技术领域
本发明涉及利用分子和细胞生物学技术筛选PPAR拮抗剂和激动剂的方法,具体涉及利用酵母双杂交技术,通过测定α-半乳糖苷酶的活性大小筛选PPAR拮抗剂和激动剂的方法。
背景技术
近年来,在我国随着人们生活水平质量的不断提高,饮食结构及生活方式的变化,患有血脂代谢异常的人数在逐年升高,由此而引发的高血压病、高脂血症、冠心病、糖尿病、肥胖症等“富裕性疾病”的发病率亦明显上升,同时,这类疾病还是威胁中老年人健康的第一大杀手,已经引起了全社会的高度重视。有资料显示,目前,高脂血症、糖尿病、肥胖症等相关性十分密切的病症,其相加总发病率已达10%。我国90年代初期与80年代相比,人群血脂水平明显增加,尤其在北方大城市,约30~40%的人患有不同程度的超过边缘性标准的血脂和血糖代谢异常。因此,在全社会范围内重视、控制这类疾病已势在必行。
近十年来的研究表明,核受体PPAR(Peroxisome Proliferator-activated Receptors)又称过氧化物酶体增长因子活化受体,是一类由配体激活的核转录因子,属II型核受体超家族成员,能被脂肪酸以及外源性过氧化物酶体增殖剂(Peroxisome Proliferator,PP)激活。在两栖类、啮齿类动物及人类等PPARs均有3种亚型,即PPARα、PPARβ(亦称PPARδ或NUC1)和PPARγ。人类PPARα、β和γ分别含有468、441和479个氨基酸残基。作为甾体激素受体超家族成员之一,PPAR共有6个区域(A-F)、4个功能结构域。位于受体分子中部的C区是DNA结合区(DBD);羧基端的E/F区是配基结合区(LBD),在激素信号转换成转录激活信号过程中发挥重要作用;氨基端的A/B是调节区,PPARγ的活化可被MAPK介导的A/B区的一个丝氨酸残基的磷酸化而降低,而PPARα的磷酸化可正调节受体-配体的亲和力。活化的PPAR与视黄酸类受体(RXR)形成异二聚体,然后与所调节基因的启动子上游的过氧化物酶体增殖物反应元件(PPRE)结合而发挥转录调控作用。同时,活化的PPAR能同一系列转录共激活因子(如CBP SRC-1;RIP140;TIF1;TIF2;SUG1;P300)结合,从而激活转录。PPAR介导的效应主要包括脂类代谢、免疫反应等,因此,动脉粥样硬化、肥胖、糖尿病、肿瘤以及炎症性疾病等的发生发展与PPAR有密切关系。
PPAR的不同亚型具有不同的生物学作用,PPARα在肝脏脂类代谢中发挥重要作用,PPARβ在脂肪细胞分化中的作用还存在着争议,而PPARγ参与脂肪形成和免疫反应,与脂肪细胞分化、肥胖及胰岛素抵抗关系密切,被认为是治疗糖尿病、肥胖症、坏脂血症、炎症、高血压和癌症药物设计的重要靶标分子,成为近年来研究热点。
对于PPARγ激动剂的研究主要集中在其在治疗II型糖尿病方面的应用。胰岛素增敏剂噻唑烷二酮类药物(TZDs)作为PPARγ的一类激动剂可以明显降低白色脂肪组织(WAT)中甘油三酯的含量,同时还可降低血浆中的葡萄糖,脂质和II型糖尿病实验动物的胰岛素水平。三类TZD类药物罗格列酮rosiglitazone(Rezulin),皮格列酮piogitazone(ACTOS),和曲格列酮Troglitazone(Avandia)已被FDA认证通过,作为治疗II型糖尿病的药物已经在市场上销售。至于PPARγ拮抗剂,理论上可减少脂肪、防治肥胖,但临床尚未应用。
