[0081] 用于特异性扩增的引物的有效设计是决定实验成功的最关键环节。目前虽有一些 设计引物的软件和设计引物的一些基本原则,但根据这些设计软件和设计原则,可以设计 出多条不同的引物序列,而采用不同的引物序列会产生不同的效果,个别碱基的替换或增 减都会对试验结果产生重要影响。因此,对于扩增引物的设计并非是常规的软件设计所能 得到的。本实施例中,用于特异性扩增猪传染性胸膜肺炎放线杆菌、猪瘟病毒、猪副嗜血杆 菌、猪流感病毒、猪肺炎支原体、猪圆环病毒II型、猪细小病毒、猪蓝耳病毒、猪伪狂犬病病 毒和猪链球菌病原基因最初设计有30对引物序列,具体如下:
