本发明涉及生物技术领域,具体涉及一种基因工程菌生产β-胡萝卜素的方法及其基因工程菌。



背景技术:

β-胡萝卜素是一种橘黄色的脂溶性的类胡萝卜素,是联合国粮农组织和世界卫生组织食品添加剂联合专家委员会认定的A类的营养食品添加剂。在人体中,β-胡萝卜素是维生素A的前体;可以作为抗氧化剂清除体内的自由基,降低老年人心力衰竭的风险;激发免疫系统,有防癌和抗癌的功能。此外β-胡萝卜素还可以用于食品色素、动物饲料添加剂、保健品、化妆品及药品中。目前β-胡萝卜素的市场需求不断增加,它的应用领域也从食品着色转向营养食品、保健品和药品三大领域。

β-胡萝卜素制备生产有三种方法:化学合成法;天然提取法;微生物发酵法。采用化学合成法人工合成的β-胡萝卜素几乎完全是反式异构体形式,不具备许多天然β-胡萝卜素的生理功能,不能完全被人体吸收,长期服用会对人体产生一定程度的毒副作用,随着人们对绿色天然食品的重视化学合成的β-胡萝卜素不被大多数食品科学家所接受。从植物中提取天然β-胡萝卜素受原料含量、气候、产地等条件限制,且工艺复杂,难以大量生产,应用也收到限制。

微生物发酵法因不受环境等条件的影响,并且容易培养,产量高而广泛受到国内外的重视,是生产β-胡萝卜素的最好方法。发酵常用的菌株主要分为三类:藻类、酵母和真菌。藻类和酵母的产量相对较低无法达到工业化生产的要求。

三孢布拉霉(Blakeslea trispora)是目前生产β-胡萝卜素的首选菌株,但是以三孢布拉霉发酵生产β-胡萝卜素的培养基成分复杂、成本相对较高。三孢布拉霉是异宗结合菌,进行有性生殖的方式是异宗接合,即由来源和性质不同的“+”“-”菌株才能相互融合,进行有性生殖。接合孢子产生时通常进行减数分裂,于是产生含有单倍体孢子的孢子囊。诱导配囊柄的激素成为三孢酸,三孢酸的主要功能是诱导有性生殖,促进β-胡萝卜素的生物合成。在工业生产中,常常要考虑“+”“-”菌株的配比,发酵条件相对复杂,这也给工业生产带来许多不必要的困难。



技术实现要素:

本发明针对现有技术中存在的三孢布拉霉发酵生产β-胡萝卜素产量较低的不足,提供一种一种基因工程菌生产β-胡萝卜素的方法及其基因工程菌,该基因工程菌生产β-胡萝卜素产量较高,发酵条件简单。

本发明解决上述技术问题的技术方案如下:

本发明的技术方案之一为:一种含有茄红素合酶与番茄红素环化酶(carRA)基因/茄红素脱氢酶(carB)基因的耶罗维解脂酵母(Yarrowia lipotica)基因工程菌。

其中,所述的茄红素合酶/番茄红素环化酶(carRA)基因是常规的,较佳的是来源于三孢布拉霉β-胡萝卜素合成途径的carRA(茄红素合酶与番茄红素环化酶)基因,更佳的,是根据耶罗维解脂酵母的密码子偏好性做相应的优化后的,最佳的其核苷酸见序列表中SEQ ID NO.3所示。

其中,所述的茄红素脱氢酶(carB)基因是常规的,较佳的是来源于三孢布拉霉β-胡萝卜素合成途径的carB(茄红素脱氢酶)基因;更佳的,是根据耶罗维解脂酵母的密码子偏好性做相应的优化后的,最佳的其核苷酸序列见序列表中SEQ ID NO.4所示。

其中,所述的耶罗维解脂酵母基因工程菌的制备方法是常规的,较佳的用基因工程手段构建而得,更佳的包括以下步骤:将含有carRA基因和carB基因重组载体导入耶罗维解脂酵母即可。

较佳的,所述的耶罗维解脂酵母基因工程菌还进一步含有双香儿基焦磷酸合酶(GGS)基因。

其中,所述的双香儿基焦磷酸合酶(GGS)基因是常规的,较佳的来源于耶罗维解脂酵母ATCC20362,更佳的其核苷酸序列如序列表SEQ ID NO.2所示。

其中,所述的耶罗维解脂酵母基因工程菌的制备方法是常规的,较佳的用基因工程手段构建而得,更佳的包括以下步骤:将含有carRA基因、carB基因和GGS基因的重组载体导入耶罗维解脂酵母即可。

其中,所述的含有carRA基因、carB基因和GGS基因的重组载体,较佳的,由包括以下步骤的方法构建而得:克隆来源于三孢布拉霉的carRA基因、carB基因,在其上游和下游连接来源于耶罗维解脂酵母的启动子和终止子,使其成为完整的表达元件,并与GGS基因的表达元件整合到一个质粒上,即获得的重组载体。该重组载体可一次性将基因整合到耶罗维解脂酵母基因组上。较佳的,所述的重组载体是质粒pS-B-RA-GGS-F-U(L)。

进一步更佳的,所述的carB基因,carRA基因和GGS基因在所述的耶罗维解脂酵母基因工程菌中的拷贝数大于1个,较佳的为2-100个,甚至1000个。

其中,获得高拷贝数的carB基因,carRA基因和GGS基因的耶罗维解脂酵母基因工程菌的方法是常规的,较佳的可以导入多拷贝数外源基因的重组表达载体,或者用具有多整体位点的重组表达载体导入外源基因。

进一步更佳的,所述的耶罗维解脂酵母基因工程菌还进一步含有HMG-CoA还原酶的活性部位基因(HMGcata)。

其中,所述的HMG-CoAcata还原酶是常规的,较佳的来自耶罗维解脂酵母的HMG-CoA还原酶,更佳的是HMG-CoA还原酶的催化部位。

其中,所述的耶罗维解脂酵母基因工程菌的制备方法同常规,较佳的,用基因工程手段构建而得,更佳的,包括以下步骤:用含有HMG-CoA还原酶活性部位基因(HMGcata)的重组表达载体导入耶罗维解脂酵母即可。其中,所述的重组表达载体较佳的是质粒pSK-Hmg(y)-Ura3-IntB,其核苷酸序列如序列表中SEQ ID NO.6所示。

进一步更佳的,所述的耶罗维解脂酵母基因工程菌还进一步敲除了3-磷酸甘油脱氢酶(Gut2)基因。所述的敲除方法是常规的,较佳的,包括以下步骤:采用质粒pS-B-RA-GGS-Gut2-U(L)敲除,该质粒的核苷酸序列如序列表中SEQ ID NO.7所示。

进一步更佳的,所述的耶罗维解脂酵母基因工程菌还进一步敲除了过氧化物酶(Pox2)基因。所述的敲除方法是常规的,较佳的包括以下步骤:采用质粒pS-B-RA-GGS-Pox2-U(L)敲除,该质粒的核苷酸序列如序列表中SEQ ID NO.8所示。

本发明中,所述的耶罗维解脂酵母是常规的。

本发明的技术方案之二,一种基因工程菌生产β-胡萝卜素的方法,包括以下步骤:

1)制备含有carRA(茄红素合酶与番茄红素环化酶)基因与carB(茄红素脱氢酶)基因的耶罗维解脂酵母基因工程菌;

2)发酵培养步骤1)所得的耶罗维解脂酵母基因工程菌,从发酵液中获得β-胡萝卜素。

本发明步骤1)所述的耶罗维解脂酵母基因工程菌如上所述。

本发明步骤2)所述的发酵培养骤1)所得的耶罗维解脂酵母基因工程菌的方法的常规的耶罗维解脂酵母发酵方法,在常规的耶罗维解脂酵母发酵液中发酵即可,发酵条件和参数是常规的。较佳的包括在YPD液体培养基中28℃震荡培养所述耶罗维解脂酵母基因工程菌即可。步骤2)所述的从发酵液中获得β-胡萝卜素的方法也是常规的。