目前国内在PPARγ激动剂和拮抗剂的高通量筛选上大都局限于蛋白质水平的小分子化合物的筛选,如SPR技术,而细胞水平的筛选则基本依赖于哺乳动物细胞,成本较高,操作也相对烦琐。酵母双杂交技术是一项采用报告基因来检测蛋白质与蛋白质间相互作用的技术(美国专利No.5,283,173;Proc.Natl.Acad.Sci.,USA,vol.88,pp.9578-9582,1991)。许多转录因子可以分为DNA结合域(DNA binding domain,DNA-BD)和转录激活域(transcriptional activation domain,AD),而只有DNA-BD和AD这两个结构域共存的情况下,才能激活下游基因的表达。酵母双杂交系统中,将待检测的蛋白X、Y分别与DNA-BD、AD表达为融合蛋白DNA-BD-X、AD-Y,只有在X、Y有相互作用的情况下,DNA-BD、AD方形成完整的复合物,激活下游报告基因的表达。目前通常选用LacZ作为酵母双杂交的报告基因,然而β-半乳糖苷酶是一种胞内酶,因此酵母细胞的裂解彻底与否会对酶活的测定造成直接影响,且操作相对复杂。

发明内容
本发明的目的在于提供一种利用酵母双杂交系统来筛选PPAR拮抗剂和激动剂的新方法。
本发明的另一个目的是提供一种通过测定α-半乳糖苷酶的活性大小,来方便而快捷的检测小分子化合物对PPAR与共激活因子间相互作用影响的方法。
本发明的再一个目的是提供用于本发明方法筛选PPAR拮抗剂和激动剂的酵母筛选系统。该系统包括(1)小鼠共激活因子CBP和酵母转录因子GAL4-AD融合蛋白表达质粒pGADT7-CBP(Escherichia E.coli DH5α)保藏号为CGMCC No.1076,保藏日期2003年12月15日,地址北京中关村中国微生物菌种保藏管理委员会普通微生物中心,邮编100080;(2)含人PPARγLBD和酵母转录因子GAL4-BD融合蛋白表达质粒pGBKT7-PPARγLBD(Escherichia E.coli DH5α)保藏号为CGMCC No.1077,保藏日期2003年12月15日,地址北京中关村中国微生物菌种保藏管理委员会普通微生物中心,邮编100080;(3)宿主酵母AH109。
本发明采用了酵母双杂交这一有利的研究蛋白质与蛋白质之间相互作用的技术,在PPARs与共激活因子的结合是配体依赖性前提下,实现了PPAR的激动剂和拮抗剂的筛选。本发明采用GAL4转录因子,其DNA-BD、AD区域则已分别构建在酵母表达质粒pGBKT7与pGADT7中。本发明采用酵母AH109基因组自带的MEL1基因作为双杂交的报告基因,通过测定其分泌型的产物α-半乳糖苷酶的活性来检测小分子化合物对PPARγ与其共激活因子相互作用程度的影响。
本发明利用酵母双杂交系统来筛选PPAR拮抗剂与激动剂的方法由下列步骤组成1、CBP和酵母GAL4AD融合蛋白(pGADT7-CBP)的质粒构建;本步骤是借助细胞分子生物学方法,选用pGADT7载体构建CBP和酵母GAL4AD融合蛋白的质粒(pGADT7-CBP)。
(1)用PCR技术从CMV-mCBP中获得CBP(1-464aa)的DNA片段引物FW5’AACATATGATGGCCGAGAACTTGCTGGACG 3’RV5’AAGGATCCCTGTTGCCCTGCACCAACAG 3’95℃变性8min,开始循环95℃ 1min,65℃ 1min,72℃ 1min30s,重复30循环,72℃ 10min。
采用琼脂糖凝胶电泳鉴定PCR产物,回收DNA。
(2)将CBP(1-464aa)的PCR产物克隆到pGADT7载体上CBP(1-464aa)的PCR产物和pGADT7分别用Nde I和BamHI酶切;用试剂盒回收酶切产物,其中回收CBP(1-464aa)的PCR酶切产物近1400bp,pGADT7酶切产物近8.0kb。用T4DNA连接酶连接;连接产物转化入感受态细胞DH5α;通过酶切鉴定挑选正确的克隆。
2、PPARγLBD和酵母GAL4BD融合蛋白(pGBKT7-PPARγLBD)的质粒构建;本步骤是借助细胞分子生物学方法,选用pGBKT7载体构建PPARγLBD和酵母GAL4DNA-BD融合蛋白的质粒(pGBKT7-PPARγLBD)。
(1)利用PCR技术从pCDNA3.1-PPARγ中获得PPARγLBD(193aa-475aa)的DNA片段引物FW5’AACATATGGCGGAGATCTCCAGT 3’RV5’AAGTCGACCTAGTACAAGTCCTT 3’95℃变性5min,开始循环95℃ 30s,60℃ 30s,72℃ 1min,重复30循环,72℃10min。
采用琼脂糖凝胶电泳鉴定PCR产物,回收DNA。
(2)将PPARγLBD的PCR产物克隆到pGBKT7载体上PPARγLBD的PCR产物和pGBKT7分别用Nde I和Sal I酶切;用试剂盒回收酶切产物,其中回收PPARγLBD的PCR酶切产物近850bp,pGBKT7酶切产物近7.3kb;用T4DNA连接酶连接;连接产物转化入感受态细胞DH5α;通过酶切鉴定挑选正确的克隆。
3、将质粒pGBKT7-PPARγLBD和pGADT7-CBP依次转入酵母细胞AH109,得到酵母PPARγLBD-CBP_AH109(Saccharomyces cerevisiae AH109)保藏号为CGMCC No.1075,保藏日期2003年12月15日,地址北京中关村中国微生物菌种保藏管理委员会普通微生物中心,邮编100080采用LiAc转化法。先将pGBKT7-PPARγLBD质粒转入酵母细胞AH109,用色氨酸缺陷(T-)的培养基筛选阳性克隆,然后将pGADT7-CBP质粒转入含有pGBKT7-PPARγLBD质粒的酵母中,用色氨酸,亮氨酸双缺陷(T-L-)的培养基筛选阳性克隆,得到同时含有pGADT7-CBP质粒和pGBKT7-PPARγLBD质粒的酵母细胞。
4、通过测定α-半乳糖苷酶的活性筛选化合物在得到的含pGADT7-CBP和pGBKT7-PPARγLBD双质粒的阳性酵母细胞中分别加入阳性对照物、阴性对照物及待筛选的化合物,孵育后测定α-半乳糖苷酶的活性,并计算其激动率,根据各个待筛选药物的激动率的比较,筛选出PPAR拮抗剂和激动剂。
本发明提供了利用酵母双杂交系统定量检测小分子化合物对蛋白质相互作用强度影响方法采用酵母基因组自带的MEL1基因作为双杂交的报告基因,通过测定其分泌型的产物α-半乳糖苷酶的活性来检测小分子化合物对PPARγ与其共激活因子相互作用程度的影响。

有益效果1、利用本发明可进行PPARγ拮抗剂和激动剂的高通量筛选,本发明采用酵母基因组自带的MEL1基因作为双杂交的报告基因,通过测定其分泌型的产物α-半乳糖苷酶的活性来检测小分子化合物对PPARγ与其共激活因子相互作用程度的影响,无需裂解酵母细胞,是一种非常方便快捷且准确的方法,加之酵母细胞的易操作性有利于实现真核水平的高通量筛选。
2、利用本发明,首次在酵母水平进行了已知PPARγ激动剂-罗格列酮EC50的测定。
3、利用本发明可实现PPARγ拮抗剂和激动剂的高通量筛选,为药物的结构修饰和天然先导化合物的发现提供实验基础。

实施例1 CBP和酵母GAL4AD融合蛋白(pGADT7-CBP)的质粒构建(一)试验方法1)以CMV-mCBP为模板,用PCR技术复制扩增获得CBP(1-464aa)的DNA片段(基因序列依据S66385(GENEBANK))引物设计FW5’AACATATGATGGCCGAGAACTTGCTGGACG 3’RV5’AAGGATCCCTGTTGCCCTGCACCAACAG 3’PCR扩增反应(100μl)CMV-mCBP模板2μl;10×buffer10μl;dNTP(2.5mM)8μl;FW4μl;RV4μl;pyrobestTMDNA聚合酶1μl;水71μl。
反应条件为95℃变性8min,开始循环95℃ 1min,65℃ 1min,72℃ 1min30s,重复30循环,72℃ 10min。
采用琼脂糖凝胶电泳鉴定PCR产物,回收DNA。
2)将CBP(1-464aa)的PCR产物克隆到pGADT7载体上CBP(1-464aa)的PCR产物和pGADT7分别用Nde I和BamHI酶切;用试剂盒方法回收酶切产物,其中回收CBP(1-464aa)的PCR酶切产物近1400bp;pGADT7酶切产物近8.0kb;用T4DNA连接酶连接;连接产物转化入感受态细胞DH5α;涂布在含氨苄西林(100mg/l)的平板上;用酶切鉴定挑选正确的克隆。
(二)试验结果1、采用琼脂糖凝胶电泳鉴定PCR产物的结果见图2。
2、酶切鉴定结果在载体上1480bp,2280bp处有两个Hind III的酶切位点,1480bp-2280bp间存在着多克隆位点,CBP就克隆在这个区域内,因而正确的克隆应切出约2200bp和7.2kb左右的片段。如图3所示,经进一步酶切鉴定结果显示确实有正确的克隆(5号和9号)。
3、DNA测序报告pGADT7-CBP表达载体测序报告如图4所示,pGADT7-CBP DNA测序结果和CBP(1-464aa)原DNA序列比较,原序列1131处是一个未知碱基N,5号在该处测出的是A,但这个位置是密码子的第三位,所以无论是AGCT哪种,都不影响氨基酸编码。
实施例2 PPARγLBD和酵母GAL4BD融合蛋白(pGBKT7-PPARγLBD)的构建(一)试验方法以pCDNA3.1-PPARγ为模板,用PCR技术复制扩增获得PPARγLBD(193aa-475aa)的DNA片段(基因序列依据NM005037(GENEBANK))引物设计FW5’AACATATGGCGGAGATCTCCAGT 3’RV5’AAGTCGACCTAGTACAAGTCCTT 3’PCR扩增反应(100μl)pCDNA3.1-PPARγ模板2μl;10×buffer10μl;dNTP(2.5mM)8μl;FW4μl;RV4μl;pyrobest聚合酶1μl;水71μl。
反应条件为95℃变性5min,开始循环95℃ 30s,60℃ 30s,72℃ 1min,重复30循环,72℃ 10min。
采用琼脂糖凝胶电泳鉴定PCR产物,回收DNA。
2)将PPARγLBD的PCR产物克隆到pGBKT7载体上PPARγLBD的PCR产物和pGBKT7分别用Nde I和Sal I酶切;用试剂盒方法回收酶切产物,其中回收PPARγLBD的PCR酶切产物近850bp;pGBKT7酶切产物近7.3kb;用T4DNA连接酶连接;连接产物转化入感受态细胞DH5α;涂布在含卡那霉素(50mg/l)的平板上用酶切鉴定,挑选正确的克隆。
(二)试验结果1、采用琼脂糖凝胶电泳鉴定PCR产物的结果见图6。
2、酶切鉴定结果在载体上6313bp处有一个EcoRV的酶切位点,PPARγLBD基因内部16bp处也有一个EcoRV的酶切位点,因而正确的克隆应切出约2200bp和6kb左右的片段。如图7所示,经进一步酶切鉴定结果显示确实有正确的克隆(8号)。
3、DNA测序报告pGADT7-CBP表达载体测序报告如图8所示,PPARγLBDDNA测序结果和PPARγLBD原DNA序列比较,结果完全一致。
实施例3 将pGADT7-CBP与pGBKT7-PPARγLBD分步转入酵母细胞AH109中
(一)试验方法1)按LiAC法将质粒pGBKT7-PPARγLBD转入酵母细胞AH109接种酵母菌株AH109于约3ml YPDA中,30℃、250rpm培养16-18h(OD600约为1.4);菌液按1∶5转接于新鲜的YPDA中,30℃、200rpm培养5-7h(OD600至0.8~1.2);取10ml菌液,4000rpm*5min离心收集酵母细胞;弃上清,1ml无菌水重悬,转移入另一离心管中,再加入约10ml无菌水,混匀;4000rpm*5min再次离心收集酵母细胞;弃上清,1ml无菌水重悬,转移入1.5ml eppendorf管中;4000rpm*5min再次离心收集酵母细胞,小心吸去上清;依次加入240μl PEG 3500(50%W/V)、36μl 1.0M LiAc(pH7.5)、5μl煮沸的SS-carrier DNA(10mg/ml)、60μl无菌水和10μl待转化质粒。
剧烈振荡1min;30℃静置30min;42℃水浴静置30min,每隔5min颠倒混匀一次;4000rpm*5min离心收集转化的酵母细胞;吸去上清,1ml无菌水重悬,取200μl涂于涂布于色氨酸缺陷(以下简称为T-)的选择平板上,30℃培养3-4d;2)鉴定携带pGBKT7-PPARγLBD质粒的阳性克隆以上T-选择平板上长出的克隆中,挑取1-3号至T-液体培养基中,30℃、250rpm培养16-18h;收集酵母,提取质粒,PCR鉴定所提质粒引物序列FW5’GTAATACGACTCACTATAGGGCGA 3’RV5’TCCGAGTCACTTTAAAATTTGTAT 3’PCR扩增反应(20μl)模板(所提质粒)14μl;10×buffer2μl;dNTP(2.5mM)2μl;FW1μl;RV1μl;taq DNA polymerase0.5μl。
反应条件为95℃变性5min,开始循环95℃ 45s;50℃ 45s;72℃ 1min,重复35循环,72℃变性10min。
3)PCR鉴定得到拥有pGBKT7-PPARγLBD质粒的阳性克隆1,结果如图9所示。
4)将质粒pGADT7-CBP转入到上述1号克隆中接种1号克隆于约3ml T-液体培养基中,30℃、250rpm培养16-18h(OD600约为1.4);菌液按1∶5转接于新鲜的T-液体培养基中,30℃、200rpm培养5-7h(OD600至0.8~1.2);
按以上所述步骤转化1号克隆,最终涂布于色氨酸、亮氨酸双缺陷(以下简称为T-L-)平板,30℃培养3-4d;5)PCR鉴定质粒pGADT7-CBP是否转入1号克隆挑取T-L-平板上的1-3号克隆至T-L-液体培养基中,30℃、250rpm,培养16-18h后提质粒(方法同前),PCR鉴定引物序列FW5’GTAATACGACTCACTATAGGGCGA 3’RV5’AGATGGTGCACGATGCACAGTT 3’PCR扩增反应(20ul)模板(所提质粒)14μl;10×buffer2μl;dNTP(2.5mM)2μl;FW1μl;RV1μl;taq DNA聚合酶0.5μl。
反应条件95℃变性5min,开始循环95℃ 45s;50℃ 45s;72℃ 1min,重复35循环;72℃变性10min6)鉴定得到拥有pGBKT7-PPARγLBD&pGADT7-CBP两个质粒的阳性克隆2,结果如图10所示。
实施例4 筛选化合物1、罗格列酮(阳性化合物)对CBP与PPARγ相互作用影响的鉴定1)将拥有pGBKT7-PPARγLBD质粒的1号克隆(以下简称为P1)和拥有双质粒pGBKT7-PPARγLBD&pGADT7-CBP的2号克隆(以下简称P1C2)分别划线于T-、T-L-平板,30℃培养3-4d后,取新鲜克隆(1-3周),分别接种于2ml T-、T-L-液体培养基中,30℃、250rpm,过夜培养16-18h;2)分别用T-、T-L-液体培养基稀释P1、P1C2过夜菌(至OD600约0.3),按1∶100加入罗格列酮(自制合成,溶于二甲亚砜(DMSO),10mM)或DMSO,实验分组情况如下表

*每组均设三份平行。
