技术领域
本发明涉及一种重组嵌合蛋白,尤其是一种携带轮状病毒抗原表位的重组嵌合蛋白及其制备,属于医学生物技术领域。
背景技术
轮状病毒(Rotavirus,简称RV)是婴幼儿腹泻的主要病原体。世界范围内每年大约有60多万例RV感染患者死亡,其中80%发生在发展中国家。RV属呼肠病毒科(Meoviridae),轮状病毒属iRotavirus\ RV基因组由11个片段的双链RNA组成,分别编码6个结构蛋白(VP1-VP4, VP6和VP7)和6个非结构蛋白(NSP1-NSP5/NSP6)。 根据RV组(亚组)抗原蛋白VP6的抗原性的不同,可将至今已发现的轮状病毒RV分为A G七个组,A组是婴幼儿腹泻的主要病原体。根据病毒蛋白编码框(ORF)核苷酸序列同源性差异,可将A组轮状病毒RV的12个蛋白(基因)分为不同的基因型。根据RV基因型分类标准,A组轮状病毒RV的结构蛋白VP6至少可分为16个基因型(11-116型)。VP6蛋白由第6基因片段编码,含397个氨基酸,分子量约为44. 8KDa。VP6蛋白是轮状病毒RV中含量最丰富的蛋白,约占病毒总蛋白量的51%。RV内壳中间层由260个致密的VP6蛋白三聚体共780个蛋白分子构成。VP6蛋白具有重要的生物学功能,是病毒基因组转录和病毒组装不可缺少的成分。VP6蛋白也具有重要的免疫学性质。近年进行的VP6蛋白免疫学研究表明,一些抗VP6蛋白的IgA单克隆抗体诸如7D9所产生的IgA具有保护作用,在SCID鼠中可清除其慢性RV病毒感染。一些研究表明VP6蛋白的抗病毒作用可能是通过IgA的分泌途径而阻碍了病毒的复制循环过程,或作为一种黏膜的分泌型IgAJf RV感染的上皮细胞排除,从而起到保护作用。研究发现VP6蛋白分子中至少存在两个Th细胞抗原表位和一个交叉反应的细胞毒性T淋巴细胞(CTL)表位。有研究表明在使用麦芽糖结合VP6蛋白对小鼠进行黏膜免疫后,IFN- y被认为是⑶4+T细胞分泌的唯一有效的抗RV的细胞因子。众多的实验表明,经VP6蛋白免疫后,在动物体内确实能引起抗RV感染的作用。抗VP6蛋白单克隆抗体具有抑制病毒转录活性和保护小鼠免受病毒感染的作用。用重组表达的VP6蛋白免疫小鼠,也可诱导小鼠产生免疫保护作用。VP6蛋白可自我组装成病毒样颗粒(VLPs)。由于VLPs没有感染性,在研究细胞相互作用,开发诊断制剂和疫苗方面,具有更好的安全性。与单一分子的重组抗原相比,VLPs具有更好的免疫原性,可刺激机体产生体液和细胞免疫反应。由于VLPs具有高密度的呈递抗原表位,可产生很强的免疫反应,是许多疫苗研制中很有前景的材料。至今所有已用于人体的轮状病毒RV疫苗都是减毒活疫苗。1997年始用于人体的多价减毒重组活疫苗Rotashield因后来发现与肠套叠有关联而在使用一年后即停止使用。2006年和2008年分别获准在人群中使用的5价重组减毒活疫苗RotaTeq和单价重组减毒活疫苗Rotarix现也已发现有猪环状病毒DNA污染。Rotarix已在美国人群中停止使用,RotaTeq在人群中尚可正常使用。由于上述用于人类的RV疫苗存在肠套叠和其他病毒核酸污染等风险,且都存在只能降低RV感染引起的死亡率和重症腹泻,不能完全保护RV感染的不足,在Rotarix停用后,RV感染率似有所提高。在尚无理想的治疗轮状病毒RV感染的药物,以及轮状病毒RV疫苗尚不十分理想的今天,进一步研制更安全有效的新型轮状病毒RV疫苗具有重要的经济与社会价值。

