技术领域
:本发明涉及一种制备抗原的方法,尤其涉及一种利用重组杆状病毒在昆虫体内表达口蹄疫抗原的方法,属于基因工程领域。
背景技术
:口蹄疫是由口蹄疫病毒(FoofflwJ-moW/zWn,FMDV)弓I起偶蹄动物的一种急性、高度接触性、发热性传染病,以传播迅速、感染率高而著称。该病一旦爆发将对发病国造成巨大的经济损失,世界各国对该病的研究极为重视,国际兽医局将其列为A类传染病之首。口蹄疫属于小RNA病毒科口蹄疫病毒属的成员,有A、O、C、Asial、SAT1、SAT2禾BSAT3型7个血清型。目前我国口蹄疫的防制仍以预防为主。尽管传统疫苗在口蹄疫预防控制中仍占主导地位,但生产成本昂贵,免疫期短,疫苗制备过程中病毒逃逸等危险因素很难避免。因此,传统疫苗亟需加以改进,研制安全、高效的口蹄疫分子疫苗仍显得非常必要。随着生物技术、基因工程等高新技术的发展,人们意识到通过基因工程的手段,利用生物反应器来高效表达口蹄疫基因,可望达到大幅度提高口蹄疫抗原产量、降低生产成本的目的(ConneelyO.M.,BiotechnologyintheFeedIndustry,T.P.Lyons(Ed),AlltechTechnicalPublications.Nicholasville,KY.57-66,1992)。杆状病毒表达系统这一生物反应器是八十年代建立起来的。自从1983年首次利用杆状病毒表达系统高效表达了人的a-干扰素以来(Smith,Mol.CellBiol.,3:2156-2165,1983),已有数十个外源基因得到了高效表达,仅我国就有a-干扰素(杨冠珍等,生物化学与生物物理学报,22:355-361,1990)、慈菇蛋白酶抑制剂(季平等,蚕业科学,21:223-227,1995)、马立克氏病毒糖蛋白B(肖庆利等,蚕业科学,23:104-108,1997)等多种。利用此系统来生产口蹄疫抗原的优点在于1.这一表达系统的表达效率极高,产量可达10毫克级/虫的水平。因而可大大降低口蹄疫抗原的生产成本并使通过基因工程方法大规模生产口蹄疫抗原成为可能;2.这一表达系统为真核表达系统,其表达的外源蛋白质可进行翻译后修饰,使其在生化性质和生物活性等与天然产品相似,这为所表达的口蹄疫抗原具有正常的蛋白结构和生物学免疫活性提供了保证。目前,杆状病毒表达系统,尤其是其中的家蚕(Bm)-家蚕杆状病毒(BmNPV)表达系统是世界上最具有商业开发价值的真核生物个体表达系统之一。
发明内容本发明所要解决的技术问题是克服现有技术的不足,提供一种利用杆状病毒表达系统在昆虫体内安全、高效的表达口蹄疫抗原的方法。本发明所要解决的技术问题是通过以下技术方案来实现的一种制备口蹄疫抗原的方法,包括将口蹄疫不同的基因组合分别克隆到杆状病毒运载载体中,获得转移载体;用所获得的转移载体转染杆状病毒进行DNA重组,获得重组杆状病毒;用重组杆状病毒感染昆虫宿主;培养被感染的昆虫宿主使其进行口蹄疫抗原表达;收获并纯化所表达的抗原。其中,所述的口蹄疫不同的基因组合优选自SEQIDNO:l、SEQIDNO:3或SEQIDNO:5所示的碱基序列;所述的杆状病毒运载载体优选自AcRP23-/acZ,AcRP6-SC,AcUWl-/acZ,BacPAK6,BactoPac,Bacmid,BlucBacII(pETL),p2Bac,p2Blue,p89B310,pAc360、pAc373,pAcAB3、pAcAB4,PAcAS3,pAcC129,pAcC4,DZI,pAcGP67,pAcIEl,pAcJPl,pAcMLF2、pAcMLF7、pAcMLF8,pAcMPl、pAcMP2,pAcRP23、pAcRP25,pAcRW4,pAcsMAG,pAcUWl、pAcUW21、pAcUW2A、pAcUW2B、pAcUW3、pAcUW31、pAcUW41、pAcUW42、pAcUW43、pAcUW51,pAcVC2、pAcVC3,pAcYMl,pAcJcC5,pBacl、pBac2,pBlueBacIII,pBlueBacHis,pEV55、mXIV,pIEINeo,pJVETL,pJVNhel,pJVP10,pJVrsMAG,pMBac,pP10,pPAKl,pPBac,pSHONEX1.1,pSYNXIVVI+,pSYNVI+wp,pSYNXIVVI-,pVL1391、pVL1392、pVL1393、pVL941、pVL945、pVL985,pVTBac,pBM030,pUAC-5或其它类似的杆状病毒同源重组或转座载体,更优选为pVL1393。所构建的转移载体优选为pVL1393(P1-2A3C)、pVL1393(ORF)或pVL1393(VPl)。所述的杆状病毒选自BmNPV、AcMNPV、ApNPV、BsSNPV、CfMNPV、EoSNPV、HaNPV、HzNPV、LdMNPV、MbMNPV、OpMNPV、S1MNPV、SeMNPV、或TeNPV,优选为家蚕杆状病毒Bm-NPV-ZJ8;所述的重组杆状病毒优选为以下任意一种(1)家蚕核型多角体病毒rBmNPV(ORF),其微生物保藏号是CGMCCNO.1980;保藏时间是2007年3月20H;保藏单位是中国微生物菌种保藏管理委员会普通微生物中心;保藏地址是北京市海淀区中关村北一条13号,中国科学院微生物研究所;(2)家蚕核型多角体病毒rBmNPV(P1-2A3C),其微生物保藏号是CGMCCN0.1979;保藏单位是中国微生物菌种保藏管理委员会普通微生物中心;保藏地址是北京市海淀区中关村北一条13号,中国科学院微生物研究所;(3)家蚕核型多角体病毒rBmNPV(VPl),其微生物保藏号是CGMCCN0.1975;保藏单位是中国微生物菌种保藏管理委员会普通微生物中心;保藏地址是北京市海淀区中关村北一条13号,中国科学院微生物研究所。所述的昆虫宿主选自包括家蚕(Bombyxmori)、野蚕(Bombyxmandarina)、蓖麻蚕(Philosamiacynthiaricim)、樟蚕(Dictyoplocajapanica)、樗蚕(Philosamiacynthiapryeri)、柞蚕(Antheraeapernyi)、日本柞蚕(Antheraeayamamai)、野天蚕(Antheraeapolyphymus)、苜蓿尺蠖(Atographacaliforica)、茶尺蠖(Ectropisobliqua)、甘兰夜蛾(Mamestrabrassicae)、斜纹夜蛾(Spodopteralittoralis)、秋粘虫(Spodopterafrugiperda)、粉纹夜蛾(Trichoplusiani)、行军虫(Thaumetopoeawilkinsoni)、棉铃虫(Heliothisarmigera)、美国棉铃虫(Heliothiszea)、烟青虫(Heliothisassulta)、'烟草夜虫我(Heliothisvirescens)、东方茅占虫(Pseudaletiaseparata)、舞毒蛾(Lymantriadispar)等;更优选为家蚕(Bombyxmori)。所述的感染是指重组杆状病毒通过口食或透过表皮来感染1-5龄的昆虫幼虫或蛹体(更优选为将重组家蚕杆状病毒感染家蚕细胞或穿刺接种l-5龄的家蚕幼虫或蛹,在感染3-6天后收集含口蹄疫抗原的家蚕幼虫或蛹的体液或组织匀浆);其中,所述的蛹体为l-2天的早期嫩蛹。本发明采用基因重组技术,将来源于口蹄疫病毒的不同基因组合,包括P1-2A3C、ORF、VP1构建到各种杆状病毒运载载体(如AcRP23-lacZ,AcRP6-SC,AcUWl-lacZ,BacPAK6,BactoPac,Bacmid,BlueBacII(pETL),p2Bac,p2Blue,p89B310,pAc360、373,pAcAB3、4,pAcAS3,pAcC129、C4、DZ1,pAcGP67,pAcIEl,pAcJPl,pAcMLF2、7、8,pAcMPl、2,pAcRP23、25,pAc證4,pAcsMAG,pAcUWl、21、2A、2B、3、31、41、42、43、51,pAcVC2、3,pAcYMl,pAcJcC5,pBacl、2,pBlueBacIII,pBlueBacHis,pEV55、mXIV,pIEINeo,pJVETL,pJVNhel,pJVP10,pJVrsMAG,pMBac,pP10,pPAKl,pPBac,pSHONEX1.1,pSYNXIVVI+,pSYNVI+wp,pSYNXIVVI-,pVL1391、1392、1393,pVL941、945、985,pVTBac,pBM030,pUAC-5)上,使口蹄疫病毒不同的基因组合,包括ORF、P1-2A3C、VP1基因在多角体启动子、p10启动子或别的病毒和真核生物的强启动子控制之下,通过体内或体外(invivo/invitro)重组,将口蹄疫病毒不同的基因组合,包括P1-2A3C、ORF、VP1整合到杆状病毒的基因组上,得到重组病毒。重组病毒可通过经口食下或采用各种手段透过表皮感染l-5龄(最优时间为四或五龄)的昆虫幼虫或蛹体(最优时间为l-2天的早期嫩蛹),表达生产口蹄疫病毒抗原。本发明中最优选的一个技术方案是将SEQIDNO:l(P1-2A3C)、SEQIDNO:3(ORF)或SEQIDNO:5(VP1)所示的碱基序列插入到运载载体pVL1393上,再通过体内重组将全长P1-2A3C、ORF、VP1基因转移到家蚕杆状病毒亲本株Bm-NPV-ZJ8的基因组上,替代基因组上的Polyhedrin基因,通过空斑筛选技术和PCR检测技术,获得携带口蹄疫不同基因组合的重组家蚕杆状病毒rBmNPV(ORF)、rBmNPV(P1-2A3C)、rBmNPV(VP1)。将其感染家蚕细胞系或穿刺接种1-5龄的家蚕幼虫或蛹,大量繁殖rBmNPV(ORF)、rBmNPV(P1-2A3C)、rBmNPV(VP1)。当rBmNPV(ORF)、rBmNPV(P1-2A3C)、rBmNPV(VP1)在蚕体内复制时,ORF、P1-2A3C、VP1在多角体蛋白基因(polh)启动子控制下表达,产生口蹄疫抗原。在感染3-6天(最佳为5天)后收集含口蹄疫抗原的家蚕幼虫或蛹的体液(或整体匀浆),每毫升蚕血淋巴可产生IO毫克以上的口蹄疫抗原,杀灭感染性病原后,经过蛋白纯化后便得到安全、高效的口蹄疫抗原,此种抗原可用于制备疫苗。本发明方法采用杆状病毒表达系统在家蚕生物反应器中安全、高效的生产口蹄疫抗原,其生产成本显著低于传统的制备口蹄疫抗原方法(例如通过细胞繁殖病毒制备口蹄疫抗原),无需投资建厂,无三废,电力和水资源等能源消耗极少。由于家蚕已经我国卫生部批准为食药兼用昆虫,所以将本发明方法所制备的抗原纯化后,安全性极高,可直接制作疫苗免疫动物。总体而言,本发明方法可以大幅度降低口蹄疫抗原的生产成本,具有安全、高效、能耗少、成本低等诸多优点。图l、口蹄疫病毒P1-2A3C在家蚕幼虫中的表达。图2、口蹄疫病毒P1-2A3C在家蚕蛹中的表达。图3、口蹄疫病毒全长ORF在家蚕幼虫中的表达。图4、口蹄疫病毒全长ORF在家蚕蛹中的表达。图5、口蹄疫病毒VP1在家蚕幼虫中的表达。图6、口蹄疫病毒VP1在家蚕蛹中的表达。具体实施方式为了进一步阐述本发明,下面给出一系列实施例。这些实施例完全是例证性的,它们仅用来对本发明进行具体描述,不应当理解为对本发明的限制。试验材料1.大肠杆菌株E.coliTG1和DHsa购自Promega公司;运载载体pVL1393(购自Invitrogen公司)、家蚕细胞BmN、家蚕核型多角体病毒亲本株Bm-NPV-ZJ8由中国农业科学院生物技术研究所保存;口蹄疫病毒由中国农业科学院兰州兽医研究所口蹄疫国家参考实验室保存;抗原检测试剂盒由中国农业科学院兰州兽医研究所制备,家蚕品种JY1由中国农业科学院生物技术研究所保存。2.酶与试剂限制性内切酶、连接酶为Promega公司产品。3.生化试剂IPTG、X-Gal为Promega公司产品。Lipofectin、低融点琼脂糖LMP、PCR试剂盒、T4DNA连接酶、RNA酶、ProteinaseK、胎牛血清及其他试剂购于Invitrogen公司,细胞培养基TC-100购于Sigma公司。4.培养基大肠杆菌培养基为LB(l。/。蛋白胨、0.5%酵母提取物、l%NaCl,pH7.0);家蚕细胞培养基为TC-IOO。5.口蹄疫病毒基因不同组合表达产物的动物实验在中国农业科学院兰州兽医研究所动物隔离实验室进行。实施例1口蹄疫抗原的制备、纯化及动物免疫实验及病毒攻击保护实验1、P1-2A、3C基因的克隆和序列分析1.1目的基因的获得设计引物,通过RT-PCR的方法扩增出口蹄疫病毒抗原蛋白P1-2A基因与非结构蛋白3C基因。所设计的抗原蛋白P1-2A基因和非结构蛋白3C基因的扩增引物为,P1-2A上游5,-ATAGGATCCACCATGGGAGCCGGGCAATCCAGCC陽3,,Bfl/wHl酶切位点P1-2A下游5,-CGCGAATTCTGACATGTCCTCCTGCATCTGGTTG-3,。£coRI酶切位点3C上游5,-GCGGAATTCAAGAAACCTGTCGCTTTGAAAGT-3'。£coRI酶切位点3C下游5,-ATAAGATCTCTACTCGTGGTGTGGTTCGGGAT-3'Bg/n酶切位点从口蹄疫病毒细胞培养液中提取总RNA。将提取的总RNA用01igo(dT)183物在AMV反转录酶的作用下,42。C反转录制备cDNA。以获得的cDNA为模板,用特异性引物进行PCR扩增。PCR反应体系如下表lPCR反应条件<table>tableseeoriginaldocumentpage9</column></row><table>PCR反应过程94。C变性10分钟;94。Cl分钟,58。Cl分钟,72。C8分钟,共30个循环。最后延伸反应10分钟。1.2.PCR产物的纯化将PCR扩增的P1-2A、3C基因产物进行1%琼脂糖凝胶电泳,发现扩增出约2.4kb、0.7kb的片段。在紫外灯下用灭菌手术刀切取含相应DNA片段的凝胶,然后用Geneclean试剂盒进行纯化。方法如下切取凝胶片段并称重,将其放入灭菌的1.5ml小离心管中,加入3倍(v/w)体积的6MNal,37。C将凝胶溶解后,加入10pl玻璃奶(Glassmilk),混匀后室温下放置5分钟,使DNA充分吸附在玻璃奶
