技术领域
本发明属于基因工程技术领域,具体地说涉及一种基因。
背景技术
胰岛素样生长因子(IGFs)是一类多功能的细胞增殖因子,具有广泛的生物学效应,在胚胎和个体发育过程中起着重要作用。IGFs通过细胞表面两种类型的IGF受体发挥生物学作用。IGFs受体有两种,分别称为IGF-I受体(IGF-IR或称I型IGF受体)和IGF-II受体(IGF-IIR或称II型IGF受体)。IGFs对细胞的作用绝大多数都是通过与IGF-IR结合而实现的。IGF-IR为四聚体糖蛋白,由2个位于细胞外部的α亚基和2个跨膜β亚基组成。IGF-IR的结构与胰岛素受体极其相似。α亚基的半胱氨酸富集区是其配体的特异识别位点,细胞质部分的β链含有一个酪氨酸激酶活性区。一个α亚基和一个β亚基由二硫键连接起来形成一个半受体,两个半受体由α亚基上的二硫键构成一个完整的受体。IGF-I与IGF-IR结合会引发受体上位于细胞内的β亚基上的酪氨酸和丝氨酸残基的磷酸化,进而导致一系列的生物学效应。IGFs在鱼类的生长繁殖上起着重要作用,而仅仅对IGFs的研究是不足的,应该进一步了解IGFs与其受体的相互作用,这对于认识IGF的分子作用机理具有重要意义。而现有技术中,对IGFs受体的研究主要集中在哺乳动物中,对于鱼类的IGFs受体的研究很少。
石斑鱼是海洋珊瑚礁鱼类,属鮨科(Serranidae),石斑鱼属(Epinephelus),有30多种。石斑鱼是一种名贵的海水鱼类,具有极高的经济价值。石斑鱼的苗种培育是养殖生产持续发展的关键,但目前在石斑鱼人工养殖中还没有一种能有效促进其苗种生长的饵料添加剂。

发明内容
本发明的目的在于针对目前石斑鱼的苗种培育问题,提供一种重组石斑鱼胰岛素样生长因子I受体基因,利用该基因可以生产重组石斑鱼胚胎期胰岛素样生长因I受体蛋白,该受体蛋白能够有效促进石斑鱼苗种的生长,在制备鱼苗促生长剂或添加剂中有着重要的应用。
本研究对斜带石斑鱼IGFs受体的cDNA进行克隆和序列分析,以完善IGFs与其受体作用的调控理论,为IGFs系统用于鱼类养殖、促进鱼类生长与繁殖提供理论基础。
氨基酸序列分析表明石斑鱼胰岛素样生长因子I受体前蛋白原氨基酸全序列为1413aa,成熟肽为1384aa。使用Clustalx算法,将斜带石斑鱼IGF-IR前蛋白原氨基酸全序列与其它物种的IGF-R序列进行同源性比较,发现两个主要的功能区即富含亮氨酸区和酪氨酸激酶区的保守性非常高,羧基端的变异性比较大,本发明的石斑鱼IGF-IR序列属于b型。
本发明的石斑鱼胰岛素样生长因子I受体基因是以石斑鱼下丘脑总RNA反转录得到的cDNA为模板,经PCR和Gene RACER方法扩增而得到的。
本发明用于表达石斑鱼IGF-IR重组蛋白的IGF-IR成熟肽基因序列是以石斑鱼胰岛素样生长因子I受体基因双链DNA为模板,经PCR方法扩增而得到的,它来源于石斑鱼胰岛素样生长因子I受体基因的α亚基所对应的基因片段。
本发明还构建了上述表达石斑鱼IGF-IR重组蛋白的IGF-IR成熟肽基因序列的载体;特别是含有该基因的大肠杆菌克隆载体以及表达载体。这些载体的构建方法是按常规方法,将通过PCR方法合成的斜带石斑鱼胰岛素样生长因子I受体基因经酶切和分离纯化后,接入到已有的相应载体的相应酶切位点之间,即构建成所需的含有石斑鱼胰岛素样生长因子I受体基因的表达载体。
上述含石斑鱼胰岛素样生长因子I受体基因的大肠杆菌表达载体优选由本发明合成的石斑鱼胰岛素样生长因子I受体基因与大肠杆菌表达载体pET15b构建成的重组表达载体,命名为pET15b-IGF-IR。
本发明还用上述含有石斑鱼胰岛素样生长因子I受体基因的相应表达载体分别构建了能高效表达石斑鱼胰岛素样生长因子I受体基因的大肠杆菌重组株。
本发明的上述能表达石斑鱼胰岛素样生长因子I受体大肠杆菌重组菌株优选由含有石斑鱼胰岛素样生长因子I受体基因的表达载体pET15b-IGF-IR转化大肠杆菌BL21而得到的菌株,命名为pET15b-IGF-IR-BL21。