发明内容
本发明的目的在于提供一种携带有轮状病毒P[4] 223抗原表位的重组蛋白及其制备,以用作联合重组嵌合疫苗。本发明的目的在于提供一种携带有轮状病毒P [4] 56抗原表位的重组蛋白及其制 备,以用作联合重组嵌合疫苗。本发明的目的又在于提供一种携带有轮状病毒Gl [142]抗原表位的重组蛋白及其制备,以用作联合重组嵌合疫苗。本发明的目的还在于提供一种携带有轮状病毒G4[208]抗原表位的重组蛋白及其制备,以用作联合重组嵌合疫苗。本发明的目的再在于提供一种携带有轮状病毒P [8] 296抗原表位的重组蛋白及其制备,以用作联合重组嵌合疫苗。本发明的目的还在于提供一种携带有轮状病毒G2 [87]抗原表位的重组蛋白及其制备,以用作联合重组嵌合疫苗。本发明的目的在于提供一种携带有轮状病毒P [4] 223、P [4] 56,Gl [142]、G4 [208]、P[8]296和G2[87]抗原表位的重组蛋白及其制备,以用作联合重组嵌合疫苗。本发明提供的携带轮状病毒P[4]223抗原表位的重组蛋白的氨基酸序列为

8)、将步骤7)得到的VP6载体蛋白编码基因用常规的基因克隆和重组技术,连接到pET表达质粒DNA的Nde I/BamH I位点,得到携带VP6F载体蛋白编码基因的重组质粒pET6F。所述步骤2 )的轮状病毒TB-Chen株为公知的病毒株。本发明具有下列优点和效果采用上述方案获得的各个携带轮状病毒不同抗原表位的重组嵌合蛋白,经免疫学Western Blot检测,所述各个重组嵌合蛋白均能与抗轮状病毒全病毒,抗轮状病毒的VP6和抗相应轮状病毒的VP4/VP7抗体发生特异性免疫反应。说明上述各个重组表达的重组嵌合蛋白具有VP6载体蛋白和所插入的轮状病毒抗原表位的良好的免疫反应性,且不具有传染性,没有毒力返祖和肠套叠等副作用,对进一步开发轮状病毒RV重组嵌合疫苗,具有较高的应用价值。

A、6174F酶切片段制备酶切反应体系为3iil的6174 片段,311的Nde I (15U),3iU的Sac I (15U),8u I的IOX缓冲液,63 yl的水;将酶切混合液分别于37°C水浴反应3小时后,得酶切片段,用I. 5%的琼脂糖凝胶电泳分离和纯化该酶切片段;
B、除使用SacI/BamH I双酶切外,6174R酶切片段制备与6174F酶切片段相同;
C、酶切片段的连接将6174F和6174R酶切片段与T4DNA连接酶及缓冲液混合后,在18°C连接反应6h,得到携带有Sac I位点(gagctc)的VP6编码基因
3)运用步骤2)的PCR扩增、酶切和连接,使用对应的PETL5/P197R和P197F/PVP6-3引物对及内切酶Nde I/BspT104 I和BspT104 I/BamH I,在步骤2)获得的携带有Sac I位点的VP6编码基因上插入BspT104 I (ttcgaa)位点,得到携带有Sac I位点和BspT104 I位点的VP6编码基因;
4)用与步骤2 )相同的PCR扩增、酶切和连接,使用对应的PETL5/P244R和P244F/PVP6-3引物对及内切酶Nde I/Kpn I和Kpn I/BamH I,在步骤3)所得携带有Sac I位点和BspT104 I位点的VP6编码基因上插入Kpn I (ggtacc)位点,得到携带有Sac I位点、BspT104 I位点及Kpn I (ggtacc)位点的VP6编码基因;

本发明提供的携带有P[4]223抗原表位的重组蛋白通过下列步骤制备
A、根据抗原表位氨基酸序列,设计下列引物

P[4]223R CTGGAACTACATTTCTAGTATTTTGAATTGGTGGGAGCT ;
B、将步骤A的引物进行下列配对
将VP4抗原表位P [4] 223的两条人工合成的寡核苷酸即正链P [4] 223F和负链P[4]223R 配成引物对P[4]223F/ P[4]223R ;
C、将步骤B的引物对在下列条件下进行退火,退火混合液中含有2iU的正向引物,
2Ul的反向引物,以及7 ill的PCR缓冲液和59 u I的水;将退火混合液于65°C水浴反应5分钟后,再于37°C水浴反应45分钟,反应体系中加入400 ii I的TE,并与等体积异丙醇充分混匀,室温放置10分钟后,4°C,15,OOOrpm离心15min,沉淀用质量浓度为75%的乙醇漂洗,干燥后,用10 Ul的TE溶解;得到在5’端和3’端分别带Sac I位点的互补双链P[4]223互补双链;
D、将携带有SacI位点的重组轮状病毒VP6载体蛋白编码基因的质粒pETP6F,在下列条件下用Sac I酶切酶切混合液含3 ill的Sac I内切酶(15U),以及10 的pETP6FDNA,7u I的IOX缓冲液,50iU的水;将酶切混合液于37°C水浴反应3小时,得酶切质粒片段Sac I酶切质粒片段、用I. 5%的琼脂糖凝胶电泳分离和纯化所得酶切质粒片段;
E、将步骤D所得酶切质粒片段与步骤C所得互补双链进行如下相应的配对

