊、混悬剂、乳剂、溶液剂、气溶胶、胶囊、注射用溶液等形式。合适的载体、赋形剂或稀释剂包括水、乳糖、葡萄糖、蔗糖、山梨醇、甘露醇、硅酸钙、纤维素、聚乙烯吡咯烷酮、羟基苯甲酸甲酯、羟基苯甲酸丙酯、滑石粉、硬脂酸镁和矿物油等。制剂还可以包括填充剂、抗凝血剂、润滑剂、保湿剂、调味剂、乳化剂、防腐剂等。本发明提供了一种之前实施方案中所述的重组基因载体、所述的桥接分子、所述的宿主细胞或所述的药物组合物在制备用于治疗癌症、感染或其它疾病的药物中的用途。本发明的积极效果包括:在药物临床试验之前,需要分子阶段、细胞阶段、动物阶段的试验数据,而动物的试验数据是必不可少的。本发明的人-动物交叉反应性双特异性抗体既能够结合人的抗原分子,又能够交叉识别动物的抗原分子,有利于借助模式动物来评价双特异性抗体类药物的安全性与有效性,以促进药物的开发进程。附图说明图1:表达抗cd19(scfv)/cd3(scfv)双特异性抗体的微环dna载体结构图。图2:表达抗cd19/cd3人-动物交叉反应性桥接分子crbtec(dart)的微环dna载体结构图。图3:表达抗cd19/cd3人-动物交叉反应性桥接分子crbtec(dart-fc)的微环dna载体结构图。图4:表达抗epcam(scfv)/cd3(scfv)双特异性抗体的微环dna载体结构图。图5:表达抗epcam/cd3人-动物交叉反应性桥接分子crbtec(dart)的微环dna载体结构图。图6:表达抗epcam/cd3人-动物交叉反应性桥接分子crbtec(dart-fc)的微环dna载体结构图。图7:采用page检测epcam/cd3人-动物交叉反应性桥接分子crbtec的表达。具体实施方式下述实验方法如无特别说明,均为常规方法,所使用的实验材料如无特别说明,均可容易地从商业公司获取。在不背离本发明精神的情况下,本领域技术人员结合公知技术,可以对本发明做出许多修改,这样的修改也落入本发明的范围。实施例1、人-动物交叉反应性桥接分子(crbtec)的构建设计crbtec并构建相应的微环dna表达载体,crbtec表达框包括多种结构,如下:(1)scfv-scfv结构:其中,sp为信号肽(signalpeptide);linker为连接序列。tar表示靶细胞(target)抗原,如cd19、cd20、epcam、her2等。anti-tar.vh的氨基酸序列如seqidno:1、5或9所示,anti-tar.vl的氨基酸序列如seqidno:3、7或11所示;anti-cd3.vh的氨基酸序列如seqidno:13所示,anti-cd3.vl的氨基酸序列如seqidno:15所示;anti-cd16.vh的氨基酸序列如seqidno:17所示,anti-cd16.vl的氨基酸序列如seqidno:19所示。其中还包括上述各个元件的编码基因;scfv-scfv双特异性抗体的编码基因如seqidno:21、24、27、30、33或36所示。(2)双特异性抗体dart(dual-affinityre-targetingantibody)结