在符合本领域常识的基础上,上述各优选条件,可任意组合,即得本发明各较佳实例。

本发明所用试剂和原料均市售可得。

本发明的积极进步效果在于:本发明通过基因工程方法向耶罗维解脂酵母(Yarrowia lipotica)引入来源于三孢布拉霉(Blakeslea trispora)β-胡萝卜素基因茄红素合酶与番茄红素环化酶(carRA)和茄红素脱氢酶(carB)基因,进一步增强双香儿基焦磷酸合酶(GGS)和3-羟基3-甲基乙酰辅酶A还原酶(HMG-CoAcata reductase)的表达,更进一步敲除3-磷酸甘油脱氢酶(Gut2)与过氧化物酶(Pox2),获得了异源表达β-胡萝卜素耶罗维解脂酵母菌株。本发明选择耶罗维解脂酵母为表达宿主,异源表达来自三孢布拉霉的β-胡萝卜素合成途径,发酵条件简单,β-胡萝卜素产量较高,具有重要的生产价值。

具体实施方式

本发明人通过克隆来源于三孢布拉霉β-胡萝卜素合成途径的carRA(茄红素合酶与番茄红素环化酶)与carB(茄红素脱氢酶)基因,在其上游和下游连接来源于耶罗维解脂酵母的启动子和终止子,使其成为完整的表达元件,并与GGS基因的表达元件整合到一个质粒上,一次性将基因整合到耶罗维解脂酵母基因组上,得到异源产β-胡萝卜素的菌株。而后克隆表达HMG-CoAcata,增强甲羟戊酸代谢,使β-胡萝卜素的产量得到大幅提升。敲除3-磷酸甘油脱氢酶(Gut2)与过氧化物酶(Pox2)基因分别降低3-磷酸甘油的产生和过氧化物的β氧化,获得β-胡萝卜素的高产耶罗维解脂酵母菌株。

下面通过实施例的方式进一步说明本发明,但并不因此将本发明限制在所述的实施例范围之中。下列实施例中未注明具体条件的实验方法,按照常规方法和条件,或按照商品说明书选择。

1、材料和方法

应用在实施例中的标准重组DNA和分子克隆技术是本领域熟知的,相关描述可参见:(1)由Cold Spring Harbor Laboratory;Cold Spring Harbor,NY(1989)出版Sambroo,J.Fritsch,E.F.和Maniatis,T.所著的Molecular Cloning:A Laboratory Manual;(2)由Cold Spring Harbor Laboratory:Cold Spring Harbor,NY(1984)出版T.J.Silhavy.M.L.Bennan和L.W.Enquist所著的Experiments with Gene Fusions;(3)由Greene Publishing Assoc.和Wiley Interscience(1987)出版Ausubel,F.M等人所著的Current Protocols in Molecular Biology。

适用于微生物培养的维持和生长的材料和方法是本领域熟知的。适用于下述实施例的技术可参见American Society for Microbiology:Washington,D.C.(1994)出版的Manual of Methods for General Bacterioloy或Thomas D.Brock在Sinauer Associates:Sunderland,MA(1989)出版的第二版Biotechnology:A Textbook of Industrial Microbiology中所述的相关内容。耶罗维解脂酵母的操作参见Barth G,Gaillardin C.1996.Yarrowia lipotica,p 313–388.In Wolf KE(ed),Nonconventional yeasts in biotechnology。

2、耶罗维解脂酵母(Yarrowia lipotica)的培养

耶罗维解脂酵母(Yarrowia lipotica)该菌株一般在28℃下培养,采用的培养基主要有以下几种,固体培养基添加20g/L的琼脂。

YPD培养基:10g/L酵母浸出物(OXOID),20g/L蛋白胨(OXOID),20g/L葡萄糖(国药)。

MM培养基:20g/L葡萄糖,6.7g/L YNB培养基(无氨基酸)。

YNB培养基购自BIOSHARP。

MM+5-FOA培养基:20g/L葡萄糖,6.7g/L YNB培养基(无氨基酸),1g/L 5-FOA(5-氟乳清酸),0.075g/L尿嘧啶,0.075g/L尿苷。