本发明还提供了利用上述大肠杆菌重组株生产石斑鱼胰岛素样生长因子I受体的方法。先将将菌种接种在含有氨苄青霉素的LB液体培养基中,37℃,200转/分培养过夜,次日接种于相同培养基中,并经IPTG诱导,37℃,200转/分培养,10小时后收集菌体,经分离纯化得到重组石斑鱼胰岛素样生长因子I受体产品。
本发明的有益效果石斑鱼具有重大的经济价值,采用常规的基因工程技术,利用本发明的石斑鱼胰岛素样生长因子I受体基因,可以生产重组石斑鱼IGF-I受体蛋白,用于制备鱼苗苗种促生长剂或添加剂,促进了鱼苗的快速生长,具有重大的经济效益。

具体实施例方式
实施例1石斑鱼胰岛素样生长因子I受体基因中间片段的合成根据已经在GeneBank里登录的十二种物种的IGF-IR基因序列,用Dnassist软件对它们进行相似性比较,在保守区域设计合成一套用于嵌套PCR扩增的简并引物。正义引物Primer17035’T(G/T)AA(A/G)CC(C/T)TGGAC(A/C/T)CA(A/G)TA(C/T)GC3’(22bp);
反义引物Primer29355’CACTGAA(A/G)TA(C/T)TC(A/C/G/T)GG(A/G)TT(A/C/G)AC3’(22bp);反义引物Primer31005’TC(A/G)TT(A/C/G/T)AC(A/C/T)GTCTT(A/G/T)ATGGC3’(20bp)。反应条件为95℃预变性5分钟;下边为38个循环,分为两个阶段(1)95℃变性30秒,从50℃向37℃每两个循环降1℃,退火30秒,共28循环,72℃延伸1分钟;(2)95℃变性30秒,37℃退火30秒,共8循环,72℃延伸1分钟;最后72℃延伸7分钟。第一轮PCR结果Primer1703与Primer3100扩增出一条约1.4kb的条带,嵌套PCRPrimer1703与Primer2935扩增出一条约1.25kb的条带,大小与预期的石斑鱼IGF-IR cDNA中间片段相一致,如图1。
将1.25kb的PCR回收片段连接到pGEM-Teasy(Promega公司)载体上,转化DH5α后蓝白斑筛选重组子,随机挑选了10个重组子扩大培养,PCR鉴定后发现8个阳性克隆。PCR扩增产物的电泳鉴定图见图2。提取阳性质粒后进一步用EcoRI酶切鉴定质粒,选择PCR鉴定和酶切鉴定均为阳性的质粒进行测序。登录http//www.ncbi.nlm.nih.gov/网站,利用BLAST软件对已获得的序列和Genebank里已知的其它物种IGF-IR的序列进行同源性比较分析,结果表明所得的序列为石斑鱼的IGF-IR cDNA中间片段。
实施例2石斑鱼胰岛素样生长因子I受体基因5’端片段的合成操作按GeneRacer Kit Protocol来进行。根据试剂盒的要求,依据已测序的IGF-IR cDNA中间片段序列设计了两条特异性下游引物反义引物Primer 5R3365’CCTCTTCCTGGTCTCCCACACCTATG3’(26bp),嵌套反义引物Primer 5R104n5’GGTGCGGATGTAGACCACTTTGC3’(23bp)。
根据GeneRacer Kit试剂盒的通用引物和这两条特异性下游引物做嵌套PCR,最后结果得到一条分子量约为2.4kb的条带。第一次PCR反应条件为95℃预变性5分钟;下边为40个循环,分为两个阶段(1)95℃变性30秒,从62℃向47℃每个循环降0.5℃,退火30秒,共32循环,72℃延伸1分钟;(2)95℃变性30秒,46℃退火30秒,共8循环,72℃延伸1分钟;最后72℃延伸7分钟。第二次PCR以第一次PCR产物为模板,反应条件为95℃预变性5分钟;下边为35个循环95℃变性30秒,60℃退火30秒,72℃延伸1分钟30秒;最后72℃延伸7分钟。二次PCR扩增产物的电泳鉴定图如图3和图4所示。
