背景技术:

表皮生长因子受体和同源HER-2/ErbB-2蛋白激酶属于I型或者ErbB/HER受体酪氨酸激酶家族,与细胞的增值、存活、黏附、迁移和分化有密切联系。已成为在家族四名成员中最可行的抗肿瘤分子靶点,其他的分别是HER-3and HER-4。在乳腺、卵巢、非小细胞肺癌中经常能发现表皮生长因子受体和HER-2过度表达或者激活突变,使得它们成为这些癌症的治疗目标。表皮生长因子受体以及HER-2成为当前研究热点,它们的过度表达以及异常激活通常能导致细胞恶性转化。并且它们与术后不良、放疗和化疗耐受以及肿瘤血管生成有关。EGFR和ErbB-2的封锁已被临床验证成为肿瘤治疗的一个有吸引力的方法。

二氢吡唑是很多天然化合物和合成药物中的核心结构单元,据报道含有二氢吡唑骨架一些新颖的化合物作为有效的靶向EGFR抑制剂,其中一些已经被证明具有良好的抗癌活性。此外,苯甲酰肼类化合物具有抗高血糖、抗氧化、抗惊厥、抑菌、杀菌、抗癌等药理活性。本发明的一类含二氢吡唑结构的新型苯甲酰肼衍生物可能有希望成为开发新的,有效的和安全的导向化合物。



技术实现要素:

本发明的目的在于提供一类作为EGFR抑制剂的新型苯甲酰肼衍生物及其制备方法。

本发明的一类含二氢吡唑结构的苯甲酰肼衍生物,其结构如式所示,

具体实施方式

本发明的制备过程和相关产物的结构式如下所述:

步骤1.室温条件下,向反应容器中依次加入结构如式A所示的化合物,MeOH,SOCl2反应,充分反应后,得到结构如式B所示的化合物。

步骤2.依次向反应容器中加入结构如式C所示的化合物及结构如式D所示的化合物,EtOH,KOH,室温反应,充分反应后,得到结构如式E所示的化合物。

步骤3.依次向反应容器中加入无水乙醇,乙酸,结构如式B所示的化合物,结构如式E所示的化合物,80℃充分反应,得到结构如式F所示的化合物。

步骤4.依次向反应容器中加入甲醇溶液,结构如式F所示的化合物,水合肼,DMF,70℃充分反应后得到结构如式G所示的化合物。

步骤5.依次向反应容器中加入乙酸,乙醇溶液,RCHO以及结构如式G所示的化合物,室温充分反应后得到结构如式H所示的化合物。