酵母单菌落接种于YPD斜面上,28℃培养72h;接种斜面上的酵母菌落于20ml YPD液体培养基(pH 5.5)中,28℃,200rpm培养72h;取1ml培养液,12000rpm离心1min;弃上清加入1ml丙酮,超声波破碎30min后12000rpm离心;取上清用于液相检测。

3、β-胡萝卜素的HPLC检测

检测方法参见Shao Z,Zhao H,Zhao H(2009)DNA assembler,an in vivo genetic method for rapid construction of biochemical pathways.Nucleic Acid Res 37:e16。采用注射器5μl进样量,检测柱为Agilent Diamonsil SB-C18column,在472nm处采集。乙腈和四氢呋喃洗脱,具体洗脱程序见表1。

实施例1耶罗维解脂酵母基因的导入

该实施例主要描述耶罗维解脂酵母的基因转化方法。

感受态细胞制备过程

1.接种耶罗维解脂酵母单菌落至5ml YPD液体培养基(柠檬酸-柠檬酸钠缓冲液调节pH至4.0)中,28℃,220rpm振荡培养10-12h;

2.将50-100μL上述培养液转移至10ml YPD液体培养基(pH4.0)中,28℃,220rpm过夜培养直至细胞OD600约为5.5左右;

3.培养液28℃,3500rpm离心5min;

4.弃上清,用10ml 1×TE缓冲液冲洗细胞2次;

5.用20ml左右0.1M LiAc(pH6.0)稀释悬浮,使细胞浓度达到5×107/ml;

6.悬浮液在28℃,100rpm轻柔孵育1h;

7.培养液28℃,3500rpm离心5min后弃上清,用2ml 100mM LiAc(pH6.0)重新悬浮,使细胞浓度达到5×108/ml;

注:感受态细胞可以在4℃下保存一周,使用前需离心弃上清后重悬;长时间保存需加25%甘油在-80℃下保存,取用时需在冰上融化,使用前用0.1M LiAc冲洗重悬。

转化过程

1.取5μL carrier DNA(carrier DNA为鲑鱼精DNA,10mg/ml溶于50mM Tris,5mM EDTA溶液中,长度约为500bp)100℃孵育5min放置在冰上,1μg线性化载体置于2mL EP管底部,60℃孵育5min后放在冰上,然后将二者混匀;

2.将100μL感受态细胞与上述DNA混匀,28℃,100rpm孵育15min,然后加700μL 40%PEG4000(用0.1M LiAc溶解,过滤除菌)混匀,28℃,100rpm孵育1h;

3.细胞39℃热激10min,然后加入1.2mL 100mM LiAc,200ml悬浮液涂布在筛选平板上平板上,28℃培养3~4后待菌落长出(充足的O2)。

实施例2URA3缺失株的获得

HMG-CoA还原酶是甲羟戊酸途径中重要的限速酶,催化HMG-CoA到甲羟戊酸的转化。为了增加β-胡萝卜素的产量,我们通过克隆表达HMG-CoA还原酶的催化部位使碳代谢流向类异戊二烯途径。克隆来自耶罗维解脂酵母的HMG-CoA还原酶基因,共3000bp,截取其催化部位1500-3000bp,并在起始部位添加ATG起始密码子。为了使该序列正常表达,在其上游和下游分别添加Exp1启动子和Aco终止子,得到完整的克隆表达元件Exp1-HMG-CoAcata-Aco,然后整合URA3基因的上下游同源片段,得到质粒pSK-HMGcata-URA3.5N-3N,该质粒的序列见SEQ ID NO1(参见表2,图2)。将PmeI/SphI线性化的该质粒转化进入耶罗维解脂酵母ATCC20362(购自ATCC),获得菌株株Y2224。,故而经采用URA3同源臂获得了Y2224,同时导入了催化部位。