将2.4kb的PCR回收片段连接、转化后挑选了12个重组子,PCR鉴定后有6个阳性克隆,如图5,进一步用EcoRI酶切鉴定质粒,选择PCR鉴定和酶切鉴定均为阳性的质粒进行测序。利用BLAST软件对已获得的2.4kb序列和Genebank里已知的其它物种IGF-IR的序列进行同源性比较分析,结果表明所得的序列为石斑鱼IGF-IR cDNA的5’片段。
实施例3石斑鱼胰岛素样生长因子I受体基因酪氨酸激酶区片段的合成根据已得到的中间片段和已报道的其它物种的IGF-IRβ亚基的高度保守区域-酪氨酸激酶活性区的序列,设计两条特异性正义引物和一条简并反义引物,获取石斑鱼IGF-IR酪氨酸激酶活性区片段。正义引物Primer 3R7075’GCCGTTCACAGTTTACCGCATCG3’(23bp),正义引物Primer 3R1109n5’CGTCCTCTACGCTATGATCTTCG3’(23bp),反义引物Primer3RYR15’GGCTTGTTSTCMKCRCTGTAG3’(21bp),第一次PCR反应条件为95℃预变性,5min,下边为30个循环,94℃变性,30sec;50℃-65℃退火,30sec;72℃延伸,1min30sec,最后72℃延伸,10min。第二次PCR以第一次PCR 52℃退火扩增的产物稀释50倍做为模板,反应条件为95℃预变性,5min,下边为30个循环,94℃变性,30sec;48℃-68℃退火,30sec;72℃延伸,1min,最后72℃延伸,10min。二次PCR扩增产物的电泳鉴定图如图6和图7所示。
将1.1kb的PCR回收片段连接、转化后挑选了9个重组子,PCR鉴定后有8个阳性克隆(图8),进一步用EcoRI酶切鉴定质粒,选择PCR鉴定和酶切鉴定均为阳性的质粒进行测序。利用BLAST软件对已获得的1.06kb序列和Genebank里已知的IGF-IR的序列进行同源性比较分析,结果表明所得的序列为石斑鱼IGF-IR cDNA酪氨酸激酶活性区片段。
实施例4石斑鱼胰岛素样生长因子I受体基因3’端片段的合成根据获得的酪氨酸激酶活性区片段序列和几种鱼类的IGF-IR 3’非编码区序列,设计合成了两条3’特异性引物和与之配对的3’非编码区的特异性引物。正义引物Primer 3R37005’GTGTTGGCAGTACAATCCTAAG3’(22bp),正义引物Primer3R3500n5’AGGGTTTGCTTCCTGTCCGATG3’(22bp),反义引物Primer 3R60005’CCCACTGTGACATCAGGTTC3’(20bp)。第一次PCR反应条件为95℃预变性,5min,下边为35个循环,94℃变性,30sec;48℃-68℃退火,30sec;72℃延伸,1min,最后72℃延伸,10min。第二次PCR以第一次PCR 60℃退火扩增的产物稀释50倍做为模板,反应条件为95℃预变性,5min,下边为30个循环,94℃变性,30sec;48℃-68℃退火,30sec;72℃延伸,1min,最后72℃延伸,10min。二次PCR扩增产物的电泳鉴定图如图9和图10所示。
将560bp的PCR回收片段连接、转化后挑选了9个重组子,PCR鉴定后有9个阳性克隆,如图11,阳性率为100%,进一步用EcoRI酶切鉴定质粒,选择PCR鉴定和酶切鉴定均为阳性的质粒进行测序。利用BLAST软件对已获得的560bp序列和Genebank里已知的IGF-IR的序列进行同源性比较分析,结果表明所得的序列为石斑鱼IGF-IRcDNA的3’片段。
实施例5石斑鱼胰岛素样生长因子I受体基因全长序列的拼接将测序得到的正确IGF-IR中间片段、5’端片段、酪氨酸激酶活性区片段、3’端片段进行拼接得到石斑鱼IGF-IR cDNA全长序列,包括5’端595bp的非编码区,完整的α亚基,β亚基酪氨酸激酶保守区,3’非编码区71bp,总长度4908bp。开放阅读框4242bp,起始密码子从596bp处开始,到4837bp处的终止密码子结束。用DNAsist软件推测出相应蛋白质的氨基酸序列,长度为1413aa,包括29aa的信号肽和1384aa的成熟肽,分为N-末端区、富含半胱氨酸区、蛋白裂解位点、跨胞膜区、酪氨酸激酶区(TK)和羧基末端区等几个功能区。IGF-I受体基因全长和推测的氨基酸序列见序列表。