F、将步骤E得到的重组质粒pETP6F174S/P[4]223,在下列条件下转化BL21(DE3)感受态细胞=IOiIl的重组质粒pETP6F174S/P[4]223与IOOiIl的BL21 (DE3)感受态细胞混匀,冰上放置30分钟,42 °C放置70秒钟,加入500 的LB培养液后,在37°C培养30分钟,培养物涂含有200 u g/ml氨苄青霉素的I. 5%琼脂培养板,于37°C培养14个小时,挑取单个菌落放入含200 u g/ml氨苄青霉素的LB培养液中培养和表达,得到携带有P [4] 223抗原表位的重组蛋白rVP6/P[4]223。实施例3 A、引物设计为

1)将携带有轮状病毒P[4] 223抗原表位的重组蛋白rVP6/P [4] 223,携带有轮状病毒P[4] 56抗原表位的重组蛋白1^ 6 外[4]56,携带有轮状病毒61[142]抗原表位的重组蛋白rVP6F/Gl[142],携带有轮状病毒G4[208]抗原表位的重组蛋白rVP6F/G4 [208],携带有轮状病毒P [8] 296抗原表位的重组蛋白rVP6F/P[8]296,携带有轮状病毒G2[87]抗原表位的重组蛋白 rVP6F/G2[87],和同时携带 P[4]223、P[4]56、Gl [142]、G4[208]、P[8]296 和G2[87]六个抗原表位的重组蛋白rVP6F⑥R,以及用作对照的重组轮状病毒NSP2蛋白和用作对照的牛血清白蛋白,各取g分别溶于20iU水,分别加入5iU 5X变性缓冲液,混匀后,在100。。变性5min ;
2)将上述I)的变性蛋白样品分别用10%聚丙烯酰胺凝胶电泳(10%SDS-PAGE)分离,并在电压15V、电流50mA的条件下,电泳转印25 min,转移到PVDF膜上;将膜置于含5%脱脂奶粉的PBS/T液中于4° C封闭过夜;
3)用PBS/T洗膜5次,分别加入抗轮状病毒全病毒豚鼠免疫血清抗体(5%脱脂奶粉1:400稀释),或抗重组轮状病毒VP6蛋白豚鼠免疫血清抗体(1%脱脂奶粉1:400稀释),或抗重组轮状病毒NSP2蛋白豚鼠免疫血清抗体(1%脱脂奶粉1:400稀释),或抗重组轮状病毒VP4蛋白豚鼠免疫血清抗体(1%脱脂奶粉1:400稀释),或抗重组轮状病毒VP7蛋白豚鼠免疫血清抗体(1%脱脂奶粉1:400稀释),于37°C孵育I. 5 h ;用PBS/T液洗膜5次,每次
5min ;
4)加入HRP标记的羊抗豚鼠IgG(1:2,000如上稀释),于371孵育I h ;用PBS/T漂洗5次,DAB显色;显色后,用蒸馏水漂洗膜,终止显色,观察并分析结果如下

本发明提供一种携带轮状病毒抗原表位的重组嵌合蛋白及其制备,分别是携带有轮状病毒P[4]223,P[4]56、G1[142]、G4[208]、P[8]296、G2[87]抗原表位的六个重组蛋白,以及同时同时携带有P[4]223、P[4]56、G1[142]、G4[208]、P[8]296和G2[87]抗原表位的重组蛋白,经免疫学WesternBlot检测,所述各个重组嵌合蛋白均能与抗轮状病毒全病毒,抗轮状病毒的VP6和抗相应轮状病毒的VP4/VP7抗体发生特异性免疫反应。说明上述各个重组表达的重组嵌合蛋白具有VP6载体蛋白和所插入的轮状病毒抗原表位的良好的免疫反应性,且不具有传染性,没有毒力返祖和肠套叠等副作用,对进一步开发轮状病毒RV重组嵌合疫苗,具有较高的应用价值。