实施例3三孢布拉霉β-胡萝卜素基因的表达

该实施例主要描述三孢布拉霉β-胡萝卜素在耶罗维解脂酵母菌株的构建和表达(见图1)。

构建的整合质粒pS-B-RA-GGS-F-U(L)主要包括来源于三孢布拉霉的carB(茄红素脱氢酶),carRA(茄红素合酶和番茄红素环化酶)和GGS基因(焦磷酸双香叶儿基合酶)。

GGS基因克隆于耶罗维解脂酵母ATCC20362,GGS基因具体序列见SEQ ID NO.2。

CarRA基因克隆自三孢布拉霉,根据耶罗维解脂酵母的密码子偏好性做相应的优化,优化后的CarRA基因序列见SEQ ID NO.3。

CarB基因克隆自三孢布拉霉,根据耶罗维解脂酵母的密码子偏好性做相应的优化,优化后的CarB基因序列见SEQ ID NO.4。将上述基因添加启动子与终止子构建功能组件,然后整合IntF同源片段,得到pS-B-RA-GGS-F-U(L)质粒(见表3、图3),该质粒的序列见SEQ ID NO.5。该质粒的同源片段为IntF基因,该质粒整合至耶罗维解脂酵母IntF位点。

将质粒pS-B-RA-GGS-F-U(L)用AscI/SphI酶切,包含表2序列的线性片段,根据实施例1中的方法将该片段转化到耶罗维解脂酵母菌株Y2224(Ura3-)。转化细胞涂布于MM培养基平板上,28℃培养48-72h,得到转化子YH202。将长出的黄色单菌落YH202重新在MM培养基上划线,培养48-72h后接种于YPD斜面上。待斜面生长72h后,转移到20ml YPD液体培养基中,28℃,200rpm震荡培养72h后液相检测,得到该菌株β-胡萝卜素产量为4.35mg/L。

实施例4HMG-CoA还原酶催化部位的克隆及表达

HMG-CoA还原酶是甲羟戊酸途径中重要的限速酶,催化HMG-CoA到甲羟戊酸的转化。为了增加β-胡萝卜素的产量,我们通过克隆表达HMG-CoA还原酶的催化部位使碳代谢流向类异戊二烯途径。克隆来自耶罗维解脂酵母的HMG-CoA还原酶基因,共3000bp,截取其催化部位1500-3000bp,并在起始部位添加ATG起始密码子。为了使该序列正常表达,在其上游和下游分别添加Exp1启动子和Aco终止子,得到完整的克隆表达元件Exp1-HMG-CoAcata-Aco。将该元件连接到表达载体上,得到质粒pSK-Hmg(y)-Ura3-IntB(表4、图4)。该质粒整合至耶罗维解脂酵母Int B位点。该质粒具体序列见SEQ ID NO.6。

将质粒pSK-Hmg(y)-Ura3-IntB用PmeI/SphI酶切,回收含上述序列的片段,根据实施例1中的方法将该片段转化到耶罗维解脂酵母菌株YH202(Ura3-)中。转化细胞涂布于MM培养基平板上,28℃培养48-72h得到转化子YH306。将长出的黄色单菌落重新再MM培养基上划线,培养48-72h后接种于YPD斜面上。待斜面生长72h后,转移到20ml YPD液体培养基中,28℃,200rpm震荡培养72h后液相检测。YH306产量为7.13mg/L,说明过表达HMG-CoAcata能够增强类异戊二烯途径,增加β-胡萝卜素的产量。然后将YH306单菌落接种于20ml YPD培养基中培养24h,涂布于MM+5-FOA平板上,培养48-72h后得到URA3缺失的菌株YH306(Ura3-)。

实施例5敲除Gut2基因增加β-胡萝卜素产量

构建的整合质粒pS-B-RA-GGS-Gut2-U(L)(表5、图5)。主要包括carB(茄红素脱氢酶),carRA(茄红素合酶和番茄红素环化酶)和GGS基因(焦磷酸双香叶儿基合酶)。该质粒的同源臂为Gut2基因的上下游片段,目的是敲除Gut2基因,并且补入第二个carB(茄红素脱氢酶),carRA(茄红素合酶和番茄红素环化酶)和GGS基因(焦磷酸双香叶儿基合酶)。该质粒具体序列见SEQ ID NO.7。