实施例6石斑鱼胰岛素样生长因子I受体基因的合成根据在克隆IGF-IR 5’端序列时获得的包括完整α亚基部分的质粒序列,结合Pet-15b载体的多克隆位点,设计合成一对扩增α亚基片段的特异性引物。正义引物Primer Sen135’ACGCTCGAGCGACCCAGCATTGACATC3’(27bp),反义引物Primer Anti17025’ATGGATCCGTGGCACCCGGCAAATG3’(25bp),以克隆IGF-IR5’端序列时获得的包括完整α亚基部分的质粒(1∶100)为模板,扩增IGF-IR-α亚基片段。PCR反应条件为95℃预变性4分钟;下边为30个循环94℃变性30秒,60℃退火30秒,72℃延伸1.5分钟;最后72℃延伸7分钟。PCR扩增后出现约1.6kb的亮带,与预期大小相一致,连接进pGEMT-easy载体(Promega公司)后取PCR鉴定和酶切鉴定均为阳性的克隆提取质粒,该质粒为pGEMT-IGF-IR。
实施例7含石斑鱼胰岛素样生长因子I受体基因的大肠杆菌表达载体pET15b-IGF-IR的构建用BamHI和XhoI对分别Pet-15b(Novagen公司)和pGEMT-IGF-IR(实施例6所得)两种载体进行双酶切,将分别得到的5.7kb和1.6kb预期大小片段回收纯化,T4 ligase(Takara公司)连接后转化到DH5α,筛选阳性克隆提取质粒,送上海博亚生物公司测序,测序结果证明IGF-IR基因成功转进表达载体Pet-15b中,重组载体pET15b-IGF-IR成功构建,质粒构建图见图13。
实施例8能高效表达石斑鱼胰岛素样生长因子I受体基因的大肠杆菌重组株pET15b-IGF-IR-BL21的构建CaCl2法将pET15b-IGF-IR转化E.coli BL21,在含氨苄青霉素的LB平板上筛选转化子,经质粒检测和酶切分析获得含pET15b-IGF-IR的重组转化子pET15b-IGF-IR-BL21。
实施例9利用大肠杆菌基因工程菌pET15b-IGF-IR-BL21生产重组石斑鱼胚胎期胰岛素样生长因I受体挑取大肠杆菌基因工程菌pET15b-IGF-IR-BL21单菌落接种在含有100ug/ml氨苄青霉素的LB液体培养基中,37℃,200转/分培养过夜,次日按1∶50接种量接种于适当体积的相同培养基中,37℃培养至A600为0.5~0.6,加入IPTG(终浓度为1.0mmol/l),于37℃诱导培养10小时后收菌。合成的石斑鱼胰岛素样生长因子I受体在大肠杆菌基因工程菌pET15b-IGF-IR-BL21中已获得高效表达。
取少量细胞加2×电泳上样缓冲液,煮沸5分钟后按标准方法跑SDS-PAGE凝胶电泳。结果见图12,显示经诱导的pET15b-IGF-IR-BL21在约61kD的位置上出现一条新的蛋白带,未诱导的pET15b-IGF-IR-BL21不出现此带,证明石斑鱼胚胎期胰岛素样生长因子I受体已在pET15b-IGF-IR-BL21中诱导表达。
将重组石斑鱼胚胎期胰岛素样生长因子I受体采用免疫印迹(Western blot)方法进行免疫活性鉴定。使用Pet-15b本身序列His-tag的抗体作为一抗,羊抗鼠IgG-HRP做为二抗。结果见图12,显示重组蛋白具有特异的免疫活性。

本发明公开了一种石斑鱼胰岛素样生长因子I受体基因,该基因是以石斑鱼下丘脑总RNA为模板,经RT-PCR和Gene RACER方法而得到的具有石斑鱼胰岛素样生长因子I受体基因cDNA序列的基因片段。利用本发明可以获得来源稳定、成本便宜的石斑鱼胰岛素样生长因子I受体的重组蛋白,在制备鱼苗促生长剂或添加剂中有着重要的应用。