30℃培养6h后,测定α-半乳糖苷酶活性(按照Yeast Protocol Handbook(CLONTECH)进行)混匀酵母培养液,取200μl测定OD 600(HITACHI,分光光度计U-2010);另取200μl菌液于1.5ml eppendorf管中,10,000rpm*5min离心;
取16μl上清于96孔板中(T-、T-L-液体培养基分别用作空白对照),每孔加入48μl分析缓冲液(100mM PNP-a-Gal水溶液与0.5M醋酸钠,pH4.5按体积比1∶2混合);30℃避光孵育30min;加入136μl 10X终止缓冲液(1M Na2CO3),终止反应;测定410nm处的OD值(BIO-RAD酶标仪MODEL 550)。
α-半乳糖苷酶活性的计算公式为α-半乳糖苷酶活性[mIU/(ml×cell)]=OD410×Vf×1,000/[(ε×b)×t×Vi×OD600]其中,t=反应时间(分)Vf=测值时的终体积(200ul)Vi=加入酵母培养液上清的体积(16ul)OD600=酵母培养液中酵母在600nm处的吸光度ε×b=对-硝基苯酚在410nm处的摩尔吸光度×比色杯的底边边长(cm)=10.5(ml/μmol)(对于200μl菌液而言)3)试验结果,各试验组的α-半乳糖苷酶活性检测结果如图11所示由图11可见,加入罗格列酮能有效提高实验菌株P1C2的α-半乳糖苷酶活性,作为溶剂的DMSO对酶活则基本无影响;而相同条件下对P1菌株进行α-半乳糖苷酶活性检测,结果均为阴性。可见,罗格列酮作为一种已知的PPAR.γ激动剂,其与PPAR.γ的结合改变了PPAR.γ的构象,使得配体依赖性的PPAR.γ与CBP的结合得以增强,最终促进了GAL4依赖的MEL1基因表达其产物α-半乳糖苷酶。
2、药物孵育时间对检测灵敏性的影响由于目前尚无用α-半乳糖苷酶做为报告基因来定量检测药物效力,借以筛选新的先导化合物的相关报道,而文献报道的采用β-半乳糖苷酶做为酵母双杂交中的报告基因进行药物筛选时,有关加药时间的确定也并不统一,从4-5h至过夜孵育不等,更无人做过加药时间与灵敏度间相关性的有关工作。
接P1C2于T-L-液体培养基中,30℃、250rpm培养过夜,基于更灵敏地检测,用T-L-液体培养基对过夜菌作不同程度的稀释(按下表),以便测酶活时,各组菌均能达到对数生长中期。

按1∶100加入罗格列酮(10mM)或DMSO,30℃孵育。在孵育2h、4h、6h、8h、10h、12h、14h时(每隔2h)取样,按照试验1所述的方法,测定各试验组α-半乳糖苷酶活性。并计算α-半乳糖苷酶相对活性,其公式为
结果如图12所示,延长药物作用的时间可提高该系统检测的灵敏度,12-14小时的孵育时间可使本系统灵敏度足以满足药物筛选的需要。
由以上实验,可以看出,PPAR.γ、CBP间这种配体依赖性的相互作用在以上的实验系统中灵敏稳定地被定量检测与衡量,可用于筛选能充当PPAR.γ配体的天然化合物(激动剂或拮抗剂)。
3、罗格列酮(阳性化合物)EC50的测定接P1C2于T-L-液体培养基中,30℃、250rpm培养过夜,用T-L-液体培养基稀释过夜菌至OD600约0.08后,以1∶100分别加入不同浓度的罗格列酮(10-9M、10-8M、10-7M、10-6M、10-5M、10-410-5M)或DMSO,30℃培养12h后,按照试验1所述的方法,测定各试验组α-半乳糖苷酶活性,并计算α-半乳糖苷酶相对活性。结果如图13所示,罗格列酮对CBP与PPARγ间相互作用的促进作用是浓度依赖性的,并据此可得到其在该系统中EC50为1.3E-6。由此,罗格列酮在本系统中可作为阳性化合物来衡量待筛药物的活性。