将质粒pS-B-RA-GGS-Gut2-U(L)用AscI/SphI酶切,包含表5序列的线性片段,根据转化方法将该片段转化到耶罗维解脂酵母菌株YH306(Ura3-)中。转化细胞涂布于MM培养基平板上,28℃培养48-72h得到转化子YH413。将长出的黄色单菌落重新再MM培养基上划线,培养48-72h后接种于YPD斜面上。待斜面生长72h后,转移到20ml YPD液体培养基中,28℃,200rpm震荡培养72h后液相检测。YH413的β-胡萝卜素的产量为37.4mg/L。说明敲除Gut2可以降低3-磷酸甘油的产生,提升乙酰CoA的产量,增加β-胡萝卜素的产量。然后将YH413单菌落接种于20ml YPD培养基中培养24h,涂布于MM+5-FOA平板上,培养48-72h后得到URA3缺失的菌株YH413(Ura3-)。

实施例6敲除Pox2基因增加β-胡萝卜素产量

构建的整合质粒pS-B-RA-GGS-Pox2-U(L)(表6、图6)。主要包括carB(茄红素脱氢酶),carRA(茄红素合酶和番茄红素环化酶)和GGS基因(焦磷酸双香叶儿基合酶)。该质粒的同源臂为Pox2基因的上下游片段,目的是敲除Pox2基因,并且补入第二个carB(茄红素脱氢酶),carRA(茄红素合酶和番茄红素环化酶)和GGS基因(焦磷酸双香叶儿基合酶)。该质粒具体序列见SEQ ID NO.8。

将质粒pS-B-RA-GGS-Pox2-U(L)用AscI/SphI酶切,包含表6序列的线性片段,根据实施例1中的方法将该片段转化到耶罗维解脂酵母菌株YH413(Ura3-)中。转化细胞涂布于MM培养基平板上,28℃培养48-72h得到转化子YC511。将长出的黄色单菌落重新再MM培养基上划线,培养48-72h后接种于YPD斜面上。待斜面生长72h后,转移到20ml YPD液体培养基中,28℃,200rpm震荡培养72h后液相检测。菌株YC511的β-胡萝卜素的产量为53.0mg/L,说明敲除Pox2可以降低过氧化物,提升乙酰CoA的产量,增加β-胡萝卜素的产量。然后将YH511单菌落接种于20ml YPD培养基中培养24h,涂布于MM+5-FOA平板上,培养48-72h后得到URA3缺失的菌株YH511(Ura3-)。

实施例7一次性补入多个基因增加β-胡萝卜素产量

本实施例主要描述一次性补入多个carB(茄红素脱氢酶),carRA(茄红素合酶和番茄红素环化酶)和GGS基因(焦磷酸双香叶儿基合酶)对β-胡萝卜素产量的影响。酵母基因组的rDNA有多个100-200个重复单元,以此为整合位点提高整合拷贝数的应用策略在酵母中常被应用。依赖于rDNA的整合可以一次性导入几个甚至几十个的拷贝,从而实现高效表达。构建的整合质粒pS-B-RA-GGS-rDNA-U(L)(表7、图7)具体序列见SEQ ID NO.9。

将质粒pS-B-RA-GGS-rDNA-U(L)用AscI/SphI酶切,根据实施例1中的方法将该片段转化到耶罗维解脂酵母菌株YH504(Ura3-)中。转化细胞涂布于MM培养基平板上,28℃培养48-72h得到YC607。将长出的黄色单菌落重新再MM培养基上划线,培养48-72h后接种于YPD斜面上。待斜面生长72h后,转移到20mlYPD液体培养基中,28℃,200rpm震荡培养72h后液相检测。YC607的β-胡萝卜素的产量为72.9mg/L。