4、药物筛选接P1C2于T-L-液体培养基中,30℃、250rpm培养过夜,T-L-液体培养基稀释后,按1∶100分别加入罗格列酮(10mM)、DMSO以及待筛药物(10mM),30℃孵育12h后,进行α-半乳糖苷酶活性测定,并计算其激动率 本试验中待筛药物包括H001-H007为计算机药物辅助设计所得,并经BIOCORE筛选;Z001-Z013为本实验室所提取的中药(初提物或纯化合物);结果如图14所示H007、Z007表现出了强的激动效应,H001、H003、Z001、Z002也表现出了较好的激动效应;而H002、Z004则表现出了强的抑制效应。活性强的化合物可进一步测试其细胞或动物水平上的药效,活性不强的则可通过计算机辅助设计进行结构修饰,以提高其活性。

1.一种PPAR拮抗剂和激动剂的筛选方法,其特征在于利用酵母双杂交系统,采用酵母AH109基因组自带的MEL1基因作为双杂交的报告基因,通过测定α-半乳糖苷酶的活性来检测小分子化合物对PPAR与其共激活因子相互作用程度的影响,从而筛选PPAR拮抗剂和激动剂,包括如下步骤(1)CBP和酵母GAL4AD融合蛋白(pGADT7-CBP)的质粒构建;(2)PPARγLBD和酵母GAL4BD融合蛋白(pGBKT7-PPARγLBD)的质粒构建;(3)将质粒pGBKT7-PPARγLBD与质粒pGADT7-CBP转入酵母细胞AH109,得到酵母PPARγLBD-CBP_AH109(保藏号为CGMCC No.1075,菌种于2003年12月15日保藏在北京中关村中国微生物菌种保藏管理委员会普通微生物中心);(4)通过测定α-半乳糖苷酶的活性筛选化合物。
2.根据权利要求1所述的PPAR拮抗剂和激动剂的筛选方法,其特征在于将质粒pGBKT7-PPARγLBD与质粒pGADT7-CBP依次转入酵母细胞AH109的方法为采用LiAc转化法,先将pGBKT7-PPARγLBD质粒转入酵母细胞中,用色氨酸缺陷的培养基筛选阳性克隆,然后将pGADT7-CBP质粒转入含有pGBKT7-PPARγLBD质粒的酵母中,用色氨酸,亮氨酸双缺陷的培养基筛选阳性克隆,得到同时含有pGADT7-CBP质粒和pGBKT7-PPARγLBD质粒的酵母细胞。
3.根据权利要求1所述的PPAR拮抗剂和激动剂的筛选方法,其特征在于通过测定α-半乳糖苷酶的活性筛选化合物的方法为在得到的含pGADT7-CBP和pGBKT7-PPARγLBD双质粒的阳性酵母细胞中分别加入阳性对照物、阴性对照物及待筛选的化合物,孵育后,测定α-半乳糖苷酶的活性,并计算其激动率,根据各个待筛选药物的激动率的比较,筛选出PPAR拮抗剂和激动剂。
4.根据权利要求3所述的PPAR拮抗剂和激动剂的筛选方法,其特征在于通过测定α-半乳糖苷酶的活性筛选化合物时,药物孵育时间为12-14小时。
5.根据利用权利要求1所述的PPAR拮抗剂和激动剂的筛选方法,其特征在于该方法筛选PPAR拮抗剂和激动剂所利用的酵母筛选系统包括(1)小鼠共激活因子CBP和酵母转录因子GAL4-AD融合蛋白表达质粒pGADT7-CBP(菌种保藏号为CGMCC No.1076,菌种于2003年12月15日保藏在北京中关村中国微生物菌种保藏管理委员会普通微生物中心);(2)含人PPARγLBD和酵母转录因子GAL4-BD融合蛋白表达质粒pGBKT7-PPARγLBD(菌种保藏号为CGMCC No.1077,菌种于2003年12月5日保藏在北京中关村中国微生物菌种保藏管理委员会普通微生物中心);(3)宿主酵母AH109。

本发明涉及利用酵母双杂交系统,通过测定α-半乳糖苷酶的活性大小筛选PPAR拮抗剂和激动剂的方法。该方法包括(1)CBP和酵母GAL4AD融合蛋白(pGADT7-CBP)的质粒构建;(2)PPARγLBD和酵母GAL4BD融合蛋白(pGBKT7-PPARγLBD)的质粒构建;(3)将质粒pGBKT7-PPARγLBD与质粒GADT7-CBP依次转入酵母细胞AH109;(4)通过测定α-半乳糖苷酶的活性筛选化合物。
